67 patients (mean age 48 ± 1 year) with diffuse liver diseases of various origins were examined. It has been established that an increase in the degree of fibrosis is associated with an increase in the resonance intensity of glucose-6-phosphates, adenosine monophosphate (AMP), inosine monophosphate (IMP), pyrophosphates, 2,3-DPH and a decrease in the resonances of inorganic phosphates and phosphocreatine, γ-, alpha- and beta- ATP, -UTP, Hb- and Mg-ATP, as well as beta- and alpha-ADP. The predictive value of a positive result of erythrocyte 31P NMR spectroscopy for liver fibrosis is 92.3% (sensitivity 85.71%, specificity 80%). In the IR spectra of the blood serum of patients with severe fibrosis, in contrast to moderate fibrosis, the resonance intensity is higher at 1280, 3190 and significantly lower at 776, 818, 889, 966, 1399.1450, 1570, 1635 cm-1, which indicates the presence of protein molecules with a secondary structure mainly in the form of a helix (alpha-helix) compared with the structure in the form of a folded sheet (beta-sheet) (p <0.001-0.05). The combined use of 31P NMR spectroscopy of erythrocyte suspensions and IR spectroscopy of blood serum made it possible to increase diagnostic accuracy (96%), sensitivity (97%) and specificity (91%) in distinguishing between degrees of liver fibrosis in contrast to the isolated use of these methods.

The effects of long-term exposure (5 h) of Phycomyces blakesleeanus mycelium to 5 mM KCN on respiration and phosphate metabolites were tested. Exposure to cyanide, antimycin A and azide lead to a decrease in the activity of cyanide-sensitive respiration (CSR), and the ratio of core polyphosphates (PPc) and inorganic phosphates (Pi), which is a good indicator of the metabolic state of a cell. After 5 h of incubation, the activity of CSR returned to control values. For this, the recovery of cytochrome c oxidase (COX) was required. In addition, the PPc/Pi ratio started to recover shortly after initiation of COX recovery, but never reached control values. This led us to conclude that the regulation of polyphosphate (PPn) levels in the cell is tightly coupled to respiratory chain functioning. In addition, acutely applied cyanide caused two different responses, observed by 31P NMR spectroscopy, that were probably mediated through the mechanism of glycolytic oscillations, triggered by the effect of cyanide on mitochondria.

Phosphorus is one of the most important elements essential for all living beings. Plants accumulate and store phosphorous in various forms that have diverse physiological and biochemical functions. In this study,we determine and then examine the phosphorus profiles of seeds of plants belonging to different taxa based on extractable inorganic phosphates and organic forms of phosphorus. We paid particular attention to the presence of natural phosphonates in the tested materials. The inorganic phosphates were determined colorimetrically, whereas phosphorus profiles were created by using 31P NMR spectroscopy. Our study on phosphorus profiles revealed that the obtainedsets of data vary significantly among the representatives of different taxa and were somehow specific for families of plants. It should be emphasised that the measurements obtained using 31P NMR spectroscopy undoubtedly confirmed—for the first time—the presence of phosphonates among the natural components of plant seeds. Hence, the classification of plants considering the phosphorus profiles, including the presence of phosphonates, may be a new additional chemotaxonomic feature.
