Identification of 3β-hydroxy-5,7-pregnadien-20-one and 3β-hydroxy-5,7-androstadien-17-one as endogenous steroids in the fetal horse gonad

The 5,7-dienes, 3β-hydroxy-5,7-pregnadien-20-one and 3β-hydroxy-5,7-androstadien-17-one were extracted from fetal horse gonads and purified by solvent partition, thin-layer chromatography and high performance liquid chromatography. The isolated steroids were identified by comparison with the synthetic steroids using ultraviolet and mass spectroscopy and by gas chromatography–mass spectroscopy. The identification of these compounds as endogenous steroids, together with the data on their biosynthesis reported previously, support the proposal that in the fetal horse gonad there is a 5,7-diene pathway biosynthesizing 3β-hydroxy-5,7-androstadien-17-one, which is the proposed precursor for equilin in the placenta.

Vitamin E, the fat soluble vitamin is present naturally in some foods and added in food supplements, nutraceuticals etc due to its vital biological function as an antioxidant. Various methods are available for the analysis of vitamin E. Especially High Performance Liquid Chromatography (HPLC) and Gas Chromatography (GC) are exclusively used for the quantitative evaluation of vitamin E, which has also identified the four different isomeric forms of this vitamin. The rate of losses of this vitamin during food processing and analysis, in addition to their transient dynamics, presents complexities in developing a highly sensitive procedure for their separations. Though effective, HPLC instrument is expensive and comparatively cumbersome. In this prospective, the study was to evaluate the usefulness of High Performance Thin Layer Chromatography (HPTLC) in the analysis of vitamin E. There are methods available using Thin Layer Chromatography for its analysis, but they are not sensitive enough to identify the isomeric forms of vitamin E. In this HPTLC method, the different isomeric forms of vitamin E - α, β, γ and δ were identified. This technique shall be considered as an alternative to the other methods such as HPLC and GC.

ABSTRACT A suitable antioxidant for a poly(ethylene-co-propylene-co-5-ethylidene-2-norbornene) (EPDM) seal in tap water applications was determined with respect to volatilization and decomposition of the antioxidants. Seals were prepared by mixing EPDM with 1 phr antioxidant and other ingredients followed by vulcanizing the mixture at 433 K for 20 min. The resulting EPDM seals were immersed into chlorinated water to investigate accelerated degradation. The change in antioxidant content was measured by gas chromatography/mass spectroscopy (GC/MS) and high-performance liquid chromatography (HPLC). The weight loss of amine antioxidants during vulcanization was quite low due to their low volatility and decomposition. Antioxidant weight loss during accelerated degradation depended on both the antioxidant's ability to trap radicals and solubility in chlorinated water.

ABSTRACT Identification of tetrodotoxin (TTX) and its derivatives produced from a Vibrio strain in the intestine of the puffer fishFugu vermicularis radiatus was performed by thin-layer chromatography, electrophoresis, high-performance liquid chromatography, and gas chromatography-mass spectrometry, together with a mouse bioassay for toxicity. It was demonstrated that the isolated bacterium produced TTX, 4-epi-TTX, and anhTTX during cultivation, suggesting that Vibrio strains are responsible for the toxification of the puffer fish.
