AbstractEnzootic pneumonia caused by hyopneumoniae (MHYO) remains a serious concern to the swine industry in many countries including Nigeria. MHYO strains isolated from pigs from different countries and geographical locations are known to vary in pathogenicity. There is a paucity of information on the pathogenicity of the MHYO strain affecting pigs in Nigeria. This study investigated the pathogenicity of the MHYO strain in naturally infected pigs using immunohisto-chemistry and electron microscopy. Two hundred and sixty four lungs of slaughtered pigs were randomly collected from abattoirs at Abeokuta, Ibadan and Lagos, in Southwest Nigeria. A sub-sample of 104 pneumonic and 20 apparently normal lungs was selected, processed for routine histopathological examination and immunohistochemistry, while 3 lung tissues samples were selected for ultrastructural studies. The most significant microscopic changes observed were suppurative broncho-interstitial pneumonia associated with varying degrees of lymphoid hyperplasia of the bronchus-associated lymphoid tissue (BALT) and thickened alveolar septa due to cellular infiltration consisting predominantly of neutrophils and a few mononuclear cells. Immunohistochemically, MHYO antigen was detected in 86/104 (82.69 %) of MHYO-infected lung tissues and typically exhibited a granular brown reaction on the bronchial and bronchiolar epithelial lining, mononuclear cells in the BALT and luminal cellular exudates within the airways. Transmission electron microscopy revealed numerous Mycoplasma organisms in the lumina of the airways, in between degenerated cilia, while a few Mycoplasmas were located within the alveoli. It was concluded that the MHYO strain detected in this study was pathogenic to pigs and capable of inducing pneumonia, and therefore implicated in the pathogenesis.

Pneumonia is a major economic threat to swine industry worldwide, however, there is still dearth of information on the pathology and associated pathogens in Nigeria, and these were therefore investigated. Lungs from 408 slaughtered pigs were randomly collected from abattoirs in Abeokuta, Ibadan and Lagos. The lung samples were cultured for bacterial pathogens using standard techniques, while formalin-fixed tissues were processed for histopathological examination. Grossly, the most consistent lesion was bronchopneumonia (35.3%). The main histopathological findings were lymphoid hyperplasia of bronchus-associated lymphoid tissue (BALT) (88.2%), suppurative bronchopneumonia (63.6%), suppurative bronchitis and bronchiolitis with concurrent epithelial hyperplasia (57.1%), as well as thickened alveolar septa due to cellular infiltration consisting predominantly of neutrophils (54.1%). Ten different species of bacteria were isolated from the lung samples in which two or more pathogens were isolated from each sample (82.7%). Pasteurella multocida was the most frequently isolated bacterium (54.8%). Among the bacteria isolated, there were significant (P < 0.05) differences in the frequencies of isolation of β-haemolytic Streptococci, P. multocida, Haemophilus species and Escherichia coli between the pneumonic lungs and apparently normal lungs. The results of bacterial culture, gross and histopathological changes recorded in this study are consistent with bacterial pneumonia possibly caused by most of the bacteria identified in the present study.

The importance of examining stained 1 to 2μ plastic sections by light microscopy has long been recognized, both for increased definition of many histologic features and for selection of specimen samples to be used in ultrastructural studies. Selection of specimens with specific orien ation relative to anatomical structures becomes of critical importance in ultrastructural investigations of organs such as the lung. The uantity of blocks necessary to locate special areas of interest by random sampling is large, however, and the method is lacking in precision. Several methods have been described for selection of specific areas for electron microscopy using light microscopic evaluation of paraffin, epoxy-infiltrated, or epoxy-embedded large blocks from which thick sections were cut. Selected areas from these thick sections were subsequently removed and re-embedded or attached to blank precasted blocks and resectioned for transmission electron microscopy (TEM).

In secretory cells a dense and complex network of actin filaments is seen in the subplasmalemmal space attached to the cell membrane. During exocytosis this network is undergoing a rearrangement facilitating access of granules to plasma membrane in order that fusion of the membranes can take place. A filamentous network related to secretory granules has been reported, but its structural organization and composition have not been examined, although this network may be important for exocytosis.Samples of peritoneal mast cells were frozen at -70°C and thawed at 4°C in order to rupture the cells in such a gentle way that the granule membrane is still intact. Unruptured and ruptured cells were fixed in 2% paraformaldehyde and 0.075% glutaraldehyde, dehydrated in ethanol. For TEM (transmission electron microscopy) cells were embedded in Lowicryl K4M at -35°C and for SEM (scanning electron microscopy) they were placed on copper blocks, critical point dried and coated. For immunoelectron microscopy ultrathin sections were incubated with monoclonal anti-actin and colloidal gold labelled IgM. Ruptured cells were also placed on cover glasses, prefixed, and incubated with anti-actin and colloidal gold labelled IgM.

AbstractMetal combinations have been attracting the attention of scientists for some time. They usually exhibit new characteristics that are different from the ones possessed by their components. In this work, Au/ZnO/Ag nanoparticles were synthesized biologically using Glechoma hederacea L. extract. The synthesized Au/ZnO/Ag nanoparticles were characterized by UV-Vis, Scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), Transmission electron microscopy (TEM), and Atomic Force Microscopy (AFM). The microscopic methods confirmed the presence of spherical nanoparticles of 50–70 nm. The influence of biologically synthesized Au/ZnO/Ag nanoparticles on the vitality of human cells was evaluated in vitro with the use of established human Acute T Cell Leukemia cell line, Jurkat (ATCC® TIB-152™), as well as mononuclear cells isolated from peripheral blood (PBMC) of voluntary donors. Cell survival and the half-maximal inhibitory concentration index (IC50) were analyzed by the MTT test. The studies showed that the total loss of cell viability occurred at the Au/ZnO/Ag nanoparticle concentration range of 10 µmol–50 µmol. The use of Au/ZnO/Ag nanoparticles at the concentration of 100 µmol eliminated almost all living cells from the culture in 24h. The above observation confirms the result obtained during the MTT test.

We investigated the influence of transpulmonary (Ptp) and vascular pressures on the size of the pores of Kohn in primary alveolar septa. Dogs lungs, perfused and ventilated in situ, were rapidly frozen with Freon 22 in zone II or III conditions following deflation to Ptp of 5, 15, or 25 cmH2O. Frozen samples were freeze-substituted for transmission electron microscopy. Five fields containing at least one pore each were selected randomly from each section of tissue, and the minimum diameter visible in the cut section was measured. For both zone II and III conditions, as Ptp increased, mean pore size increased. The mean pore size under zone III conditions was 1.2015, 1.788, and 2.249 micrometer for Ptp of 5, 15, and 25 cmH2O, respectively. For zone 2 conditions, the corresponding values were 1.1438, 1,8757, and 2.08 micrometer. For both zones II and III, increasing capillary hydrostatic pressure had no significant effect on pore size. The results support the notion that alveolar pores can increase collateral ventilation by dynamically stretching as Ptp increases. Capillary pressure does not influence pore size probably because of collagen fibers, which surround the pore lumen. Presumably, these fibers resist encroachment of capillaries on the pore lumen as vascular pressures increase.

AbstractFour species of Cystoisospora, C. canis, C. ohioensis, C. neorivolta and C. burrowsi are described from feces of dogs. Of these, the oocysts of C. canis are the largest and easily distinguished from the remaining three species. Oocysts of C. ohioensis, C. neorivolta and C. burrowsi are difficult to distinguish because of overlap in their sizes. However, based on endogenous developmental stages, C. ohioensis is distinct from C. neorivolta and C. burrowsi because its endogenous stages are confined to surface epithelium of intestine whereas endogenous stages of C. neorivolta and C. burrowsi are predominantly in the lamina propria. There are uncertainties regarding the endogenous stages of C. neorivolta and C. burrowsi and there is no way now to determine whether C. burrowsi and C. neorivolta are different parasites; therefore, these are referred as C. ohioensis-like organisms. Additionally, mode of division of asexual stages of coccidia of dogs is largely unknown and ultrastructural studies are lacking. In the present study, development of asexual and sexual stages of a C. ohioensis-like organism in a naturally infected dog is described by light microscopy and by transmission electron microscopy. Merozoites divided by endodyogeny/merogony. Meronts were crescent/merozoite-shaped and contained a maximum of eight nuclei. A distinctive feature of merozoites was the presence of many PAS-positive amylopectin granules that were absent or rare in immature microgamonts making it possible to distinguish them.

The aim of given investigation is to study the effect of cooling upon rat hepatocyte structure using transmission electron microscopic and computer morphometric methods. Ultrastructural and morphometrical characteristics of hepatocytes under liver cooling for various levels under in vivo and in vitro conditions were investigated. Vistar rats of 180-250 g were used in the experiment. Liver cooling (in vivo) was performed by means of original cryoapplicator with different probe temperature (1,2). Liver tissue for transmission electron microscopy was fixed in glutaraldehyde fixator on cocadylate buffer and OsO4. Dehydration was completed on acetone (3). Tissue embedding was done into the mixture of Epon/Araldite epoxy rasin. Ultrathin slices were contrasted by the method of Reinolds. Cell viewing and imaging were accomplished by electron microscope at accelerating power of 75kV.Morphometrical and stereometrical analysis was performed using the “Morpho-Tools” original computer system (c) 1994-1996 A.S. Kaprelyants, A.A. Kaprelyants, A.N. Reylan .

AbstractAlthough the rat model of mammary tumors chemically induced byN-methyl-N-nitrosourea (MNU) has been frequently used by several research teams, there is a lack of ultrastructural studies in this field. The main aim of this work was to perform an ultrastructural characterization of MNU-induced mammary tumors in female rats. Some alterations previously reported in human mammary tumors, such as nucleus size and shape, accumulation of heterochromatin in the perinuclear region, and interdigitating cytoplasmic processes between cancer cells were also observed in MNU-induced mammary tumors. Although a low number of samples were analyzed by transmission electron microscopy in the present study, we consider that it may contribute to a better understanding of MNU-induced mammary carcinogenesis in a rat model. The ultrastructural characteristics of the two most frequently diagnosed mammary carcinomas described in the present work can be useful to differentiate them from other histological patterns. In addition, the loss of cytoplasm in neoplastic cells and formation of vacuoles were described.
