Objective: To evaluate the antioxidant and antibacterial efficiency of alkaloids of root, leaf, and fruit of Cucumis trigonus. Methods: The antioxidant and antibacterial properties of alkaloid extracts were assessed by ferric thiocynate (FTC), 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, hydrogen peroxide radical scavenging assay, superoxide radical scavenging activity and ferric reducing power were analyzed separately for its inhibition percentage at different concentration (20µg, 30µg, 40µg and 50µg/ml) and antibacterial activity by agar disc diffusion and minimum inhibitory concentration (MIC) against Staphylococcus aureus (MTCC Code-9886), Pseudomonas aeruginosa (MTCC Code-6458). Results: The highest percentage of extraction yield was observed in fruit followed by leaf and root. The maximum content of alkaloid was found in fruit and root compared to leaf. The scavenging efficiencies of the extracts increased with the increasing concentrations. Appreciable levels of total antioxidant activity by FTC (2.112±0.011%), DPPH radical scavenging activity (1.912±0.001%), superoxide radical scavenging activity (0.955±0.021%) and hydrogen peroxide radical scavenging activity (0.914±0.087 %) were observed at 50µg/ml of fruit extract. However, ferric reducing power was more in the root (0.893±0.0870%) compared to fruit (0.791±0.023%) and leaf (0.520±0.00 %) at 50µg/ml. The alkaloid extracts of root and fruit exhibited more antibacterial activity against Staphylococcus aureus (16.51±0.05 and 19.68±0.03 mm) and Pseudomonas aeruginosa (12.2±0.03 and14.2±0.06) at 100µg/ml than leaf. The minimum inhibitory concentration (MIC) of the root, leaf, and fruit was in the range of 3.125µg/ml to 35µg/ml for the pathogenic bacteria. Conclusion: Data from the present results revealed that the alkaloid extracts of fruit and root of C. trigonus show good antioxidant and antibacterial potential than leaf. Hence, may be explored for the formation of new antibacterial with antioxidant drugs.

Glucuronomannan oligosaccharides (Gs) were derived from fucoidan, which was extracted from the brown alga Sargassum thunbergii. Sulfated glucuronomannan oligosaccharides (SGs) were obtained by the sulfation of Gs. NMR techniques were used to reveal that the order of sulfation was Man-C6 > Man-C4 > Man-C1R > GlcA-C3 > Man-C3 > GlcA-C2. Finally, the antioxidant activities (hydroxyl radical scavenging activity, superoxide radical scavenging activity, reducing power and DPPH radical scavenging activity) of Gs and SGs were determined. The findings showed that the higher the degree of polymerization, the better the activity, except for the hydroxyl radical scavenging activity. In addition, the higher the sulfate content, the lower the activities for the reducing power and DPPH radical scavenging activity. Opposite results were found for the superoxide radical scavenging activity. Finally, compared with fucoidan, most Gs and SGs had higher antioxidant activity, suggesting that they might be good candidates for antioxidants.

The present study aimed to evaluate the potential of standardized methanolic extracts from seven Ficus deltoidea varieties in inhibiting the formation of AGEs, protein oxidation, and their antioxidant effects. The antiglycation activity was analyzed based on the inhibition of AGEs, fructosamine, and thiol groups level followed by the inhibition of protein carbonyl formation. The antioxidant activity (DPPH radical scavenging activity and reducing power assay) and total phenolic contents were evaluated. After 28 days of induction, all varieties of Ficus deltoidea extracts significantly restrained the formation of fluorescence AGEs by 4.55–5.14 fold. The extracts also reduced the fructosamine levels by 47.0–86.5%, increased the thiol group levels by 64.3–83.7%, and inhibited the formation of protein carbonyl by 1.36–1.76 fold. DPPH radical scavenging activity showed an IC50 value of 66.81–288.04 μg/ml and reducing power activity depicted at 0.02–0.24 μg/ml. The extent of phenolic compounds present in the extracts ranged from 70.90 to 299.78 mg·GAE/g. Apart from that, correlation studies between the activities were observed. This study revealed that seven varieties of Ficus deltoidea have the potential to inhibit AGEs formation and possess antioxidant activity that might be attributed to the presence of phenolic compounds.

Two novel antioxidants, obtusilactone A (1) and (-)-sesamin (2) have been identified in Cinnamomum kotoense Kanehira. Both showed effective 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity compared with vitamin C, and reducing power compared with BHA. These results suggest that these constituents of C. kotoense act as natural antioxidants and play a potential role in cancer prevention.

The Ablmoschus manihot (L.) Medic, an edible hibiscus of the Malvaceae family, is abundant with total flavonoid. The content of total flavonoid from A. manihot flowers (TFA) was extracted by 70% ethanol (ethanol:water, 70:30). The scavenging effects of TFA on superoxide anions and hydrogen radicals were related to the total flavonoid concentrations with the IC50 values of 63.90 ±2.21 μg/mL and 266.88±28.32 μg/mL, respectively. However, the IC50 values of standard ascorbic acid were 436.52 ±14.36 μg/mL and 439.58±21.41 μg/mL, respectively. The DPPH radical scavenging activity increased with increasing of TFA concentrations and the highest inhibition was 94.63 ±3.01% at 50 μg/mL, as compared to ascorbic acid 34.94 ±1.50%. The extract also showed good reducing power. The data from this study suggest A. manihot flower extract has significant potential to use as health supplements and nutraceuticals.

Pulverized rhizome of Curcuma peliolata on hydrodistillation, afforded light yellow essential oil in the yield of 0.13% v/w. The chemical constituent of the essential oil was determined by GC-MS. Nine compounds representing 95.34 % of the total oil were identified. The major compounds of the oil were 2-methyl-5-pentanol, 1H-pyrrol-1-amine,2-(4-methoxyphenyl)-n,n,5-trimethyl, and curcumol. The distillated aqueous was extracted with dicloromethane (DCM), ethyl acetate (EtOAc), and n-butanol. The DPPH radical scavenging and reducing power of the extracts were investigated. Curcumin and ascorbic acid were used as standard references. The DPPH radical scavenging activity was decrease in order: curcumin ascorbic acid EtOAc extract > DCM extract > n-butanol extract > aqueous extract. The EtOAc extract showed significantly highest inhibition of the radical scavenging activity with IC50 0.092 ± 0.001 mg/mL (curcumin and ascorbic acid (IC50 0.012 and 0.013 mg/mL, respectively (P<0.05)). While, the DCM extract showed highest reducing power with IC50 = 0.271 mg/mL which lower than that of standard curcumin (IC50 = 0.013 mg/mL) and ascorbic acid (IC50 = 0.002 mg/mL).Introduction

Callus cultures from stems, leaves and roots of colocynth were initiated on MS media supplemented with various combinations of 2,4 dichlorophenoxyacetic acid (2,4-D) with kinetin (KIN) and benzyladenine (BA) with α-naphthaleneacetic acid (NAA). The highest percentage of callus formation frequency (98.9%) was obtained from stem explants grown on MS media supplemented with (1.0 mg/L) 2,4-D + (1.0 mg/L) KIN. The total phenolics and flavonoid content of the colocynth callus cultures were measured. The results showed that the MS medium supplemented with 6.0 mg/L 2,4-D + 2.0 mg/L KIN (MD3) gave the highest content of total phenolics (19.2 mg/100g d.w.) in leaf-derived calli. The highest content of flavonoids (47.3 mg/100g d.w.) was obtained in stem derived calli grown on the same medium (MD3). Antioxidant activities of extracts were determined using different assays, including DPPH radical scavenging activity, hydrogen peroxide (H2O2) scavenging activity and ferric reducing power. Leaf-derived calli cultured on MS medium + 2.0 mg/L 2,4-D + 1.0 mg/L KIN (MD1) showed the highest DPPH radical scavenging activity (85.3%). The highest percentage of H2O2 scavenging activity (61.4%) was detected in leaf explant-derived calli growing on MD1. The leaf-derived calli growing on (MD3) gave the highest ferric reducing power (22.3 μg/g d.w.), compared to the activities of stems, leaves and roots of in vitro grown seedlings (3.28, 12.9 and 2.85 μg/g d.w.), which were used as controls. On the basis of the current findings, we conclude that MS media supplemented with different combinations of 2,4-D and KIN yields higher phenolics, flavonoids contents and antioxidant activities than MS media supplemented with BA and NAA.

Objective: In the present study, antioxidant activity in the leaf of the pet-ether, chloroform, acetone and methanolic extracts from Litsea laevigata Gamble. Leaf was investigated by employing established in vitro studies. L. laevigata belongs to the Lauraceae family. Methods: The capability of the plant extract to act as hydrogen/electrons donor or scavenger of radicals were determined by in vitro antioxidant assays using 2,2-diphenyl-2-picrylhydrazyl free radical (DPPH.) scavenging, reducing power assay, superoxide radical (O2*-) scavenging activity, phosphomolybdenum assay, FRAP, ABT and metal chelating activity were performed to know the antioxidant potency of the plant extract of leaves of L. laevigata. Results: Results are evaluated higher in leaf extract of L. laevigata recorded total phenol, total flavonoid, and tannin. The present state of work was designed to evaluate the phytochemical, antioxidant in the plant leaf extracts of L. laevigata. The plant L. laevigata methanolic extract of leaf showed greater IC50 antioxidant activity of DPPH assay (5.264 µg/ml) and compare to other extract, higher phosphomolybdenum reduction (164.36 mg/g), better Reducing power activity leaf in methanol (0.711%), higher ferric reducing power (4060.66MmolFe(II)E/mg), and higher in superoxide radical scavenging activity in (78.12 mg/ml). However, the better metal chelating ability was shown by the water extracts of the leaf (5.145 EDTAE/100g) compared to other solvent extracts. Conclusion: The result indicates the total phenol and antioxidant activity potential of L. laevigata.

Organic Rices, such as Jasmine variety white organic rice, Saodah variety red organic rice, and Java variety black organic rice, are many cultivated by farmer in Indonesia, especially Sleman area, DI Yogyakarta. The potency of three varieties of organic rice as antioxidant source hasn’t been studied. Therefore this research was done to know the effect of pigment color difference in three varieties of organic rice on antioxidant activity, especially 1,1-diphenyl-2- pycrylhydrazyl (DPPH) radical scavenging activity and ion reducing power. The results showed that Saodah variety red organic rice had the highest antioxidant potency because it had the highest total phenol and total flavonoid, 37.93 mg GAE/g sample dry base and 0.85 mg CE/g sample dry base, respectively. However Saodah variety red organic rice had total anthocyanin lower than that of Java variety black organic rice, 0.0025 mg/g sample dry base and 0.024 mg/g sample dry base, respectively. Anthocyanin compounds identified in three varieties of organic rice were cyanidine-3-glucoside at 32 minute time retention and peonidine -3-glucoside at 37 minute time retention. 1,1-diphenyl-2-pycrylhydrazyl (DPPH) radical scavenging activity and ion reducing power of Saodah variety red organic rice were the highest, 0.90 mg equivalent vitamin E/g sample dry base and 278.28 mg equivalent vitamin E/g sample dry base, respectively.Keywords: Pigment, organic rice, antioxidant activity ABSTRAKBeras organik, meliputi beras putih varietas Jasmine, merah varietas Saodah, dan hitam varietas Jawa, banyak dibudidayakan oleh masyarakat di Indonesia, terutama daerah Sleman, D.I. Oleh karena itu penelitian ini dilakukan untuk mengetahui pengaruh perbedaan pigmen pada ketiga jenis beras organik tersebut terhadap aktivitas antioksidan, khususnya kemampuan menangkap radikal bebas 2,2-difenil-1-pikrilhidrasil (DPPH) dan mereduksi ion besi. Hasil menunjukkan bahwa beras organik merah varietas Saodah paling berpotensi sebagai sumber antioksidan. Hal ini ditunjukkan dengan total fenol dan total flavonoid beras merah tertinggi dibandingkan kedua beras yang lain, masing-masing sebesar 37,93 mg GAE/g sampel db dan 0,85 mg CE/g sampel db. Meskipun beras organik merah mempunyai total antosianin (0,0025 mg/g sampel db) lebih rendah dari beras organik hitam (0,024 mg/g sampel db). Jenis senyawa antosianin yang terdeteksi pada ketiga jenis organik ini adalah sianidin-3-glukosida yang terdeteksi pada waktu retensi 32 menit dan peonidin-3-glukosida dengan waktu retensi 37 menit. Kemampuan menangkap radikal bebas DPPH dan kemampuan mereduksi ion besi beras organik merah tertinggi, masing-masing sebesar 0,90 mg ekuivalen vit E/g sampel db dan278,28 mg ekuivalen vit E/g sampel db.Kata kunci: Pigmen, beras organik, aktivitas antioksidan

In this study, antioxidant activities of H. fursei were investigated. Different model were used for assay. IC50 for DPPH radical-scavenging activity was 7.5±1.2 μg ml-1. The IC50 value for BHA was 92.9±4.5 µg ml-1. The extract showed very good reducing power that was comparable with vitamin C (p> 0.05) but showed weak nitric oxide scavenging activity and Fe2+ chelating ability. Extract showed good activity in scavenging of H2O2. Inhibition was 99.1% at 800 μg ml-1. The IC50 values for extract and BHA were 333.1±14.7 and 52.0±4.5 μg ml-1, respectively. Total phenol compounds, by the Folin Ciocalteu method, was 274±9.6 mg gallic acid equivalent/g of extract and the total flavonoid content, by AlCl3 method was 23.9 ± 1.4 mg quercetin equivalent/g of extract. Antioxidant activity may be attributed to the presence of phenols and flavonoids in the extract.

Natural antioxidants are found in algae, are important biologically active compounds that involve in protecting the cells from oxidation thereby used in defense mechanism on ageing processes and various diseases. In this study, R-Phycoerythrin (R-PE) was extracted from Spyridia filamentosa and was assessed for their antioxidant properties. The antioxidising property of decontaminated R-PE has been evaluated by measuring the free radical scavenging activity by DPPH radical scavenging assay, ABTS radical scavenging activity, hydroxyl radical scavenging activity, superoxide radical (O2) scavenging assay, nitric oxide scavenging activity and phosphomolybdenum reduction assay. The purified R-PE extract was subjected to structural analysis by UV-Visible spectroscopy, fluorescence emission spectroscopy and infrared spectroscopy. The UV-Visible spectrum showed a peak at 496 nm, 543 nm and 564 nm and fluorescence emission spectrum of R-PE was measured 575 nm which confirmed the presence of R-Phycoerythrin. The IR spectral data showed the presence of aromatic constituents containing amide group in the protein. DPPH radical scavenging assay showed the highest antioxidant potential with an IC50 value of 128.25 μg/ml. The radical scavenging activity of Nitric oxide showed an IC50 value of 87.85 μg/ml, Phosphomolybdenum reduction assay showed an RC50 value of 61.45 μg/ml. The hydroxyl radical scavenging activity showed IC50 value of 34.56 μg/ml and Superoxide radical (O2) scavenging assay showed IC50 value of 18.58 μg/ml. The ABTS radical scavenging activity showed the lowest antioxidant potential of 3.13 μg/ml. The free radical scavenging property of R-Phycoerythrin (R-PE) concentrate of Spyridia filamentosa demonstrate that it has high antioxidant potency and it could be considered as utilitarian nourishment with essential pharmaceutical and biological importance.
