The corn leafhopper Dalbulus maidis is the main vector of the pathogens that cause corn stunt, a major disease of maize in the Americas. As host resistance is an efficient tool to control diseases, the findings of a previous report showed that some corn hybrids are resistant to D . maidis . In this work, we assessed the probing behavior of D . maidis on susceptible and resistant corn hybrids using EPG (Electrical Penetration Graph) technology. Fifteen-day-old females were monitored for 20 hours, with access to hybrids DK390, DK670, DK79-10, and DK72-10. Hybrids DK390 and DK72-10 showed resistance to D . maidis in phloem, since insects feeding on these hybrids presented more salivation events in phloem without subsequent ingestion, which are seen as failed attempts to ingest. A reduction of the total duration of phloem ingestion was observed, and accordingly of the time spent by insects with access to these hybrids on xylem ingestion. The hybrid DK390 also had mesophyll resistance, seen as less probing time and a higher number of probes of short duration. These findings support and are consistent with previous research, providing useful information to characterize maize hybrids resistant to D . maidis , and so to corn stunt.

AbstractTrehalose serves multifarious roles in growth and development of insects. In this study, we demonstrated that the high trehalose diet increased the glucose content, and high glucose diet increased the glucose content but decreased the trehalose content of Acyrthosiphon pisum. RNA interference (RNAi) of trehalose-6-phosphate synthase gene (ApTPS) decreased while RNAi of trehalase gene (ApTRE) increased the trehalose and glucose contents. In the electrical penetration graph experiment, RNAi of ApTPS increased the percentage of E2 waveform and decreased the percentage of F and G waveforms. The high trehalose and glucose diets increased the percentage of E2 waveform of A. pisum red biotype. The correlation between feeding behavior and sugar contents indicated that the percentage of E1 and E2 waveforms were increased but np, C, F and G waveforms were decreased in low trehalose and glucose contents. The percentage of np, E1 and E2 waveforms were reduced but C, F and G waveforms were elevated in high trehalose and glucose contents. The results suggest that the A. pisum with high trehalose and glucose contents spent less feeding time during non-probing phase and phloem feeding phase, but had an increased feeding time during probing phase, stylet work phase and xylem feeding phase.

Carystoterpa fingens (Walker) is an endemic xylem feeder It was chosen as a model insect for the glassy winged sharp shooter (Homalodisca coagulata (Say)) The electrical penetration graph (EPG) technique was used to measure realtime stylet penetration and feeding behaviour of 48 adult C fingens on four plant types hebe (Hebe pubescens) rengarenga lily (Arthropodium cirratum) wild iris (Dietes bicolor) and cabbage (Brassica oleracea) over 12 days EPG waveforms were recorded for 12 hours and the duration of the main penetration events (pathway ingestion and nonprobing phases) of the insects were analysed to compare host preferences There were no significant differences in the duration of ingestion indicating that all the plants were potential hosts for C fingens Insects on hebe showed the shortest nonprobing time suggesting that hebe was the most preferred host as expected from observations in the wild

AbstractThe effect of structurally related sesquiterpenoids (E,E)-farnesol and cis-nerolidol on the host-plant selection behaviour of the peach potato aphid Myzus persicae (Sulz.) was evaluated using electrical penetration graph (EPG) technique. No repellent effects of (E,E)-farnesol and (Z)-nerolidol to M. persicae were found but aphid probing activities on (E,E)-farnesol- and cis-nerolidol-treated plants were restrained. During non-phloem phases of probing, neither (E,E)-farnesol nor (Z)-nerolidol affected the cell puncture activity. On (E,E)-farnesol-treated plants, the total duration of phloem phase, the mean duration of individual sustained ingestion periods were significantly lower, and the proportion of phloem salivation was higher than on control plants. On (Z)-nerolidol-treated plants, the occurrence of the first phloem phase was delayed, and the frequency of the phloem phase was lower than on control plants. The freely moving aphids were reluctant to remain on (E,E)-farnesol- and (Z)-nerolidol-treated leaves for at least 24 h after exposure. (E,E)-Farnesol and (Z)-nerolidol show complementary deterrent properties, (E,E)-farnesol showing ingestive and post-ingestive activities and nerolidol showing pre-ingestive, ingestive, and post-ingestive deterrent activities.

Abstract Aphids, including the bird cherry-oat aphid (Rhopalosiphum padi), are significant agricultural pests. The wild relative of barley, Hordeum spontaneum 5 (Hsp5), has been described to be partially resistant to R. padi, with this resistance proposed to involve higher thionin and lipoxygenase gene expression. However, the specificity of this resistance to aphids and its underlying mechanistic processes are unknown. In this study, we assessed the specificity of Hsp5 resistance to aphids and analysed differences in aphid probing and feeding behaviour on Hsp5 and a susceptible barley cultivar (Concerto). We found that partial resistance in Hsp5 to R. padi extends to two other aphid pests of grasses. Using the electrical penetration graph technique, we show that partial resistance is mediated by phloem- and mesophyll-based resistance factors that limit aphid phloem ingestion. To gain insight into plant traits responsible for partial resistance, we compared non-glandular trichome density, defence gene expression, and phloem composition of Hsp5 with those of the susceptible barley cultivar Concerto. We show that Hsp5 partial resistance involves elevated basal expression of thionin and phytohormone signalling genes, and a reduction in phloem quality. This study highlights plant traits that may contribute to broad-spectrum partial resistance to aphids in barley.

Abstract In the current context of global climate change, atmospheric carbon dioxide (CO2) concentrations are continuously rising with potential influence on plant–herbivore interactions. The effect of elevated CO2 (eCO2) on feeding behavior of corn leaf aphid, Rhopalosiphum maidis (Fitch) on barley seedlings Hordeum vulgare L. was tracked using electrical penetration graph (EPG). The nutrient content of host plant and the developmental indexes of aphids under eCO2 and ambient CO2 (aCO2) conditions were also investigated. Barley seedlings under eCO2 concentration had lower contents of crude protein and amino acids. EPG analysis showed the plants cultivated under eCO2 influenced the aphid feeding behavior, by prolonging the total pre-probation time of the aphids (wandering and locating the feeding site) and the ingestion of passive phloem sap. Moreover, fresh body weight, fecundity and intrinsic population growth rate of R. maidis was significantly decreased in eCO2 in contrast to aCO2 condition. Our findings suggested that changes in plant nutrition caused by eCO2, mediated via the herbivore host could affect insect feeding behavior and population dynamics.
