Hypoglycemic and Hypocholesterolemic Potential of Wheat Grass Juice Extract and Powder on Diabetics

The study was designed to evaluate the hypoglycemic and hypocholesterolemic potential of wheat grass juice, extract and powder on diabetics. Two hundred male employees (35-50 y) of Tamil Nadu Electricity Board (TNEB) from Usilampatti, a town in Madurai district, Tamil Nadu were selected for the study. After screening, 60 employees whose fasting blood glucose level was ≥ 145mg/dl and not taking oral hypoglycemic drugs or insulin were considered for the study. Fifty grams of fresh wheat grass, made into a fine paste using a stone grinder was mixed in 100ml of water. Two to three drops of lemon juice was added to enhance the flavour and palatability of the juice. Wheat grass extract obtained was then squeezed and the fiber separated. Fresh wheat grass was harvested and shade dried for 2-3 days. It was then made into fine powder with a stone grinder. Five grams of wheat grass powder was added in 100ml of water and mixed thoroughly. The selected diabetic employees (60) were divided into three experimental groups and one control group of 15 members each. Experimental group I was supplemented with 100ml of wheat grass juice, group II was supplemented with 100ml of wheat grass extract and group III was supplemented with 5g of wheat grass powder mixed in 100ml of water and for a period of four months. No supplementation was given to control group. Anthropometric parameters like height, weight, Body Mass Index (BMI), waist / hip ratio, skin fold thickness and biochemical parameters namely fasting blood glucose level, post prandial blood glucose level, total cholesterol, LDL, VLDL, HDL, HbA<sub>1</sub>C and blood pressure were measured before and after supplementation. Group II supplemented with wheat grass extract showed a slight difference in weight when compared to groups I and III. The mean skin fold thickness of all the three groups decreased after supplementation. The mean initial blood glucose level of the experimental groups (I, II and III) and control group were above the normal range . The mean decrease of fasting blood glucose level in experimental group I, II, III and control group were 37.4, 32.9, 27.6 and 1.4 mg /dl respectively. The mean decrease of post prandial blood glucose level in experimental groups I, II, III and control group were 63, 60.3, 39 and 1.8 mg/dl respectively. High fibre supplement lowered total cholesterol by about 19.2 % and reduced LDL cholesterol by 13.2 % with maximum reduction in groups II and III. HDL cholesterol level was highly increased in experimental group II. The decrease in HbA<sub>1</sub>C level was higher in experimental group I (2.5%) and blood pressure was highly decreased in experimental group I compared to other experimental groups and control group.

The purpose of this project is to study the anti-diabetic effect of on a diabetic rat model. A total of Twenty male Sprague rats were used and it randomly distributed into four groups which are Group I: , Group II: negative control, Group III: and Group IV: and . In diabetic model were induced with via injection at the dosage of 65mg/kg. and FBG (Fasting Blood Glucose) level of diabetic rats were assessed every three days. Blood was collected via cardiac puncture at day 21 after the induction of treatment. Insulin level of the rats was assessed with the Mercodia Rat Insulin ELISA kit. FBG level of group I (12.16 ±3.96, p&lt;0.05) and group IV (11.34 ±3.67, p&lt;0.05) were significantly decreased. Meanwhile, the for all rats did not show any significant increase. However, the insulin level was escalated in group IV (0.74+0.25, p&lt;0.05) significantly. The present study shows that the and the combination of and lowered blood glucose level and enhanced insulin secretion.

Objective. To evaluate the importance of determining fasting glycemia at the first prenatal visit for timely diagnostics of gesta- tional diabetes mellitus (GDM). Material and methods. The study included 577 pregnant women examined with the use of the oral glucose tolerance test (OGTT) during weeks 24-28 of pregnancy. The results of the test were compared with the fasting glucose level at the first prenatal visit. Results. The mean fasting blood glucose level at the first prenatal visit was 4.70±0.61 mmol/l. In 25% of the patients, fasting glycemia at this time was 5.1 mmol/l or higher. Only in 36% of the women (52 of the145 ones) with the fasting blood glucose level ≥5.1 and <7 mmol/l at the first prenatal visit, the results of OGTT were consistent with the criteria for GDM. The mean fasting blood glucose level at the first prenatal visit (up to week 16) was significantly higher than on weeks 24-28 (4.71±0.61 and 4.53±0.69 mmol/l respectively, p<0.001). The ROC analysis failed to reveal the fasting blood glucose level that could be used to predict the development of gestational diabetes with a reliable sensitivity and specific- ity. Conclusion. Further population-based investigations are needed to elucidate the relationship between fasting glycemia in the first trimester of pregnancy and its unfavourable outcome.

Context: Scientific studies revealed the hypoglycaemic properties of momordica charantia. The present study was carried out to find out microscopically whether Momordica charantia (karela) has got any impact lowering of FBG (Fasting Blood Glucose) level in diabetes mellitus. Study type: an experiemental study. Setting: Anatomy department of the then IPGMR (Institute of Post Graduate Medicine and Research) at present BSMMU (Bangabandhu Sheikh Mujib Medical University) and BIRDEM (Bangladesh Institute of Research and Rehabilitation in Diabetes, Endocrine & Metabolic Disorders). Subjects: Sixty five healthy young Long Evans rats of male sex weighing 150 to 280gm aged between 10 to 12 weeks were used in this study. Methods: The rats were divided into four equal groups depending on their different sorts of dietry feeding and drug treatment. Main outcome measures: variation of differential FBG level in different groups of rat. Result: Mean 'initial' and 'final' (on day 7 and day 51 from Streptozotocin/vehicle injection) fasting blood glucose (FBG) level in the control group (Group-A) was 7.872 ± 0.60 and 8.55 ± 0.82 respectively. Therefore the mean (FBG) increased by about 13% (P = 0.022*) which is higher than that of the initial value. In untreated diabetic group the mean initial (FBG) level was 25.95 ± 8.90 and the mean final was 24.02 ± 4.08. So here, the (FBG) level decreased by about 13% (P = 0.557). On the other hand, in the insulintreated diabetic rats the mean initial (FBG) level was 24.35 ± 6.81 and the mean final was 8.38 ± 5.02, which is lower (P = 0.000*) & in the karela–treated diabetic rats, the initial (FBG) level was 23.03 ± 5.70 and the mean final was 5.65 ± 1.29 which is lower* (P = 0.000*). The value in the insulin-treated diabetic rats & in the karela-treated diabetic rats were significantly lower than that of the untreated diabetic rats (P = 0.007) & (P= 0.005) respectively. But there was no significant difference between the insulin-treated diabetic rats & the karela-treated diabetic rats (P = 0.605) in this regard. Conclusion: Karela showed a tendency of acting against hyperglycemic effects of Streptozotocin-induced diabetes mellitus. However, further investigations are recommended for establishing karela as a safe, useful effective anti- hyperglycemic agent as well as antidiabetogenic agent. Objective: To find out the effects of ginger (Zingiber officinale) juice blood glucose in alloxan induced-diabetes mellitus in rats. Methods: This experimental study was done in the Department of Pharmacology & Therapeutics, Dhaka Medical College, Dhaka, in collaboration with the Departments of Pathology, Ibrahim Medical College, Dhaka between January and December 2009. This experimental animal study was divided into two parts, which were Experiment-1 and Experiment-2. Experiment 1 comprises of 12 rats and divided into 2 groups each group having 6 rats. Rats of group-A was non-diabetic normal control group and group-B was fed with ginger (Zingiber officinale) in a dose of 4ml/kg body weight orally through Ryle’s tube. Experiment-2 comprised of 12 rats divided into 2 groups each containing 6 rats labeled as group C, group D. Rats of groups C administered alloxan 150 mg/kg intraperitoneally on the 2nd day of the study. Rats of group D were administered alloxan 150mg /kg intraperitoneally and ginger (4ml/kg of body weight orally) on the 2nd day of the study. Results: The fasting blood glucose level at day 12 in the rats treated with ginger (Zingiber officinale) 4 ml /kg body weight orally daily for 12 days showed reduction in fasting blood glucose level as compared to control group, but not significant, which indicates that ginger has no effect in lowering blood glucose of normal rats. The fasting blood glucose levels at day 12 in the rats of group D (treated with ginger and alloxan) showed highly significant reduction in fasting blood glucose level as compared to diabetic control group (p<0.002). Conclusion: Consumption of ginger produced a significant antihyperglycemic effect in experimentally induced diabetic rats. Objective: To determine the frequency and severity of hypoglycemia in patientswith liver cirrhosis. Study Design: Cross sectional case series study. Period: Six months.Setting: Liaquat University Hospital Hyderabad. Methods: All the patients of liver cirrhosis,of >12 years of age and of either gender were evaluated for hypoglycemia by assessing theglycemic status through random or fasting blood glucose level. The severity of liver cirrhosiswas identified according to the Child-Pugh classification whereas the severity of hypoglycemiawas grouped in mild, moderate and severe categories. The data was entered and saved inSPSS and frequency and percentage was calculated for hypoglycemia in patients with livercirrhosis. The stratification was done for age, gender, hypoglycemia and severity of the diseaseand hypoglycemia. The chi-square test was applied between categorical variables at 95%confidence interval and p -value ≤0.05 was considered as statistically significant. Results:During six months study period, total 100 cirrhotic subjects were studied for hypoglycemia,of which 59% were males and 41% were females. The mean ± SD for age in all (100)cirrhotic patients was 42.33 ± 8.87 while the mean ± SD for age in male cirrhotic patientswas 44.06±11.45 where as in female cirrhotic subjects it was 39.92±12.55 respectively. Thehypoglycemia was observed in 67%, of which 45(67.2%) were males and 22(32.8%) werefemales. The mean random blood glucose level in male and female hypoglycemic cirrhoticpatients was 67.88±8.43 and 65.62±6.75 while the mean fasting blood glucose level in maleand female hypoglycemic cirrhotic patients was 52.93±5.31 and 53.64±8.73 respectively. Outof sixty seven hypoglycemic cirrhotic subjects 45(67%) were males and 22(33%) were females.Of sixty seven, 32(47.8%) had moderate hypoglycemia while 30/67(44.8%) were in Child-Pughclass B (p<0.05). Conclusions: The hypoglycemia was detected in patients with liver cirrhosis,hence frequent blood glucose monitoring is one of the most important way to detect mildhypoglycemia and prevent serious and severe episodes.

The present study was designed to investigate the antidiabetic activity of aqueous extract ofAcacia tortilispolysaccharide (AEATP) from gum exudates and its role in comorbidities associated with diabetes in STZ-nicotinamide induced diabetic rats. Male albino Wistar rats were divided into control, diabetic control, glimepiride treated (10 mg/kg), and diabetic rats treated with 250, 500, and 1000 mg/kg dose of AEATP groups and fasting blood glucose, glycated hemoglobin, total cholesterol, triglyceride, LDL, VLDL, HDL, SGOT, and SGPT levels were measured. STZ significantly increased fasting blood glucose level, glycated hemoglobin, total cholesterol, triglyceride, LDL, VLDL, SGOT, and SGPT levels, whereas HDL level was reduced as compared to control group. After 7 days of administration, 500 and 1000 mg/kg dose of AEATP showed significant reduction(P<0.05)in fasting blood glucose level compared to diabetic control. AEATP has also reduced total cholesterol, triglyceride, LDL, VLDL, SGOT, and SGPT levels and improved HDL level as compared to diabetic control group. Our study is the first to report the normalization of fasting blood glucose level, lipid profile, and liver enzyme in AEATP treated diabetic rats. Thus, it can be concluded that AEATP may have potentials for the treatment of T2DM and its comorbidities.

Curcuma longa (C. longa), also known as curcumin, is a lipophilic polyphenol substance proven to have cholesterol-lowering, anti-diabetic, anti-inflammatory, anti-oxidant, and anti-cancer properties in both in vitro and in vivo models. Most previous studies investigated the effect of C. longa on diabetic mice and therefore, there is a need to investigate the effect of C. longa on normoglycemic mice. Depression is a common consequence of anxiety that affects 21% of the world’s population. Since the prevalence of diabetes and depression is on the rise globally, it is important to search for safer and cost-effective management for these disorders. In doing so, it is therefore essential to investigate its effect in normoglycemic mice. The current study determines the effect of C. longa on blood glucose level and anxiety-like behavior in normoglycemic Swiss albino mice. A total of 20 mice were divided into four groups of five (n=5 per group). Group I (control) received distilled water 10 ml/kg, groups II, III, and IV received C. longa at 5%, 10%, and 20%, respectively, for 14 days. We found that 20% C. longa group showed a significant (p<0.05) increase in fasting blood glucose level (195.84±14.46 mg/dl) after 14 days of administration compared with the control group (134.60±4.52 mg/dl).  We also found that 20% C. longa increased the anxiety-like behavior in normoglycemic Swiss albino mice compared with the control group. However, there was no significant (p>0.05) difference in both fasting blood glucose level and anxiety-like behavior between the mice treated with 5% and 10% C. longa and the control group. This study indicates that C. longa at high concentration is unsafe for consumption by normoglycemic Swiss albino mice.

Abstract Squaleneis a chemical compound that has been reported to have antidiabetic activity. The present study aimed to investigate the effect of squalene on fasting blood glucose level (FBGL) in type 2 diabetic rats. Diabetes type II in rats was obtained by giving nicotinamide (120 mg/kg) before high dose streptozotocin (60 mg/kg) intraperitoneally. A total of 18 diabetic rats were divided into 3 groups and served once daily for 12 days as follows; Group I Aquades (Diabetic Control) 10 ml/kg, Group II (Metformin 45 mg/kg) and Group III (Squalene 160 mg/kg). FBGL was measured at day 0, day 6 and day 12. The results showed that FBGL in both Squalene- (194.67 ± 28.32 mg/dL) and Metformin- (178.50 ± 34.27 mg/dL) were significantly decreased after 12 days intervention compared to Diabetic Control-treated groups (438.33 ± 65.79 µmol/L) with p<0.001. This study concluded that squalene was able to decrease FBGL in type II diabetic rats.

Objective: The present study was aimed at the development of partial pancreatectomy in a murine model for diabetes.Methods: Diabetes model was successfully developed by partial pancreatectomy method. In this study, cyclosporine was used for influenced the glycaemic status. Diabetes status was evaluated by fasting blood glucose level (FBG), lipid profile (by estimation of total cholesterol level and HDL-level), serum amylase and biochemical assay like glutathione estimation.Results: We report here the restoration of euglycaemic status in cyclosporine-induced diabetic in swiss albino mice after 30% pancreatectomy. In this study, Pancreatectomised (partial) group of animals showed a rapid elevation of glycaemic status, starting from 15th post observational d, but the level of significance decreased gradually from 15th (P<0.01) to 60th (P<0.05) d. This was probably due to nesidioblastotic activity which shifted the fasting blood glucose level towards normoglycaemic status with β-cells regeneration. Cyclosporine treated a group of mice showed normoglycaemic status throughout the whole experimental period, but the cholesterol level remained significant (P<0.001) till the end of the experimental d. Gradually decrements in glycaemia of the diabetic pancreatectomised animals demonstrate islets neogenesis occurring after the operative activity, leading to normoglycaemic condition, probably attributed to β-cells proliferation.Conclusion: The biochemical and histopathological evaluations suggest that there is the development of the diabetic model in the pancreatectomized group and diabetes status induced by pancreatectomy is curable to a certain extent due to the regeneration of β-cells.

Challenges in lifestyle, such as increasesin energy intake and decreasesin physical activity are causing overweight and obesity leading to epidemic increases in type II Diabetes Mellitus. The research approach used for this study was evaluative approach and the research design was true experimental design. 60 patients with type II diabetes, 30 in experimental group and 30 in control group were selected for this study by using purposive sampling technique. Data was collected with the help of self-structured interview schedule. Descriptive statistics (frequency, percentage, mean and standard deviation) and inferential statistics (chi-square, paired ‘t’ test) were used to analyse the data and to test the hypotheses. In the experimental group,the pre-test mean score was 2.966, mean percentage was 59% and standard deviation was 1.129 and in post-testmean score was 2.533, mean percentage was 50.66% and standard deviation was 1.074 with effectiveness of 8.34% and paired‘t’ test value of t=3.971,which was statistically significant (p<0.05) which is an evidence ofthe effectiveness of Amla juice in reducing blood glucose level. Comparison of blood glucose levels in experimental and control groups, shows that the value is statistically highly significant, as was observed from the unpaired ‘t’ test value of 13.39 with P value of <0.05, which is an evidence indicatingthe effect of Amla juice in reducing postprandial blood glucose levels. The resultsfound that the administration of Amla juice did have aneffect in reducing blood glucose level in the experimental group. By comparing the findings of pre-test and post test between the experimental group and the control group,the effect was identified (assessed). The study concluded that the Amlajuice is effective in reducing blood glucose level.
