AbstractIgE sensitisation in tropical areas is under-reported. A 2 year retrospective cohort study of allergy data specific to aero and food allergens in Brunei demonstrated that specific IgE levels to house dust mite (Dermatophagoides pteronyssinus, D.farinae, Blomia tropicalis) were highest in this population and correlated with atopic dermatitis (p < 0.001). Shrimp and peanut were the most common food allergens. A dominance of house/storage mite sensitization is seen in Brunei which is consistent with other tropical countries.

Introduction: Allergic rhinitis (AR) is an inflammatory disease of the nasal mucosa that can be modeled using Controlled Allergen Exposure Facilities (CACF). Recently, we clinically validated the house dust mite (HDM) Environmental Exposure Unit (EEU) facility. In the current study, we aimed to assess biological responses in the blood following HDM exposure in the HDM-EEU.Methods: Fifty-five participants passed a screening visit, where they provided consent and completed a skin prick test (SPT), then attended a modest or higher HDM exposure session. Baseline and post-exposure blood samples were collected. Complete blood counts with differentials were measured, and isolated serum was used to determine Dermatophagoides farinae- and Dermatophagoides pteronyssinus-specific IgE (sIgE) and cytokine concentrations (IL-4, IL-5, IL-6, IL-10, IL-13, TNF-α).Results: HDM-allergic participants had significantly greater SPT wheal sizes than healthy controls. sIgE concentrations were significantly greater in allergic participants, with a strong correlation between Dermatophagoides farinae and Dermatophagoides pteronyssinus. Serum eosinophil counts were significantly decreased post-exposure for allergic participants. White blood cell, neutrophil, and lymphocyte counts were significantly increased for both allergic and non-allergic participants post-exposure. Serum IL-13 concentrations were significantly reduced post-exposure in allergics while TNF-α was significantly reduced in non-allergics.Conclusion: The HDM-EEU is a useful model for investigating biologic mechanisms of HDM-induced AR. Allergic participants produced measurable biological changes compared to healthy controls following allergen exposure, specifically with serum expression of eosinophils and related markers, namely IL-5, which promotes the proliferation and differentiation of eosinophils, and IL-13, a cytokine released by eosinophils. The exact mechanisms at play require further investigation.

House dust mite antigens have been used for decades to diagnose allergic diseases in humans and animals. The objective of this study was to identify allergens in commercial Dermatophagoides farinae and Blomia tropicalis extracts by immunoblotting using sera from allergic dogs and anti-dog IgE conjugate. The analysis of antigens present in the D. farinae extract (FDA Allergenic) using sera from 10 dogs allergic to D. farinae showed that eight sera recognized a band of approximately 102 kDa, eight recognized two bands of 52 to 76 kDa, five recognized one band of approximately 76 kDa, four recognized one band of 31 to 38 kDa, and two recognized one band of 12 to 17 kDa. Immunoblot assays of the B. tropicalis extract (FDA Allergenic) using sera from 10 animals allergic to B. tropicalis showed that five sera recognized two bands of 52 to 76 kDa. These results demonstrate the importance of the two house dust mite species for the pathogenesis of canine atopic dermatitis in Brazil. In addition, the results indicate which allergens should be present in allergenic extracts used for diagnosis and allergen-specific immunotherapy.

Shrimp tropomyosin has a similar structure to house dust mite (HDM) tropomyosin. In this research, 232 adult patients with symptoms of persistent allergic rhinitis were randomly selected. In the group, 59% were sensitized to Dermatophagoides pteronyssinus and 57.8% to Dermatophagoides farinae. In total, 128 (55.2%) patients were sensitized to both HDM species and 143 (61.6%) to at least one. Slightly over a quarter (25.4%) of patients were sensitized to shrimp. Of the 35 shrimp-sensitized patients, the sensitization to Der p 10 and Pen a 1 was found in 11 cases (31.4%). There was a strong correlation between IgE Pen a 1 and IgE Der p 10 concentrations. The results indicate that there are other allergens responsible for a high incidence of shrimp sensitization in HDM-sensitized patients. A high convergence of Der p 10 and Pen a 1 levels may indicate that the determination of just one of the above is reasonable.

Abstract. House dust mites are the most found allergens in homes scattered throughout the world that associated with allergic manifestations in the respiratory tract and skin, such as bronchial asthma, allergic rhinitis and atopic dermatitis. The most common house dust mite is from the family Pyroglyphidae, four of which have a close relationship with the incidence of allergy: Dermatophagoides pteronyssinus, Dermatophagoides farinae, Dermatophagoides microceras, and Euroglyphus maynei. This study aims to determine the type and density of house dust mites in several habitats in the homes of people with allergic diseases. This study is a descriptive study using cross-sectional method. Samples were dust in allergic patient’s home in Malalayang I, taken from three habitats, namely bed (mattress and bed linen), bedrooms floor and sofa, as many as 96 samples. Of the 96 samples, there are 60 samples positive of dust mites and 36 negative samples. This study found 157 mites (124 adults and 33 larvae mites). Of the 124 adult mites were identified, there are 122 from order Astigmata with the dominant species is Dermatophagoides pteronyssinus, and two others of the order Prostigmata. In each habitat that is in the bed (mattress and bed linen), bedrooms floor and sofa, Pyroglyphidae is the most prevalent.Keywords: house dust mite, allergiesAbstrak. Tungau debu rumah (TDR) adalah alergen dalam rumah terbanyak yang tersebar di seluruh dunia yang dikaitkan dengan manifestasi alergi pada saluran pernapasan dan kulit, seperti asma bronkial, rinitis alergi dan dermatitis atopik. TDR yang paling sering ditemukan adalah dari famili Pyroglyphidae, empat di antaranya memiliki hubungan yang erat dengan kejadian alergi: Dermatophagoides pteronyssinus, Dermatophagoides farinae, Dermatophagoides microceras, dan Euroglyphus maynei. Penelitian ini bertujuan untuk mengetahui jenis dan kepadatan tungau debu rumah pada beberapa habitat di rumah penderita penyakit alergi. Penelitian ini adalah penelitian deskriptif dengan menggunakan metode cross-sectional. Sampel penelitian adalah debu di rumah penderita alergi di Kelurahan Malalayang I yang diambil dari 3 habitat, yaitu tempat tidur (kasur dan sprei), lantai kamar tidur, dan sofa sebanyak 96 sampel. Dari 96 sampel, 60 sampel positif tungau debu dan 36 sampel negatif. Pada penelitian ini ditemukan 157 tungau (124 tungau dewasa dan 33 larva). Dari 124 tungau dewasa yang terindentifikasi, dari ordo Astigmata sebanyak 122 dengan spesies yang dominan adalah Dermatophagoides pteronyssinus, dan dua lainnya dari ordo Prostigmata. Pada masing-masing habitat yaitu di tempat tidur (kasur dan sprei), lantai kamar tidur dan sofa, famili Pyroglyphidae paling banyak ditemukan.Kata kunci: tungau debu rumah, alergi
