Amaryllidaceae plants are rich in alkaloids with biological properties. Pancratium trianthum is an Amaryllidaceae species widely used in African folk medicine to treat several diseases such as central nervous system disorders, tumors, and microbial infections, and it is used to heal wounds. The current investigation explored the biological properties of alkaloid extracts from bulbs of P. trianthum collected in the Senegalese flora. Alkaloid extracts were analyzed and identified by chromatography and mass spectrometry. Alkaloid extracts from P. trianthum displayed pleiotropic biological properties. Cytotoxic activity of the extracts was determined on hepatocarcinoma Huh7 cells and on acute monocytic leukemia THP-1 cells, while agar diffusion and microdilution assays were used to evaluate antibacterial activity. Antiviral activity was measured by infection of extract-treated cells with dengue virus (DENVGFP) and human immunodeficiency virus-1 (HIV-1GFP) reporter vectors. Cytotoxicity and viral inhibition were the most striking of P. trianthum’s extract activities. Importantly, non-cytotoxic concentrations were highly effective in completely preventing DENVGFP replication and in reducing pseudotyped HIV-1GFP infection levels. Our results show that P. trianthum is a rich source of molecules for the potential discovery of new treatments against various diseases. Herein, we provide scientific evidence to rationalize the traditional uses of P. trianthum for wound treatment as an anti-dermatosis and antiseptic agent.

Polianthes tuberosa L. (Amaryllidaceae) is an ornamental and medicinal plant. Its flowers and bulbs are used traditionally as a diuretic, emetic, against rashes and gonorrhea. The aim of this work was, to evaluate the antioxidant and antimicrobial activities of bulbs and bulbils alkaloid extracts of P. tuberosa. Antiradical effect was assessed against DPPH radical. However, antimicrobial activity was measured through the disc diffusion method against Escherichae coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212, Staphylococcus aureus resistant to Methicillin (MRSA) and Candida albicans ATCC 90028. The scavenging effect against DPPH showed that the bulbs and bulbils alkaloids extracts exhibited an antiradical effect with IC50 = 0.231±0.017 mg/mL and 0.233±0.093 mg/mL respectively, less than the effect of vitamin C with IC50 = 0.0194±0.0002 mg/mL. Antimicrobial activity results reveal that both alkaloid bulbs extracts at 50 mg/mL did not have any inhibitory effect against the studied strains using the disc diffusion method. According to this work, bulbs and bulbils alkaloid extracts show a moderate antioxidant effect; that could be recommended as a natural antioxidant. Although tuberose bulbs were used traditionally as a soap substitute; bulbs alkaloid extract has no antimicrobial effect. Background: Hyoscyamus niger L. (Solanaceae) generally known as henbane, is commonly distributed in Europe and Asia. In Turkey, henbane seeds have been used in folk medicine to remove worms from the eyes. The present study aimed to investigate the insecticidal activity of H. niger seeds. Methods: n-hexane, ethyl acetate, methanol and alkaloid extracts were prepared from the seeds of the plant and their insecticidal activities on Lucilia sericata larvae were evaluated. EC50 and EC90 values of the alkaloid extract were calculated and morphological abnormalities were investigated. Results: Alkaloid extract prepared from the seeds of this plant displayed significant insecticidal activity. EC50 values of H. niger seeds alkaloid extract were found to be 8.04, 8.49, 7.96 μg/mL against first, second and third instar, respectively. It was determined that malformations of larvae included damaged larvae with small size, contraction and weak cuticle. Furthermore, HPLC analysis was performed on alkaloid extract of H. niger seeds and main components of the extract were determined. It was determined that alkaloid extract mainly contain hyoscyamine and scopolamine. Conclusions: These results confirm the folkloric usage of the plant and suggest that the alkaloid content of the plant could be responsible for the insecticidal activity.

AbstractTuberculosis is the main cause of death from a single infectious agent. Globally, according to the World Health Organization, in 2018, there were an estimated 1.2 million tuberculosis deaths. Moreover, there is a continuous appearance of drug-resistant strains. Thus, development of new antituberculosis medicines should receive high priority. Plant-derived natural products are promising candidates for this purpose. We therefore screened alkaloid extracts obtained from the root and stem barks of the Mexican Apocynaceae species Tabernaemontana alba and Tabernaemontana arborea, as well as the pure alkaloids ibogaine, voacangine, and voacamine, tested for activity against Mycobacterium tuberculosis H37Rv and cytotoxicity to mammalian Vero cells using the resazurin microtiter and the MTT assays, respectively. The extracts were analyzed by GC-MS and HPLC-UV. T. arborea root bark alkaloid extract showed the highest activity against M. tuberculosis (MIC100 = 7.8 µg/mL) of the four extracts tested. HPLC suggested that voacangine and voacamine were the major components. The latter was isolated by column chromatography, and its chemical structure was elucidated by 1H and 13C NMR, and MS. Unambiguous assignation was performed by HSQC, HMBC, and NOESY experiments. Voacamine is a dimeric bis-indole-type alkaloid and is 15 times more potent than the monomeric ibogan-type alkaloids ibogaine and voacangine (MIC100 = 15.6, 250.0, and 250.0 µg/mL, respectively). However, all of these compounds showed cytotoxicity to Vero cells, with a poor selectivity index of 1.00, 0.16, and 1.42, respectively. This is the first report of voacamine activity against M. tuberculosis.
