Abstract The validation of many anuran species is based on a strictly descriptive, morphological analysis of a small number of specimens with a limited geographic distribution. The Scinax Wagler, 1830 genus is a controversial group with many doubtful taxa and taxonomic uncertainties, due a high number of cryptic species. One example is the pair of species Scinax constrictus and Scinax nebulosus, which share a similar morphology. Scinax constrictus is restricted to the Brazilian Cerrado savanna, while S. nebulosus is widely distributed throughout northern South America. Despite the validation of many anuran species, discriminations based only on morphological traits is quite difficult due to the high conservative morphology of some groups. In this context, the present study uses mitochondrial and nuclear genes to provide a more consistent diagnosis and test the validity of S. constrictus as a distinct species from S. nebulosus, as well as evaluate the position of these taxa within the Scinax genus. The topologies obtained herein uphold the monophyletic status of Scinax based on all molecular markers assessed in this study, in all analytical approaches, with high levels of statistical support.

Primula intricata Gren. et Godr. has an unclear taxonomic status: it was originally described as a distinct species but subsequently was considered a subspecies (Primula elatior subsp. intricata) or even a variety (P. elatior var. intricata ) of Primula elatior (L.) Hill. No prior genetic studies were performed on this group of Primulaceae, therefore we considered useful to investigate taxonomies boundaries within the P. elatior-intricata group. We explored genetic differences between Primula intricata and Primula elatior group by applying three different types of molecular markers: nuclear ribosomal DNA (ITS1), chloroplast DNA (spacer trnH-psbA and intron trnL) and Amplified Fragment Length Polymorphisms (AFLP). We found a solid differentiation between P. intricata and P. elatior group, differentiation that was confirmed by all the employed molecular markers. This finding enabled us to propose a valid species rank for Primula intricata, as a separate taxon from the P. elatior group.

Epeorus (Caucasiron) turcicus sp. nov. is described based on larvae from NE Turkey. The new species can be distinguished from other Caucasiron by a unique combination of several diagnostic characters: the presence of a rounded hypodermal medial femur spot, colouration of abdominal terga and sterna, narrow gill plate VII, fine hair-like setae on the surface of abdominal terga, and absence of postero-lateral projections on tergum X. In addition to morphological analysis, two single-locus analytical approaches are employed for delimiting the new species using COI sequences (Automatic Barcode Gap Discovery, ABGD; and General Mixed Yule Coalescent Model, GMYC). Both approaches unambiguously recognized E. (C.) turcicus sp. nov. as a distinct species. Our molecular dataset contains all Caucasiron species occurring in the Caucasus and the delimitation of individual species mostly follows the morphologically defined species. This study confirms the suitability of the GMYC approach for species delimitation within Caucasiron.

Passiflora ovalis and P. contracta are species restricted to the Atlantic Rainforest (Mata Atlântica) of Brazil and were separated into two distinct taxonomic units based on different morphological traits, as inflorescence structure and indument, being the populations located between 7–19°S referred to P. contracta, while those found around 23°S belong to P. ovalis. Recently the species were grouped together under the specific name P. ovalis. The objective of this work was to understand the taxonomy of these species based in a Bayesian phylogenetic analysis using a combination of molecular markers from plastidial and nuclear genomes. The results confirmed the existence of two distinct species supported by full posterior probability values. We consequently propose accepting the two-species, which also have been validated both by morphologic differences and distinct geographic distribution.

An investigation of the taxonomic status of Pareas hamptoni (Hampton's Slug snake) based on morphological and molecular data revealed a new distinct species from the Golden Triangle region (comprising parts of southern China, and adjacent Laos and Thailand). The new species is shown to be a sister species to P. hamptoni but can be separated from the latter by having 3–5 dorsal scale rows at midbody slightly keeled (vs 5–9 scales strongly keeled); a lower number of ventrals, 170–188 (vs 185–195); and a lower number of subcaudals, 67–91 (vs 91–99). The new species is currently known from northwestern Thailand, northern Laos, and the southern part of Yunnan Province in China at elevations of 1,160–2,280 m a.s.l. We suggest that the new species to be considered of Least Concern (LC) in the IUCN‘s Red List categories. Problems of taxonomy and actual distribution of the P. hamptoni complex are briefly discussed; our results show P. hamptoni is now reliably known only from Myanmar and Vietnam, but its occurrence in Yunnan Province of China is likely.

AbstractThe Greek lentil landrace ‘Eglouvis’ is cultivated continuously at the Lefkada island for more than 400 years. It has great taste, high nutritional value and high market price. In the present study, we used morphological and molecular markers to estimate genetic diversity within the landrace. Morphological analysis was based on characteristics of the seed. Molecular analysis was performed using simple sequence repeat (SSR) molecular markers in a high-resolution melting (HRM) approach. ‘Samos’ and ‘Demetra’, two of the most widely cultivated commercial lentil varieties in Greece, were used for comparisons. Morphological analysis was performed with 584 seeds randomly selected from a lot. Analysis of seed dimensions and colour distributed the samples in different categories and highlighted the phenotypic variability in ‘Eglouvis’ lentil seeds. Genetic variability was estimated from 91 individual DNA samples with 11 SSR markers using HRM analysis. Genotyping was based upon the shape of the melting curves and the difference plots; all polymerase chain reaction products were also run on agarose gels. Genetic distances of individuals and principal coordinates analysis suggested that ‘Eglouvis’ landrace has a unique genetic background that significantly differs from ‘Samos’ and ‘Demetra’ and no overlapping could be detected. Genetic variability within the ‘Eglouvis’ landrace can be considered in targeted breeding programs as a significant phytogenetic resource of lentils in Greece.

Specimens of monstrilloid copepods collected and described in the early 20 th century by G.O. Sars from the coasts of Norway and deposited in the Sars Collection (University of Oslo) were re-examined. Monstrilla leucopis Sars, 1921 was described based on female and male specimens, but the species was later synonymized with M. conjunctiva Giesbrecht, 1902 by several authors. Females of this species were analyzed and compared with closely related congeners, particularly with M. conjunctiva. This analysis includes the description of previously unknown morphological details following upgraded descriptive standards in this group. Evidence was found to support the notion that the female type specimens from Kvalø, Norway represent a distinct species; thus, M. leucopis is redescribed and reinstated as a valid taxon. Previous tropical records of female M. conjunctiva are questionable, but differences with M. leucopis can be found in body and antennule proportions, the structure of the genital spines and fifth legs, and most probably, their geographical ranges. Furthermore, M. leucopis has a modified thick-walled seta on the endopods of legs 2–4, so far a unique character among monstrilloids. The single male specimen labeled as M. leucopis in the Sars Collection was also examined and it is not the male of this species as depicted by G.O. Sars (1921). It is in fact a male of M. longiremis Giesbrecht, 1893, a species for which a short supplementary description and taxonomic comments are also provided herein. The male of M. leucopis also shares some important characters with that of M. conjunctiva, but also with another male specimen that was questionably assigned to the latter species; this male probably represents an undescribed species.
