Fresh and Post-Thaw Seminal Attributes of Jafarabadi Buffalo Bulls

The study was conducted on four Jafarabadi breeding bulls, 5-6 years old to know the fresh and post-thawed seminal characteristics based on total of 192 semen ejaculates evaluated and cryopreserved over one year period. The mean values of fresh neat seminal characteristics of Jafarabadi bulls, viz., ejaculate volume (ml), colour/density (score), sperm concentration (million/ml), mass activity (score), initial motility (%), live sperm (%), abnormal sperm (%), HOS reactive sperm (%) and acrosomal integrity (%) were 5.19±0.18, 2.38±0.10, 1253.36±24.75, 3.73±0.05, 80.31±0.05, 86.20±0.64, 5.00±0.40, 85.75±0.43 and 93.56±0.56, respectively, whereas the mean post-thawed sperm characteristics, viz., progressive sperm motility, live sperm (%), abnormal sperm (%), HOS reactive sperm (%), acrosomal integrity (%) and first insemination conception rate (%) observed were 57.60±0.36, 66.34±0.53, 8.85±0.33, 56.97±0.46, 75.26±0.17 and 44.63±0.14, respectively. The semen quality of fresh and post-thawed samples observed was within normal limit for use in breeding program with satisfactory first insemination conception rate.

Background: The quality of the frozen thawed semen is one of the most powerful factor affecting the conception rate in a buffaloes. The season may have its effect on post thaw seminal attributes and fertility. Therefore, current study was aimed to investigate the effect of winter and summer seasons on post thaw seminal attributes and conception rate from semen of Jaffarabadi buffalo bulls.Methods: The study was conducted on four Jaffarabadi breeding bulls, 5-6 years old, to know the post-thaw semen characteristics based on total of 192 semen ejaculates evaluated and cryopreserved over one year period. Result: The mean percentages of post-thaw motility, live sperm, abnormal sperm, HOST reactive sperm, acrosomal integrity and first AI conception rate for semen cryopreserved during winter were 58.85±0.56, 67.06±0.77, 8.62±0.43, 58.52±0.67, 75.22±0.27 and 45.75±0.21, respectively, whereas, corresponding values for semen cryopreserved during summer were 56.35±0.43, 65.62±0.76, 9.08±0.51, 55.43±0.60, 74.29±0.22 and 43.50±0.11. The semen quality of post thawed samples were observed better during winter compared to summer season for artificial insemination with satisfactory first AI conception rate.

A study was conducted to assess the semen quality using Resazurin Reduction Test (RRT) in Jersey and Jersey cross bulls. A total 36 fresh ejaculates were collected for evaluation RRT, sperm motility, acrosomal integrity and sperm concentration. The mean value of RRT in bulls was 15.77±0.85 minutes.Statistical analysis revealed that RRT was highly significantly (p<0.01) and positively correlated with sperm concentration (r=0.769), sperm motility(r=0.858) and acrosomal integrity (r=0.515), hence it may be helpful in evaluating the quality of semen in bulls.

This study was undertaken during the favourable breeding season of the year 2016-17 on healthy mature Gir cattle and Surti buffalo bulls, three each, at Sperm Station of the College. The ejaculates (9/bull, total 54) collected in the morning using artificial vagina were evaluated for routine seminal attributes, including acrosomal and plasma membrane integrity. The mean values of ejaculate volume, sperm concentration, mass activity (0-5 score), individual sperm motility, live sperm, abnormal sperm, intact acrosome and HOST reactive sperms observed in fresh semen of Gir cattle and Surti buffalo bulls were 6.20±0.42 and 3.34±0.23 ml (P less than 0.01), 1169.44±61.71 and 846.30±54.82 million (P less than 0.01), 3.44±0.09 and 3.42±0.08, 76.53±0.53 and 80.76±0.39 % (P less than 0.05), 81.00±1.32 and 84.73±0.78 % (P less than 0.05), 6.00±0.37 and 5.81±0.40 %, 95.59±0.35 and 95.54±0.25 % as well as 80.30±1.90 and 84.58±0.88 % (P less than 0.05), respectively. The variation between cattle and buffalo semen was significant for most of these traits. The variations between bulls within breed were however not significant. Significant correlations were observed only between mass activity and initial motility (0.62), and live and abnormal sperm (-0.41) in Gir bulls, and for ejaculate volume with sperm concentration (-0.56) and abnormal sperm (0.45), and between live and HOS reactive sperms (0.48) in Surti bulls.

A study was carried out on semen ejaculates (40) of five healthy Surti buffalo bulls during favourable breeding season. The ejaculates with >70% IM were diluted @ 100 million sperms/ml in Tris-fructoseegg yolk-glycerol (TFYG) diluent supplemented with L-cysteine @ 1 mg/ml, and examined for quality parameters such as sperm progressive motility, viability, morphology, acrosomal and plasma membrane integrity. A part of the extended semen was preserved at 5°C and another was processed for ultra-low temperature (-196°C) preservation in LN2 using biofreezer after filled in French mini straws. The mean values of ejaculate volume, sperm concentration, mass activity (0-5 score), individual sperm motility, live sperm, abnormal sperm, intact acrosome and HOS reactive sperms observed in fresh semen were 3.04±0.11 ml, 963.05±50.97 million/ml, 2.97±0.06, 75.00±0.69 %, 88.22 ±0.48 %, 4.40±0.26 %, 93.67±0.31 % and 89.17±0.57 %, respectively. Just after dilution the percentages of progressively motile, live & abnormal sperms, intact acrosomes and HOS reactive sperms were 80.51±0.65, 85.95±0.65, 4.92±2.20, 91.00±2.30, 85.72±0.67, respectively. The corresponding values after 48 hrs of refrigeration (5°C) were 61.75±0.85, 67.85±0.85, 6.45±0.27, 89.05±0.32, 67.30±0.96%, the values at pre-freeze stage (after equilibration) were 72.62±0.69, 78.97±0.93, 5.45±0.25, 90.90±0.35, 78.12±0.79% and at post-thaw stage (37°C/30 sec) 46.50±0.72, 52.97±0.79, 7.10±0.26, 85.73±0.18, 51.62 ±0.82%, respectively. The mean motile spermatozoa observed after 1, 2 and 3 h of post-thaw incubation at 37°C in water bath were 32.75±0.82, 18.88 ±0.70 and 8.88±0.66% (P<0.01), respectively. The semen quality parameters, fresh and cryopreserved were acceptable for artificial breeding use. The seminal traits in initial, 48 hrs refrigerated, pre-freeze and post-thawed samples revealed significant (p<0.01) interrelationships (r = 0.44 to 0.84) between progressive motile sperm, live sperm and HOST reactive sperm directing more emphasis on these quality parameters for better semen evaluation.

Assam Hill goat (AHG) is an important goat germplasm found in Assam and its adjoining areas of India. The study was designed with an objective to study the semen characteristics and freezability of AHG buck semen using Tris -Egg yolk-Citrate-Fructose diluent. The mean values of fresh semen characteristics in AHG bucks viz., ejaculate volume (ml), initial sperm motility (%), sperm concentration (x106/ml), live sperm (%), sperm abnormality (%), HOST-reacted sperm (%) and intact acrosome (%) recorded were 0.39 ± 0.01, 77.97 ± 0.73, 3201.00 ± 143.78, 83.02 ± 0.65, 7.66 ± 0.73, 66.95 ± 0.74 and 93.34 ± 0.51, respectively. Mean values for post-thaw semen characteristics i.e., sperm motility (%), live sperm (%), HOST-reacted sperm (%) and intact acrosome (%) were 55.39 ± 0.97, 71.01 ± 0.78, 54.77 ± 0.55 and 82.16 ± 0.43, respectively. It can be concluded that AHG bucks donate acceptable quality of semen which can be frozen successfully in Tris-Egg yolk-Citrate-Fructose diluents for using in Artificial Insemination.

Objective: To determine the effect of age and frequency of collection on quality parameters of Jersey bull semen at National Livestock Breeding Center (NLBC), Pokhara. Materials and Methodology: Nine Jersey bulls were selected randomly from NLBC. To obtain the effect of age, six bulls of three different age interval (3-4, 5-7 and 8-9 years) were selected randomly. Remaining three bulls were used to determine the effect of frequency of collection on semen quality. Studies were conducted for three months (Nov/Dec to Dec/Jan, 2016). Semen quality was analyzed using CASA system and SMILE software. ANOVA and paired t- tests were applied to find out level of significance. All the statistical analyses were done using SPSS 17.0 Results: Sperm concentration, post- thaw motility, live sperm, and abnormal sperm were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. Significantly higher value for semen volume, sperm concentration and post- thaw motility was obtained on second collection (P<0.05). However, volume initial motility, pre-filling motility, live sperm and abnormal sperm percentage did not differ significantly among different age intervals and between the frequencies of collection. Conclusion: Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentages were higher in 5-7 years old bulls and in second collection of semen. Sperm concentration (0.551 ±0.004 to 1.084 ± 0.014x109), post- thaw motility (42.44±0.11 to 51.72±0.03 %), live sperm (45.62 ± 0.13 to 51.23±0.15 %), and abnormal sperm (11.57±0.12 to 19.59±0.24%) were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. On the other hand, volume (3.98±0.09 to 6.72±0.06 ml), initial motility (72.68±0.02 to 78.64±0.15 %) and pre-filling motility (71.34±0.17 to 77.62±0.02 %) did not differ significantly among different age intervals. Significantly, higher value for semen volume (3.98±0.12 to 6.02±0.14 ml), sperm concentration (0.572±0.119 to 1.095±0.015) and post- thaw motility (40.34±0.21 to 48.54±0.06% was obtained on second collection. However, initial motility (70.58±0.06 to 75.64±0.13 %), prefilling motility (68.11±0.12 to 71.62±0.02 %), live sperm (53.27±0.54 to 56.17±0.02 %) and abnormal sperm (12.31±0.12 to 14.3±0.47 %) did not differ significantly between the frequencies of collection. Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentage were higher in 5-7 years old bulls and in second collection of semen.

Semen quality of cocks is of utmost importance in reproduction. Breeding method, breed, strain and genetic constitution are the major components of the genetic factors capable of affecting the semen quality of cocks. This research seeks to determine the effect of genotype on the semen quality traits of 272 F crossbred cocks produced at day-old by main and 1 reciprocal crossbreeding of 69 Isa Brown and local frizzle feathered, naked neck and normal feathered chickens. The genotypes of the cocks were Isa Brow x frizzle feathered main cross (IBxF), Isa Brown x naked neck main cross (IBxNa), Isa Brown x normal feathered main cross (IBxN), frizzle feathered x Isa Brown reciprocal cross (FxIB), naked neck x Isa Brown reciprocal cross (NaxIB) and normal feathered x Isa Brown reciprocal cross (NxIB). At 36-40 weeks of age, semen was extracted from the cocks by abdominal  massage technique and used to evaluate semen volume (SV), sperm motility (SM), sperm concentration (SC) and live sperm (LS) at weekly intervals. Data obtained from the evaluation were subjected to analysisof variance and tested at 5% level of probability. Genotypes differed significantly (P <0.05) in SC at 37-40 weeks and in SM and LS at all ages. There was no significant difference (P>0.05) in SV. Regardless of genotype and age, the mean SV, SM, SC and LS ranged from 0.31±0.02 to 0.24±0.01ml, 71.84±1.33 to 58.75±2.16 %, 3.22±0.02 to2.92±0.02 x109/ml and 71.91±1.37 %, respectively. The NxIB and IBxNa genotypes produced largest numericalsemen volume at 37 and 39 weeks of age respectively. Whereas SC and LS were significantly highest for IBxF and IBxN respectively at week 37, SM was so for IBxN at all ages. Further, the LS of IBxN genotype was significantly highest at 36-38 and 40 weeks of age. The exotic Isa Brown and normal feathered main cross cocks exhibited the best semen quality traits among all genotypes studied and should be selected for improvement of the reproductive characteristics of cocks.

The study was carried out to observe the effects of preservation time on the quality of frozen semen of indigenous rams. Semen was collected using AV once a week from 4 rams. Tris based with 10% egg yolk and 7% glycerol extender was used to extend and freezing the semen. Fresh semen was evaluated for volume, density, mass motility and concentration, and mean values were observed as 0.8±0.2ml, 3.0±0.3, 3.2±0.7, 3.9±0.7×109/ml, respectively. Significant difference (p<0.05) was found for all the parameters among the rams. Mean values of motility, viability and normal morphology percentages were 83.3±4.3%, 88.2±4.4%, 84.2±3.5% in fresh semen while those of chilled semen at 40C were 74.7±2.3, 78.8±4.9 and 79.2±2.9%, respectively. For all the parameters, significant (p<0.05) difference was found among the rams. Frozen sperm motility was observed after thawing at 39-400C for 14-15 seconds. The mean motility, viability and normal morphology percentages after freezing for 24hrs, 7, 15 and 30 days of duration were 39.8±3.1, 41.1±4.3, 40.1±4.1 and 39.4±2.9%; 44.5±2.5, 45.3±2.8, 44.6±2.8 and 43.9±2.8%; 71.0±2.0, 71.7±1.5, 70.7±1.7 and 70.3±1.8%, respectively and values did not decrease significantly (p>0.05) with the increasing time of preservation. Non significantly decrease of the semen quality with advance of preservation time indicates the suitability of the protocol used for freezing of indigenous ram semen in Bangladesh.DOI: http://dx.doi.org/10.3329/bjas.v44i1.23113            Bang. A total of sixteen rabbits sourced from two locations in Southeastern Nigeria were used for this study. Semen was collected from all the rabbits twice weekly for eight weeks using an artificial vagina while observation on the effect of time of collection on semen quality were also made. The results obtained indicated that time of collection had no significant effect (P > 0.05) on all the seminal characteristics measured. However, source of the animals showed significant effect (P < 0.05) on semen volume, sperm concentration, total sperm per ejaculate and proportion of abnormal sperm. Average semen volume, sperm concentration and total sperm per ejaculate were: 0.69±0.007 ml, 104.0±1.44 x 106/m1 and 71.40±0.90x106 respectively for the rabbits sourced from Port Harcourt. The corresponding values for rabbits sourced from Owerri were: 0.66±0.008m1, 96.02 ±1.63x106iml and 63.49±1.50 x 106 respectively. Abnormal sperm proportions were 11.05±0.22% and 1216 ± 032% for the rabbits sourced from Port Harcourt and Owerri respectively. There were no significant (P> 0.05) differences observed between locations in live sperm concentration and sperm motility. Live sperm concentration and sperm motility were 87.75±1.09% and 71.62±0.74% for the Port Harcourt group, while the values obtained for the Owerri group were 88.06±034% and 71.19±0.89% respectively.
