This case study aims to determine the effect of a combination of prostaglandin F2α (PGF2α) with Gonadotropin (PG-600), and human chorionic gonadotropin (hCG) on the estrus and pregnancy rates in Holstein Friesian (HF) cows with persistent corpus luteum (PCL). A total of 15 Holstein Friesian (HF) dairy cows aged 3-5 years with body condition score (BCS) 5-7 (scale 1-9) diagnosed with PCL, were injected with 7.5 mg PGF2α intra-vulvar submucosa. Cows were then divided into three treatment groups. Group P0 was not given the combination treatment, while P1 group was injected with a  combination of 300 IU PG-600 and 150 IU hCG, meanwhile P2 group was was injected with a combination of 300 IU PG-600 and 300 IU hCG intra-muscularly. The PGF2α and PG-600 injections were carried out on the same day PCL was diagnosed. Meanwhile, the injection of hCG was carried out at the same time as the AI. The Pregnancy examination was performed 90 days after AI through rectal palpation. The results showed that all treated cows were estrus  observed with the fastest onset of estrus was the P2 group, that was significantly different (p <0.05) compared to the P0 and P1 groups. All treated cows were diagnosed as pregnant. It can be concluded that the administration of PGF2α or its combination with gonadotropins in HF cows with PCL could result in 100% estrus and pregnancy rates.

Treatments with progesterone (P4) releasing devices and estradiol benzoate (EB) have been extensively used in fixed-time AI (FTAI) programs in beef cattle. However, pregnancy rates in postpartum cows kept on pasture often have been lower than expected because of poor body condition score (BCS) and a high incidence of anestrous. A recent study (Cutaia L et al., 2003 Theriogenology 59, 216) suggested that the addition of eCG to a P4/EB FTAI program may improve pregnancy rates in cows with fair to poor BCS, but results were not conclusive. Therefore, this experiment was designed to further investigate the effect of eCG treatment on pregnancy rates in postpartum beef cows in those conditions. The experiment was performed at 2 locations with lactating Angus cows (n=93), 60 to 90d postpartum with a BCS 1.9 (1 to 5 scale; Location 1), and crossbred Zebu cows (n=290), 60 to 90d postpartum with a BCS 2.0 (Location 2). In each Location, cows were randomly allocated to 1 of the following treatment groups: Control, eCG Day6, or eCG Day8. All cows received a P4 intravaginal device (DIB, Syntex, Argentina) and 2mg EB i.m. (Syntex) on Day0, 500mg cloprostenol (Estroplan, Syntex) at the time of DIB removal (Day8), 1mg EB i.m. on Day9 and FTAI 52 to 56h after DIB removal. Cows in the Control group received no further treatment, whereas cows in the eCG-treated groups received 400 IU eCG (Novormon 5000, Syntex) on Day6 or Day8. Cows were examined on Day0 by rectal palpation (Location 1) or by ultrasonography (US; Location 2) and were classified as those with a CL or without a CL, with either large (&gt;8mm) or small (&lt;8mm)small follicles. Pregnancy was determined by US 45d after FTAI. Data were analyzed by logistic regression and the effects of location, treatment, ovarian status, AI technician and semen were considered in the model. There was no effect of location (P=0.3), AI technician (P=0.2) or semen (P=0.8) on pregnancy rates. However, there was an effect of treatment (P=0.02), attributed to higher pregnancy rates in the eCG Groups than in the Control Group (Table). Furthermore, cows with a CL or without a CL but with large follicles on Day 0 had higher pregnancy rates than those with small follicles (P=0.04). It was concluded that the use of eCG in a P4/EB FTAI program improved pregnancy rates in postpartum beef cows that were in fair topoor BCS.

A significant factor in improving the performance of dairy cows is their physiological ability to correct a negative energy balance (NEB). This study, using Simmental (SIM) and Holstein-Friesian (HF) cows, aimed to assess changes in NEB (non-esterified fatty acid; body condition score; and C16:0, C18:0, and C18:1) and its effect on the metabolic efficiency of the liver (β-hydroxybutyrate and urea). The effects of NEB on daily yield, production at peak lactation and its duration, and changes in selected milk components were assessed during complete lactation. Up to peak lactation, the loss of the body condition score was similar in both breeds. Subsequently, SIM cows more efficiently restored their BCS. HF cows reached peak lactation faster and with a higher milk yield, but they were less able to correct NEB. During lactation, their non-esterified fatty acid, β-hydroxybutyrate, C16:0, C18:0, C18:1, and urea levels were persistently higher, which may indicate less efficient liver function during NEB. The dynamics of NEB were linked to levels of leptin, which has anorectic effects. Its content was usually higher in HF cows and during intensive lactogenesis. An effective response to NEB may be exploited to improve the production and nutritional properties of milk. In the long term, it may extend dairy cows’ productive life and increase lifetime yield.

A comparative hormonal study of prostaglandin, gonadotropin releasing hormone and their combination was conducted on 30 randomly selected cross-bred dairy cows of SEMEX project for artificial insemination in Hillat Kuku, Khartoum North, Sudan. The study attempts to determine the response of the animals to hormonal induction and synchronization of estrous as well as fertility following fixed time artificial insemination. Three treatment groups of 10 cows each were undertaken. Groups 1, 2 and 3 involved administration of prostaglandin F2α (PGF2α), gonadotropin releasing hormone (GnRH) and a combination of PGF2α + GnRH, respectively. Descriptive statistical analysis was performed. The results revealed that all the three protocols induced and synchronized estrous were almost synchronous in all the treated groups. However, protocol 3 revealed a significantly (p<0.05) better result of 70% pregnancy rate compared to other two protocols that equally provided 50% pregnancy rates. The study resolves that protocol 3 be highly recommended to change the mindset of farmers about the spread of reproductive technology in Sudan. Protocols 1 and 2 provided acceptable pregnancy rates that can be enhanced with the improvement of management.

Two experiments were designed to evaluate the effect of the timing of fixed-time AI (FTAI) in relation to the removal of an intravaginal progesterone-releasing device (1 g of progesterone, DIB, Syntex SA, Buenos Aires, Argentina) on pregnancy rates in Bos indicus × Bos taurus cross-bred heifers. In experiment 1, 285 Bonsmara × zebu cross-bred heifers, between 18 and 24 months of age and with a body condition score (BCS) between 3.0 and 3.5 (1-5 scale) were used. On the day of initiation of treatment (Day 0), the heifers’ ovaries were palpated (92% of them had a CL) and they received a new DIB plus 2 mg of estradiol benzoate (EB; Syntex SA) and 250 μg of cloprostenol (Ciclase DL, Syntex SA). On Day 8, DIB devices were removed and all heifers received 250 μg of Ciclase plus 0.5 mg of estradiol cypionate (ECP; Cipiosyn, Syntex SA). At that time the heifers were randomly divided to receive FTAI between 48 to 49 h, 53 to 54 h, or 58 to 59 h after DIB removal. The heifers underwent FTAI with semen from 4 bulls by 2 inseminators. In experiment 2, 260 heifers from the same group as those used in experiment 1 (87% with a CL) were treated exactly as those in experiment 1, except that previously used DIB was inserted on Day 0. Pregnancy diagnosis was performed 30 days post-fixed-time AI by ultrasonography. The data were analyzed by logistic regression, taking into account the effect of time of FTAI, semen, and inseminator on pregnancy rates. In experiment 1, pregnancy rates were lower (P = 0.04) in the heifers undergoing FTAI between 48 and 49 h after DIB removal (46/95, 48.4%) than those undergoing FTAI 53 to 54 h (61/99, 61.6%) or 58 to 60 h (57/91, 62.6%) after DIB removal. However, no differences in pregnancy rates were found (P = 0.72) in experiment 2 between the 3 treatment groups, with 39/91 (42.9%) for the 48 to 49 h group, 45/89 (50.6%) for the 53 to 54 h group, and 35/89 (43.8%) for the 58 to 59 h group. There was no effect of the semen or inseminator (P > 0.2) in either experiment. We conclude that when Bos indicus × Bos taurus beef heifers are synchronized with new DIB devices and ECP, higher pregnancy rates are obtained in heifers undergoing FTAI late (between 53 to 60 h after DIB removal) than in those undergoing FTAI early (48 to 49 h after DIB removal). However, time of insemination does not apparently affect pregnancy rates when Bos indicus × Bos taurus beef heifers are synchronized with previously used DIB devices and ECP.

The objective of our research was to evaluate the effect of some factors on the embryonic mortality of Holstein-Friesian dairy cows in a herd. Monitoring was conducted in the course of three years (2006–2009) at a level of (9330–8970) kg/dairy cow per lactation. We monitored the reproduction parameters of 209 dairy cows (1st to 8th lactation). Ultrasonography was used to diagnose gravidity, and/or embryonic mortality, in each cow 6 times, at regular 4-day intervals between days 22 and 42 after insemination. We analysed the effect of 3 factors on embryonic mortality: age, body condition score and season of the year at the time of insemination. Embryonic mortality relative to the age of the female was significantly the lowest (20.69 %) in cows aged 42 to 47 months (3.5–3.9 years); high embryonic mortality (45.46 %) was recorded in the youngest females of 24 to 29 months of age (2.0–2.4 years). The results indicated that the body condition score of the cow at the time of insemination had a statistically significant (P > 0.01) effect on embryonic mortality (EM). Embryonic mortality was the lowest when the body condition of the cow was “ideal”, i.e. 5 points of the BCS (at the most 9 points). The season of the year when insemination was carried out had a positive effect on embryonic mortality.

Several studies, performed mainly in North America, have shown that the 5-day co-synch timed-AI (TAI) protocol results in similar or higher pregnancy rates than conventional 7-day co-synch protocols in beef cows and beef and dairy heifers. It is hypothesised that the 5-day co-synch protocol reduces the incidence of persistent follicles and provides for a longer proestrus, with increasing oestradiol concentrations due to continuous gonadotropin support for the dominant follicle (Bridges et al. 2008 Theriogenology 69, 843–851). Similar findings have been reported recently following the use of a shortened oestradiol-based protocol that also provides for a longer proestrus (named J-synch; de la Matta and Bo 2012 Taurus 55, 17–23). A study was designed to compare these 2 treatment protocols for synchronization of ovulation that allow for a prolonged proestrus with the conventional oestradiol-based protocol currently used for TAI in South America for heifers. Cycling Holstein heifers that were, on average, 17 months of age, weighting 350 to 380 kg, and with a body condition score of 2.5 to 3.5 (scale of 1 to 5), were randomly allocated to 1 of 3 treatment groups. Heifers in the 7-day estradiol benzoate (EB) group received a progesterone device (DIB 1 g of progesterone; Syntex SA, Buenos Aires, Argentina) and 2 mg EB (oestradiol benzoate, Syntex SA) on Day 0, 500 μg of cloprostenol (prostaglandin F2α; Sincronil, Proagro SA, Argentina) and DIB removal on Day 7, 1 mg of EB on Day 8, and TAI 30 h later (54 h after DIB removal). Heifers in the J-synch group received a DIB and 2 mg of EB on Day 0, PGF and DIB removal on Day 6, and 10 μg of buserelin acetate [gonadotropin-releasing hormone (GnRH); Receptal, Intervet, Millsboro, MD, USA] and TAI on Day 8 (72 h later). Heifers in the 5-day co-synch group received a DIB and GnRH on Day 0, DIB removal and prostaglandin F2α (twice, 12 h apart) on Day 5, and GnRH along with TAI on Day 8 (72 h later). Treatments were coordinated so that DIB were removed in all groups at the same time to decrease a possible time effect of follicle parameters and pregnancy rates. Two experiments were conducted; the first evaluated follicular dynamics and ovulation rates by twice daily ultrasonography (Honda 101 V, 5.0 MHz) in 20 heifers per group and the second determined pregnancy rates to TAI in 172 heifers. The mean (± standard error of the mean) interval from DIB removal to ovulation was shorter in the 7-day EB group (82.7 ± 3.1 h) than in the J-synch (103.8 ± 3.3 h) and 5-day co-synch (96.8 ± 3.3 h) groups (P = 0.01). However, the mean diameter of the dominant preovulatory follicle was smaller (P = 0.01) in the 7-day EB (12.8 ± 0.5 mm) and the J-synch (13.1 ± 0.6 mm) groups than in the 5-day co-synch group (14.9 ± 0.6 mm). Although pregnancy rates in the second experiment were numerically lower in the 7-day EB group (31/58; 53.4%) and 5-day co-synch group (30/55; 54.5%) than in the J-synch group (37/59; 62.7%), differences were not significant (P > 0.3). Preliminary results indicate that the 3 protocols evaluated result in comparable pregnancy rates to TAI in dairy heifers.

A retrospective analysis evaluated pregnancy rates and embryo losses with in vitro-produced embryos in a commercial embryo transfer program on 15 different beef farms. Recipients were beef cows and heifers (n = 1841) that were synchronized with 5 different protocols and transferred at a fixed-time (FTET). Recipients were examined by ultrasonography on Day 0, and those with a corpus luteum (CL) or a follicle ≥8 mm in diameter and with body condition score 2 to 4 (1 to 5 scale) were synchronized. The synchronization treatments were as follows. (T1) Recipients received an intravaginal device with 0.5 g of progesterone plus 2 mg of oestradiol benzoate on Day 0; device removal, plus 500 μg of cloprostenol (prostaglandin F2α), 400 IU of eCG, and 0.5 mg of oestradiol cypionate on Day 8; and FTET on Day 17. (T2) This treatment was similar to T1 but 1 mg of oestradiol cypionate was injected at device removal instead of 0.5 mg of oestradiol cypionate. (T3) This treatment was similar to T1 except that animals were tail-painted on Day 8 and observed on Day 10. Those with the tail-paint intact on Day 10 received 100 μg of gonadorelin (gonadotropin-releasing hormone) and all recipients were FTET on Day 17. (T4) Recipients received a progesterone device on Day 0; device removal, prostaglandin F2α, and eCG on Day 5; gonadotropin-releasing hormone on Day 8; and FTET on Day 15. (T5) Recipients received a progesterone device and 2 mg of oestradiol benzoate on Day 0; device removal, prostaglandin F2α, and eCG on Day 6; gonadotropin-releasing hormone on Day 9; and FTET on Day 16. On the day of FTET all recipients with CL ≥18 mm in diameter (G1), ≥16 and <18 mm in diameter (G2), and ≥14 mm and <16 mm in diameter (G3) received in vitro-produced fresh embryos. Pregnancy was diagnosed by ultrasonography at 30 and 60 days of gestation, and data were analysed by logistic regression. The overall proportion of recipients synchronized that were FTET was 80.8% (1487/1841), with a 30-day pregnancy rate to FTET (P/FTET) of 45.6% (678/1487) and the rate of 30- to 60-day embryo/fetal loses on the 528 recipients that were re-checked at 60 days was 12.8% (68/528). There were no significant differences in P/FTET among operators, animal category, time of the year, embryo stage, or body condition score; however, there was a significant effect of farm (P < 0.001) and CL diameter (P < 0.05), but no interaction between CL diameter and farm or treatment (P > 0.1). Recipients with G1 (443/953, 46%) and G2 (221/462, 47%) CL had higher pregnancy rates than those with G3 CL (23/71, 32%). There was a significant effect of synchronization treatment on the proportion of recipients transferred and on P/FTET (P < 0.01) that was highly influenced by farm (farm by treatment interaction P < 0.01). The proportions of recipients selected for embryo transfer were as follows: T1: 386/486, 79.4%; T2: 233/331, 70.3%; T3: 342/377, 90.7%; T4: 126/160, 78.7%; and T5: 400/487, 82.1%. The P/FTET were as follows: T1: 190/386, 49.2%; T2: 96/233, 41.2%; T3: 175/342, 51.1%; T4: 49/126, 38.8%; and T5: 168/400, 42.0%. Although 30- to 60-day embryo/fetal losses were not influenced by synchronization treatments, they were highly influenced by farm (P < 0.001) and ranged from 0 to 34.5%. In conclusion, P/FTET in a commercial program with beef in vitro-produced embryos was influenced by factors related to the recipient (CL diameter) and the environment (farm), whereas embryo/fetal losses were influenced by farm but not treatment or recipient factors.

In pasture-based dairying systems it is important to maintain a 365-day calving interval, which requires that cows have a rapid resumption of postpartum ovulatory activity and high conception rates. The major form of infertility in New Zealand (NZ) dairy cows is an extended postpartum anovulatory interval (ppai; Macmillan, 2002), a problem that can be exacerbated by low body condition score (BCS) at calving (McDougall, 1995). Furthermore, Holstein cows, originating from North American genetic strains (Overseas; OS), which have been widely used in NZ in recent years, have longer intervals to first mating and conceive later than do NZ strains, suggesting a possible delay in the initiation of postpartum cyclicity (Harris and Winkleman, 2000). Worldwide, there is concern over an apparent decline in the fertility of Holstein-Friesian dairy cattle (Butler et al., 1995; Royal et al., 2000). The purpose of this trial was to determine whether there was a difference in ppai and BCS between NZ and OS Holstein Friesians that may affect reproductive performance.

The objectives of the study were to compare the ovarian activity of Holstein-Friesian (CH HF), Fleckvieh (CH FV) and Brown Swiss (CH BS) dairy cows of Swiss origin with that of Holstein-Friesian (NZ HF) dairy cows of New Zealand origin, the latter being used as a reference for reproductive performance in pasture-based seasonal calving systems. Fifty, second-lactation NZ HF cows were each paired with a second-lactation Swiss cow (17, 15 and 18 CH HF, CH FV and CH BS respectively) in 13 pasture-based, seasonal-calving commercial dairy farms in Switzerland. Ovarian activity was monitored by progesterone profiling from calving to first breeding service. CH BS cows produced less energy-corrected milk (mean 22·8 kg/d) than the other breeds (26·0–26·5 kg/d) during the first 100 d of lactation. CH HF cows had the lowest body condition score (BCS) at calving and the greatest BCS loss from calving to 30 d post partum. Commencement of luteal activity (CLA) was later for NZ HF than for CH FV (51·5 v. 29·2 d; P <0·01), with CH HF and CH BS intermediate (43 d). On average, NZ HF and CH HF cows had one oestrous cycle before the onset of the seasonal breeding period; this was less (P<0·01) than either CH FV (1·7) or CH BS (1·6). There was a low prevalence of luteal persistency (3%) among the studied cows. First and second oestrous cycle inter-ovulatory intervals did not differ between breeds (20·5–22·6 d). The luteal phase length of CH BS during the second cycle was shorter (10·6 d) than that of the other breeds (13·8–16·0 d), but the inter-luteal interval was longer (9·8 d v. 7·0–8·0 d). The results suggest that the Swiss breeds investigated have a shorter interval from calving to CLA than NZ HF cows.
