This study was undertaken during the favourable breeding season of the year 2016-17 on healthy mature Gir cattle and Surti buffalo bulls, three each, at Sperm Station of the College. The ejaculates (9/bull, total 54) collected in the morning using artificial vagina were evaluated for routine seminal attributes, including acrosomal and plasma membrane integrity. The mean values of ejaculate volume, sperm concentration, mass activity (0-5 score), individual sperm motility, live sperm, abnormal sperm, intact acrosome and HOST reactive sperms observed in fresh semen of Gir cattle and Surti buffalo bulls were 6.20±0.42 and 3.34±0.23 ml (P less than 0.01), 1169.44±61.71 and 846.30±54.82 million (P less than 0.01), 3.44±0.09 and 3.42±0.08, 76.53±0.53 and 80.76±0.39 % (P less than 0.05), 81.00±1.32 and 84.73±0.78 % (P less than 0.05), 6.00±0.37 and 5.81±0.40 %, 95.59±0.35 and 95.54±0.25 % as well as 80.30±1.90 and 84.58±0.88 % (P less than 0.05), respectively. The variation between cattle and buffalo semen was significant for most of these traits. The variations between bulls within breed were however not significant. Significant correlations were observed only between mass activity and initial motility (0.62), and live and abnormal sperm (-0.41) in Gir bulls, and for ejaculate volume with sperm concentration (-0.56) and abnormal sperm (0.45), and between live and HOS reactive sperms (0.48) in Surti bulls.

The study was conducted on four Jafarabadi breeding bulls, 5-6 years old to know the fresh and post-thawed seminal characteristics based on total of 192 semen ejaculates evaluated and cryopreserved over one year period. The mean values of fresh neat seminal characteristics of Jafarabadi bulls, viz., ejaculate volume (ml), colour/density (score), sperm concentration (million/ml), mass activity (score), initial motility (%), live sperm (%), abnormal sperm (%), HOS reactive sperm (%) and acrosomal integrity (%) were 5.19±0.18, 2.38±0.10, 1253.36±24.75, 3.73±0.05, 80.31±0.05, 86.20±0.64, 5.00±0.40, 85.75±0.43 and 93.56±0.56, respectively, whereas the mean post-thawed sperm characteristics, viz., progressive sperm motility, live sperm (%), abnormal sperm (%), HOS reactive sperm (%), acrosomal integrity (%) and first insemination conception rate (%) observed were 57.60±0.36, 66.34±0.53, 8.85±0.33, 56.97±0.46, 75.26±0.17 and 44.63±0.14, respectively. The semen quality of fresh and post-thawed samples observed was within normal limit for use in breeding program with satisfactory first insemination conception rate.

A study was carried out on semen ejaculates (40) of five healthy Surti buffalo bulls during favourable breeding season. The ejaculates with >70% IM were diluted @ 100 million sperms/ml in Tris-fructoseegg yolk-glycerol (TFYG) diluent supplemented with L-cysteine @ 1 mg/ml, and examined for quality parameters such as sperm progressive motility, viability, morphology, acrosomal and plasma membrane integrity. A part of the extended semen was preserved at 5°C and another was processed for ultra-low temperature (-196°C) preservation in LN2 using biofreezer after filled in French mini straws. The mean values of ejaculate volume, sperm concentration, mass activity (0-5 score), individual sperm motility, live sperm, abnormal sperm, intact acrosome and HOS reactive sperms observed in fresh semen were 3.04±0.11 ml, 963.05±50.97 million/ml, 2.97±0.06, 75.00±0.69 %, 88.22 ±0.48 %, 4.40±0.26 %, 93.67±0.31 % and 89.17±0.57 %, respectively. Just after dilution the percentages of progressively motile, live & abnormal sperms, intact acrosomes and HOS reactive sperms were 80.51±0.65, 85.95±0.65, 4.92±2.20, 91.00±2.30, 85.72±0.67, respectively. The corresponding values after 48 hrs of refrigeration (5°C) were 61.75±0.85, 67.85±0.85, 6.45±0.27, 89.05±0.32, 67.30±0.96%, the values at pre-freeze stage (after equilibration) were 72.62±0.69, 78.97±0.93, 5.45±0.25, 90.90±0.35, 78.12±0.79% and at post-thaw stage (37°C/30 sec) 46.50±0.72, 52.97±0.79, 7.10±0.26, 85.73±0.18, 51.62 ±0.82%, respectively. The mean motile spermatozoa observed after 1, 2 and 3 h of post-thaw incubation at 37°C in water bath were 32.75±0.82, 18.88 ±0.70 and 8.88±0.66% (P<0.01), respectively. The semen quality parameters, fresh and cryopreserved were acceptable for artificial breeding use. The seminal traits in initial, 48 hrs refrigerated, pre-freeze and post-thawed samples revealed significant (p<0.01) interrelationships (r = 0.44 to 0.84) between progressive motile sperm, live sperm and HOST reactive sperm directing more emphasis on these quality parameters for better semen evaluation.

Semen was collected from four rams, using artificial vagina and viability%, motility% and plasma membrane integrity% were measured. Fresh ejaculates (n = 32) were separated by modified swim-up separation using modified human tubal fluid medium. Four fractions of supernatant were collected at 15-minute intervals. The mean volume, mass activity, concentration, motility%, viability%, normal morphology and membrane integrity% (HOST +ve) of fresh semen were 1.0 ± 0.14, 4.1 ± 0.1 × 109 spermatozoa/ml, 85.0 ± 1.3, 89.4 ± 1.0, 85.5 ± 0.7, 84.7 ± 0.5 respectively. There was no significant (P>0.05) difference in fresh semen quality parameters between rams. The motility%, viability% and HOST +ve % of first, second, third and fourth fractions were 53.4 ± 0.5, 68.2 ± 0.3, 74.8 ± 0.3 and 65.5 ± 0.4; 55.5 ± 0.4, 66.2 ± 0.4, 74.5 ± 0.3 and 73.6 ± 0.3 and 66.7 ± 0.5, 66.8 ± 0.5, 65.2 ± 0.4 and 74.7 ± 0.5 respectively. The motility%, viability% and membrane integrity% of separated semen samples differed significantly (P<0.05) between four fractions. The mean motility% and viability% were significantly higher (P<0.05) in third fraction (74.8 ± 0.3%), whereas the mean HOST +ve% was significantly higher (P<0.05) in fourth fraction (74.7 ± 0.5). All quality parameters of separated spermatozoa were significantly (P<0.05) lower than that of fresh semen. This study was undertaken on three mature bulls each of Gir cattle and Surti buffalo breeds to evaluate the comparative efficacy of egg yolk based standard TFYG (Tris-citrate-fructose-yolk-glycerol) extender and egg yolk free soybean based commercial extenders Optixcell® (IMV, France) and Andromed® (Minitube, Germany) under split-sample technique. The ejaculates (9/bull) were extended @ 100×106 sperm ml-1 with three extenders and frozen using biofreezer following 4 hr of equilibration. The pooled means of progressively motile sperm observed (irrespective of extenders) at initial, pre-freeze and post-thaw stage in Gir bulls semen were 76.53±0.53, 71.11±0.53 and 39.86±0.90% and in Surti buffalo 80.76±0.39, 74.65±0.45 and 40.35±1.07%, respectively. The corresponding values for live sperm were 75.64±0.76, 69.01±0.97 and 47.99±1.11 % for Gir and 80.90±0.45, 75.76±0.48 and 52.33±0.86 % for Surti buffalo; and those of intact acrosome 94.29±0.25, 90.29±0.27 and 79.29±0.33 % for Gir bulls, and 93.94±0.21, 89.94±0.23 and 78.95±0.26 % for Surti buffalo semen, respectively. The HOS reactive sperm at initial, pre-freeze and post-thaw stage were 76.18±0.74, 71.04±0.76 and 27.90±0.70 % for Gir, and 81.83±0.35, 76.47±0.39 and 27.83±0.68 % for Surti bulls, respectively. The overall mean post-thaw incubation (37°C) survival of spermatozoa observed at 60, 120 and 180 min were 28.40±0.91, 17.78±0.86 and 9.44±0.72% for Gir bulls semen, and 28.01±0.99, 18.40±1.01 and 10.51±0.93% for Surti buffalo semen, respectively. Optixcell was proved superior, and at par with TFYG, than the Andromed in maintaining greater motility, viability, morphology, acrosomal/plasma membrane integrity including post-thaw sperm longevity of cattle and buffalo spermatozoa with significant differences only in sperm motility and post-thaw longevity. The motile, live and HOST reactive sperm were significantly higher in buffalo semen than cattle at initial and pre-freeze stage, but not at post-thaw stage. The results showed that egg yolk free commercial Optixcell extender and egg yolk based TFYG extender were at par in terms of most of the sperm quality traits, hence any one of them can be preferred over Andromed for successful routine cryopreservation of cattle and buffalo semen.

Sixty semen ejaculates from 10 mature bulls, 5 each of Jafarabadi and Mehsana breed, were studied for sperm motility and velocity parameters of fresh and frozen-thawed spermatozoa using computer assisted sperm analyzer (CASA). The mean values of motile and progressively motile spermatozoa observed in fresh semen of Jafarabadi and Mehsana bulls (79.77±1.62 and 61.80±1.85, and 78.90±1.22 and 61.37±1.58%) were highly significantly (P<0.01) reduced (51.20±1.57 and 33.20±1.45, and 52.10±1.70 and 34.30±1.54 %, respectively) in post-thawed semen. The average path velocity, straight line velocity and curvilinear velocity (μm/sec) of spermatozoa of Jafarabadi and Mehsana bulls noted in fresh semen were also reduced highly significantly (P<0.01) in frozen-thawed semen. Among the other velocity parameters, amplitude of lateral head displacement (μm), elongation (%) and medium motile sperm (%) increased, while beat-cross frequency (Hz), straightness (%), linearity (%), sperm area (μm<sup>2</sup>) and rapidly motile sperm (%) decreased significantly in post-thawed sperms when compared with the fresh sperm of both Jafarabadi and Mehsana bulls. The initial motility and live sperm per cent were significantly correlated with CASA traits of fresh and frozen-thawed semen, and all the sperm motility and velocity traits of fresh and frozen-thawed semen assessed by CASA were significantly interrelated among both the breeds. The interrelationships were stronger in Mehsana bulls as compared to Jafarabadi bulls.

The present study was carried out on 80 ejaculates, 20 each from four healthy Jaffrabadi breeding bulls to see the relationship betweensexual behavior and semen quality parameters. The results depicted that the overall mean values of libido score, mating ability score andsexual libido score between bulls. The overall color of semen ranged from thin white to very thick creamy. The overall values for semen volume(ml), pH, sperm concentration (million/mL), mass activity, individual sperm motility (%) and live spermatozoa (%) were 2.99 ± 0.12, 6.80 ± 0.01, 1439.68 ± 48.46, 3.94 ± 0.03, 89.51 ± 0.21 and 95.00 ± 0.19, respectively. The values for semen volume, sperm concentration,mass activity (%) and live sperm (%) differed significantly (p less than 0.05) between bulls. There were significant positive correlations (p LESS THAN 0.01)of libido with mating ability, sexual behavior score and ejaculate volume of semen (r = 0.37 to 0.87), while a negative correlation wasfound with sperm concentration (-0.30). Mating ability was significantly (p less than 0.01) correlated with sexual behavior score and ejaculatevolume (0.78, 0.67), and sexual behavior was correlated with semen volume (0.84). Sperm concentration was significantly (p less than 0.05 correlated with mass activity (0.29), while individual sperm motility was positively (p less than 0.05) correlated with live sperm percentage (0.25).Correlations of libido, reflected the importance of sexual behavior of Jaffrabadi bulls in predicting their future utility as a proficient breeder under AI program.

A total of 120 ejaculates from six adult Black Bengal bucks were studied to determine the semen attributes and evaluate their fertility. The various semen attributes of fresh semen were volume, sperm concentration, mass motility, live spermatozoa and normal spermatozoa percentage. All the semen attributes differed significantly (P<0.05) except normal spermatozoa percentage between the bucks. A total of 129 goats were inseminated with diluted semen to assess the fertility rate of Black Bengal buck. The mean values for the various semen attributes were: volume 0.58 ± 0.03 ml; sperm concentration 2797.22 ± 18.73 × 106/ml; mass motility 77.82 ± 0.61%; live spermatozoa 86.72 ± 0.54% and normal spermatozoa 91.39 ± 0.24%, respectively in fresh semen. With regard to the diluted semen, individual bucks showed significant difference (P<0.05) on motility, live and normal spermatozoa percentages. The diluted semen motility varied from 68.64 ± 1.66 to 74.55 ± 1.06%. On the other hand, the live and normal spermatozoa percentages of the diluted semen varied from 83.73 ± 0.94 to 89.27 ± 1.40% and 88.73 ± 0.54 to 91.91 ± 0.78%, respectively. From the insemination trial, the average kidding rate obtained was 58.9% in Black Bengal goat. The objective of the study was to assess the semen characteristics of Jersey, Mithun and Siri breeds of bulls and to compare the suitability of Tris Eggyolk Citrate and commercial soya lecithin based BIOXcell extenders for cryopreservation of semen from the these breeds of bulls. The mean semen volume was 4.55± 1.57, 4.86±1.33 and 4.83±1.72 ml in Jersey, Mithun and Siri bulls, respectively. Mean mass activity of 3, mean initial motility of 78.0±4.22, 80±4.47 and 78.33±4.08%, mean sperm concentration of 1416±484.19, 1530.91±517.41, 1122.83±293.68 x106 and mean livability of 69.20±6.81, 70.45±8.42, 76.67±8.17% was found in Jersey, Mithun and Siri bulls, respectively. There is no significant difference in all above mentioned variables among the breeds. The post-thaw motility of semen cryopreserved in Tris Eggyolk Citrate extender was found to be 45.50 ± 4.38, 46.82 ± 6.81 and 47.50 ± 8.22% in Jersey, Mithun and Siri bulls, respectively. Whereas in BIOXcell extender, it was 45.50 ± 5.99, 46.36 ± 5.05 and 49.17 ± 5.85 % in Jersey, Mithun and Siri bulls, respectively. Similarly, the plasma membrane integrity of thawed spermatozoa in Tris eggyolk extender was 17.50±2.32, 21.00±3.12 and 21.58±3.83% in Jersey, Mithun and Siri bulls, respectively. Whereas in BIOXcell extender, it was found to be 20.45±2.86, 20.86±2.281 and 22.33±3.24% in Jersey, Mithun and Siri bulls, respectively. No significant difference (p>0.05) was observed in post-thaw motility as well as in plasma membrane integrity between extenders and between breeds, and there was no significant effect (p>0.05) of breed x extender interaction. It can be concluded that the semen characteristics of Siri and Mithun bulls fall with the range established for other breeds of bulls. Assam Hill goat (AHG) is an important goat germplasm found in Assam and its adjoining areas of India. The study was designed with an objective to study the semen characteristics and freezability of AHG buck semen using Tris -Egg yolk-Citrate-Fructose diluent. The mean values of fresh semen characteristics in AHG bucks viz., ejaculate volume (ml), initial sperm motility (%), sperm concentration (x106/ml), live sperm (%), sperm abnormality (%), HOST-reacted sperm (%) and intact acrosome (%) recorded were 0.39 ± 0.01, 77.97 ± 0.73, 3201.00 ± 143.78, 83.02 ± 0.65, 7.66 ± 0.73, 66.95 ± 0.74 and 93.34 ± 0.51, respectively. Mean values for post-thaw semen characteristics i.e., sperm motility (%), live sperm (%), HOST-reacted sperm (%) and intact acrosome (%) were 55.39 ± 0.97, 71.01 ± 0.78, 54.77 ± 0.55 and 82.16 ± 0.43, respectively. It can be concluded that AHG bucks donate acceptable quality of semen which can be frozen successfully in Tris-Egg yolk-Citrate-Fructose diluents for using in Artificial Insemination.

The current study was planned to evaluate the supplementation of Polyvinylpyrrolidone in extender on cryopreservation of Nili-Ravi buffalo bull semen. The semen samples were collected from Nili-Ravi buffalo (Bubalus bubalis) bull kept at SPU Qadirabad, District Sahiwal, Pakistan. Qualifying semen ejaculates having motility >60%, volume >5-6ml and concentration >0.5 billion/ ml were diluted 50 × 106 motile sperm ml approximately at 37°C in Tris-citric acid extender supplemented with different concentrations of PVP (0.01, 0.05, 0.1mM). The extender without PVP was kept as control. Semen was stored at 4°C for a period of 2 h and kept at 4°C for 4h. Semen was filled in 0.5 ml French straws using suction pump at 4°C, plunged and stored in liquid nitrogen (-196°C). Semen straws were rewarmed at 37°C for 30 seconds and assessed for sperm motility, plasma membrane integrity (PMI), dead sperm percentage and the live sperm percentage. The data on the role of PVP on different parameters of semen quality were analyzed by using ANOVA and RCBD. Higher percentage (P< 0.05) of sperm motility (66.1±7.51 and 59.4±10.72) and PMI (72.9±5.39 and 75.7±6.5) was observed in extenders having 0.05 mM and 0.1mM PVP compared to extenders having 1.5mM PVP and control. The percentage acrosomal integrity was observed greater (P< 0.05) in extended semen containing 0.1mM (68.2±0.50) PVP compared to extenders having 0.01 and control.
