Absence of circadian patterns of secretion of melatonin or cortisol in Weddell seals under continuous natural daylight

ABSTRACT Blood samples were obtained from adult female Weddell seals in McMurdo Sound, Antarctica, utilizing four different seals every 2 h for 24 h on two occasions in November 1987. Plasma levels of melatonin were mostly near zero, whereas the values for cortisol were high (2101 ±54 and 1908 ± 108 (s.e.m.) nmol/l, n = 52). For both hormones there was no evidence of a diurnal pattern of secretion. Juvenile seals sampled in December 1985 also had no evidence of a diurnal secretion pattern of melatonin, but blood levels of melatonin were higher than in adults. Study was of the behaviour of 9 marked female Weddell seals, Leptonychotes weddelli, and their young in a colony of 35 to 40 such pairs at Turtle Rock, McMurdo Sound, off Ross Island, Antarctica. The young sucked first about 80 min after birth. Sucking was usually initiated by the young seal, which had to raise its head to reach either of the 2 nipples. After sucking for 10 to 20 sec the young seal dropped down to the ice and waited for 1 to 2 min before sucking again. It was not clear whether the young seal had to rest after supporting itself or whether it had cleared the milk from the gland cistern and had to wait while milk from alveoli and ducts refilled the cistern. For young more than 1 week old there was a diurnal pattern of sucking with peaks between 1200 and 2359 h and least between 0300 and 0600 h. When the young began to swim they used more energy, stored less fat and sucked less. Early swimming, resulting in lower bodyweight at weaning, could affect survival during the following winter. Published values for bodyweight increments of growing seals are given. Young Weddell seals accumulate less fat than some species but learn to catch food before they are weaned.

The preservation and conservation of living resources was one of the original stated objectives of the Antarctic Treaty, 1959. The harvesting of animals near bases for food, especially Weddell Seals Leptonychotes weddelli received particular attention at the Fourth Meeting of SCAR, 1960 (Polar Record,1961) and several regulations were suggested including: a permit required before killing, records to be kept of animals killed, and use of alternative food where possible. Annex D, Section (b)i states “… Without detailed biological research it is impossible to assess the permissible annual crop which the various [Weddell] seal colonies would stand, but it is certain that this figure is being seriously exceeded in many areas.”

The alimentary tracts of seven Weddell seals taken at McMurdo Sound, Antarctica, were examined for helminth parasites. Two species of trematodes, three of cestodes, and one of nematodes were recovered. Numerical data on incidence and intensity of infection are given for each species. Heavy infections of diphyllobothriid plerocercoids were also recorded.

Summary1. The concentration of the major components of the fibrinolytic enzyme system was compared in venous and arterial blood samples from male subjects.2. The plasminogen activator concentration was higher in venous blood and the arterio-venous difference increased as its concentration rose, but the ratio of the arterial to venous level remained constant.3. No arterio-venous difference was found for anti-urokinase activity, antiplasmin, plasminogen and fibrinogen.4. It is concluded that venous blood determinations of the components of the fibrinolytic enzyme system reflect satisfactorily arterial blood levels.

The history of animal-borne instrumentation is reviewed from the first basic depth gauge invented in the late 1800s, to the complex animal-borne imagery and archival systems of the present day. A major breakthrough occurred in 1964 when the first time-depth recorder was deployed on a Weddell Seal in McMurdo Sound, Antarctica. The next phase in the study of animals at sea was the use of microprocessors as archival recorders in the mid-1980s. These also were first attached to Weddell seals in McMurdo Sound. Microprocessor technology made possible the next major step of attaching a video camera housed in a submersible case (Crittercam) to a loggerhead turtle. Since the 1990s the field of “Biologging” has flourished, with new additions of satellite and GPS tracking, and resulted in three major international symposiums in the past four years (2003-2007).

ABSTRACT In sheep, secretion of oestradiol by the ovary is stimulated by pulses of LH but the factors controlling ovarian inhibin secretion are not well understood. We have investigated the effect of a single injection of LH on the ovarian secretion of inhibin. Six anoestrous Finn–Merino ewes which had one ovary autotransplanted to a site in the neck had jugular and timed ovarian venous blood samples collected at 10-min intervals for a total of 5 h. The secretion rates of both inhibin (1–3 ng/min) and oestradiol (0·5–8 ng/min) were similar to those observed during the breeding season indicating significant follicular development in these animals. After injection of 2·5 μg NIH-LH-S25 intravenously the concentration of LH in plasma rose from a baseline of 1·8 ±0·1 (s.e.m.) μg/l to a peak of 3·9 ±0·3 μg/l (P<0·01). This LH pulse stimulated a corresponding increase (P<0·01) in oestradiol secretion from a basal level of 0·9±0·2 ng/min to a peak of 4·6±0·6 ng/min that occurred within 30 min of injection. Although inhibin secretion was episodic in nature, increases were not related to either exogenous or endogenous LH pulses. We conclude that, in contrast to oestradiol, the secretion of inhibin by the ovary is not controlled acutely by changes in plasma levels of LH during anoestrus. The Weddell seal (Leptonychotes weddelli) is one of the most vocal pinnipeds. The repertoires of subice vocalizations of Weddell seals recorded at Palmer Peninsula and at McMurdo Sound, Antarctica, are different. Although seals at both sites give some of the same vocalizations, there are subtle spectral and temporal differences. In addition each population has unique vocalizations which are not heard at the other site. At Palmer Peninsula, there are several usage characteristics not exhibited at McMurdo Sound, such as mirror-image vocalization pairs and vocalization trios. Weddell seals in McMurdo Sound make extensive use of nine auxiliary sounds, while the Peninsula repertoire has none. Factors which appear to have been important in the development of these geographic differences appear to include strong fidelity to breeding sites, a polygynous mating system, and learning. Geographically different vocal repertoires have potential for identifying discrete breeding stocks of Antarctic seals.

ABSTRACT Two experiments were conducted to explore the effectiveness of synthetic peptide-based vaccines for active and passive autoimmunization of sheep against inhibin. In the first experiment, adult Romney ewes (n = 20) were actively immunized against a synthetically produced peptide that corresponded to the N-terminus of the α-subunit of bovine inhibin (bIα(1–29)-Tyr30). This peptide was conjugated to tuberculin purified protein derivative (PPD) to increase its antigenic properties. Control groups comprised non-immunized (n = 10) and PPD-immunized (n = 10) ewes. Primary immunization (400 μg conjugate/ewe) was followed by two booster immunizations (200 μg conjugate/ewe), given 5 and 8 weeks later. Following synchronization of oestrus using progestagen sponges, ovulation rates were assessed by laparoscopy. Weekly blood samples were taken throughout the experiment. All inhibin-immunized ewes produced antibodies which bound 125I-labelled bovine inhibin (Mr 32 000), and ovulation rate in inhibin-immunized ewes (2·15 ± 0·22; mean ± s.e.m.) was significantly (P<0·01) greater than in both non-immunized (0·90 ± 0·23) and PPD-immunized (1·20 ± 0·13) control groups. Immunization against the peptide, but not against PPD alone, resulted in a modest rise in plasma FSH, with mean levels after the second boost being significantly (P<0·025) higher (22%) than those before immunization. Moreover, when blood samples were taken (2-h intervals) from randomly selected groups of control (n = 7) and inhibin-immunized (n = 7) ewes for an 84-h period following withdrawal of progestagen sponges, the mean plasma concentration of FSH during the 48 h immediately before the preovulatory LH surge was 37% greater (P< 0·025) in immunized than in control animals. However, more frequent blood sampling (every 15 min for 12 h) during follicular and mid-luteal phases of the oestrous cycle revealed no significant differences between treatment groups in mean plasma concentrations of FSH. In addition, neither mean concentrations of LH nor the frequency and amplitude of LH episodes differed between immunized and control ewes. However, the mean response of LH to a 2 μg bolus of gonadotrophin-releasing hormone, given during the luteal phase, was significantly (P<0·05) less in immunized than in control ewes. These findings indicate that active immunization of Romney ewes against a synthetic fragment of inhibin can promote a controlled increase in ovulation rate, but this response cannot be unequivocally related to an increase in plasma levels of FSH. In the second experiment, passive immunization of seasonally anoestrous ewes (mule × Suffolk crossbred; n = 6 per group) against inhibin, using an antiserum raised in sheep against a synthetic peptide corresponding to the N-terminus of the α-subunit of human inhibin promoted a rapid (<3 h), dose-dependent rise in plasma levels of FSH which remained increased (2·5-fold; P<0·001) for up to 30 h. Plasma concentrations of LH, however, were unaffected by treatment with the antiserum. It is deduced from this observation that, even in the seasonally anoestrous ewe, the ovary secretes physiologically active levels of inhibin, which exert an inhibitory action on the synthesis and secretion of FSH. Members of the genus Staphylococcus are widespread in nature and occupy a variety of niches, however, staphylococcal colonization of animals in the Antarctic environment has not been adequately studied. Here, we describe the first isolation and characterization of two Staphylococcus intermedius group (SIG) members, Staphylococcus delphini and Staphylococcus pseudintermedius, in Antarctic wildlife. Staphylococcus delphini were found exclusively in Adélie penguins. The report of S. pseudintermedius from Weddell seals confirmed its occurrence in all families of the suborder Caniformia. Partial RNA polymerase beta-subunit (rpoB) gene sequencing, repetitive PCR fingerprinting with the (GTG)5 primer, and matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry gave consistent identification results and proved to be suitable for identifying SIG members. Comparative genomics of S. delphini isolates revealed variable genomic elements, including new prophages, a novel phage-inducible chromosomal island, and numerous putative virulence factors. Surface and extracellular protein distribution were compared between genomes and showed strain-specific profiles. The pathogenic potential of S. delphini was enhanced by a novel type of exfoliative toxin, trypsin-like serine protease cluster, and enterotoxin C. Detailed analysis of phenotypic characteristics performed on six Antarctic isolates of S. delphini and eight reference strains from different animal sources enabled us to emend the species description of S. delphini.
