Pneumonia is a major economic threat to swine industry worldwide, however, there is still dearth of information on the pathology and associated pathogens in Nigeria, and these were therefore investigated. Lungs from 408 slaughtered pigs were randomly collected from abattoirs in Abeokuta, Ibadan and Lagos. The lung samples were cultured for bacterial pathogens using standard techniques, while formalin-fixed tissues were processed for histopathological examination. Grossly, the most consistent lesion was bronchopneumonia (35.3%). The main histopathological findings were lymphoid hyperplasia of bronchus-associated lymphoid tissue (BALT) (88.2%), suppurative bronchopneumonia (63.6%), suppurative bronchitis and bronchiolitis with concurrent epithelial hyperplasia (57.1%), as well as thickened alveolar septa due to cellular infiltration consisting predominantly of neutrophils (54.1%). Ten different species of bacteria were isolated from the lung samples in which two or more pathogens were isolated from each sample (82.7%). Pasteurella multocida was the most frequently isolated bacterium (54.8%). Among the bacteria isolated, there were significant (P < 0.05) differences in the frequencies of isolation of β-haemolytic Streptococci, P. multocida, Haemophilus species and Escherichia coli between the pneumonic lungs and apparently normal lungs. The results of bacterial culture, gross and histopathological changes recorded in this study are consistent with bacterial pneumonia possibly caused by most of the bacteria identified in the present study.

AbstractEnzootic pneumonia caused by hyopneumoniae (MHYO) remains a serious concern to the swine industry in many countries including Nigeria. MHYO strains isolated from pigs from different countries and geographical locations are known to vary in pathogenicity. There is a paucity of information on the pathogenicity of the MHYO strain affecting pigs in Nigeria. This study investigated the pathogenicity of the MHYO strain in naturally infected pigs using immunohisto-chemistry and electron microscopy. Two hundred and sixty four lungs of slaughtered pigs were randomly collected from abattoirs at Abeokuta, Ibadan and Lagos, in Southwest Nigeria. A sub-sample of 104 pneumonic and 20 apparently normal lungs was selected, processed for routine histopathological examination and immunohistochemistry, while 3 lung tissues samples were selected for ultrastructural studies. The most significant microscopic changes observed were suppurative broncho-interstitial pneumonia associated with varying degrees of lymphoid hyperplasia of the bronchus-associated lymphoid tissue (BALT) and thickened alveolar septa due to cellular infiltration consisting predominantly of neutrophils and a few mononuclear cells. Immunohistochemically, MHYO antigen was detected in 86/104 (82.69 %) of MHYO-infected lung tissues and typically exhibited a granular brown reaction on the bronchial and bronchiolar epithelial lining, mononuclear cells in the BALT and luminal cellular exudates within the airways. Transmission electron microscopy revealed numerous Mycoplasma organisms in the lumina of the airways, in between degenerated cilia, while a few Mycoplasmas were located within the alveoli. It was concluded that the MHYO strain detected in this study was pathogenic to pigs and capable of inducing pneumonia, and therefore implicated in the pathogenesis.

Background: There is scanty literature available about concurrent infection of Pestes des Petits (PPR) and emerging opportunistic bacterial pathogens in field conditions. Thus, study was conducted to investigate the above based on necropsy. Methods: Systematic necropsy examination was conducted on 50 small ruminant carcasses. Detailed study included virological, bacteriological, gross and microscopic investigation on tissues collected using standard techniques. PPR virus (PPRV) antigen was detected in various samples using sandwich Enzyme Linked Immunosorbent Assay (s-ELISA). Bacterial pathogens were isolated on blood agar and identified by VITEK® system.Result: Eleven goats and nine sheep had concurrent infection. Highest positivity for PPRV antigen was observed in nasal and tracheal swab, lungs and lungs associated lymph nodes. Pasteurella multocida was the major bacteria found to cause fibrinious hemorrhagic broncho-alveolar pneumonia followed by Klebsiella pneumoniae spp. pneumoniae producing suppurative bronchoalveolar pneumonia in the presence of PPR infection. Sphingomonas paucimobilis and Acinetobacter baumannii, human pathogen were also isolated and found to produce severe hemorrhagic interstitial pneumonia and serofibrinous bronchopneumonia respectively in presence of PPR which is novel finding. Our study is the first report of necropsy based investigation on concurrent infection in sheep and goats.

AbstractBackground: Known also as Osler’s triad, Austrian syndrome is a complex pathology which consists of pneumonia, meningitis and endocarditis, all caused by the haematogenous dissemination of Streptococcus pneumoniae. The multivalvular lesions are responsible for a severe and potential lethal outcome.Case report: The case of a 51-year-old female patient, with a past medical history of splenectomy, is presented. She developed bronchopneumonia, acute meningitis and infective endocarditis as a result of Streptococcus pneumoniae infection and subsequently developed multiple organ dysfunction syndromes which led to a fatal outcome. Bacteriological tests did not reveal the etiological agent. The histopathological examination showed a severe multivalvular endocarditis, while a PCR based molecular analysis from formalin fixed valvular tissue identified Streptococcus pneumoniae as the etiologic agent.Conclusions: The presented case shows a rare syndrome with a high risk of morbidity and mortality. Following the broad-spectrum treatment and intensive therapeutic support, the patient made unfavourable progress which raised differential diagnosis problems. In this case, the post-mortem diagnosis demonstrated multiple valvular lesions occurred as a result of endocarditis.

ABSTRACT: The aim of this study was to investigate the main causes of death in growing-finishing pigs in southern Brazil. During a one-year period (from 2018 to 2019), two industrial pig herds (18 and 20 thousand pigs each farm) in southern Brazil were monitored along the four seasons of the year (12 days per season on each farm), in order to perform necropsies of all pigs that died in that period. The two farms had an average monthly mortality rate ranging from 0.94 to 3.93% in the evaluated months. At necropsy, tissues were collected, fixed in 10% formalin solution and processed routinely for histopathological examination. When necessary, samples were sent for bacterial culture and PCR to identify etiologic agents. A total of 601 necropsies were performed, with 94.9% of conclusive diagnoses. Infectious diseases corresponded to 64.4% of conclusive diagnosis and non-infectious diseases to 35.6%. The most prevalent causes of death were: pneumonia (33%), gastric ulcers (15.4%), circovirosis (9.9%), systemic bacterial embolism (5.4%), polyserositis (4.4%), dilated cardiomyopathy and torsion of abdominal organs (4.3% each), and bacterial pericarditis (3.4%). Regarding pneumonias (199/601), the main agents identified in these cases were Pasteurella multocida, Influenza A virus and Mycoplasma hyopneumoniae, mainly in associations.

Background: Acute appendicitis is the most common surgical emergency. Obstruction of the lumen by fecolith is the usual cause of acute appendicitis.The aim of the study was to analyze clinical presentation of acute appendicitis and its histopathological correlation. Materials and Methods: A retrospective study of acute appendicitis was done in the Department of Surgery of Helping Hands Community Hospital from January 2009 to December 2010. Three hundred forty five patients out of 415 patients with clinical diagnosis of appendicitis underwent operative treatment. The histopathological reports were reviewed and correlated with clinical diagnosis. Results: Out of 345 patients who underwent operative procedure 98% (n= 338) came with chief complaint of pain in the periumbilical region migrating to the right iliac fossa. The mean age of presentation was 42 years. Increased leucocyte count was seen in only 65% cases. Acute appendicitis was more commonly seen in male patients (214 cases, 62%). The most common per operative finding was acutely inflammed appendix (84%) followed by perforated appendix (7.5%), gangrenous appendix (3.5%) and appendicular lump (1.5%). However, histopathological diagnoses were acute appendicitis (91.9%), resolving appendicitis (3.5%), lymphoid hyperplasia (2.6%), mucocele (0.3%) and carcinoid (0.3%). Normal histology was seen in 1.4% cases. Conclusion: Though there are other causes of acute abdomen, acute appendicitis still stands first amongst all the emergencies. Histopathological examination of appendectomy specimen should not be omitted in order to see the incidence negative appendectomy rate and to avoid complications relating to malignant conditions. Pasteurella multocida, Mannheimia haemolytica and Trueperella pyogenes are three bacterial pathogens closely associated with the bovine respiratory disease complex (BRDC). In the current study, a multiplex PCR for the simultaneous detection of these three bacteria in cultures was established. After serial optimisation, the detection limit of the method for the genomic DNA of the three bacteria was 40 pg/μl. The method could detect the genomic DNA of these three bacteria but not the genomic DNA of seven other bacterial strains. Together with the bacterial enrichment technology, the multiplex PCR could be used for detecting the three bacteria in animal tissues. This method might be valuable for speeding up laboratory diagnosis and directing the treatment of BRDC to these three bacterial pathogens.

ABSTRACT Introduction Making a protocol is a must to overcome the controversy of doing routine histopathological examination of tonsillectomy specimen. Materials and methods Histopathological report of 100 consecutive tonsillectomy specimens that reached the pathology department of Saveetha Medical College was analyzed. Results Histological examination of the 154 pediatric tonsils showed reactive lymphoid hyperplasia in all the patients. One specimen had a small cartilaginous choristoma. In the 46 adult tonsils, there were two lymphoma, one extensive osteocartilaginous choristoma, and one epidermoid cyst of tonsil. Discussion As the histopathological examination in the pediatric group did not reveal any finding, it can be done only in patients with risk factors. The adult group showed two cases of Hodgkins lymphoma, one epidermoid cyst of tonsil and one osteocartilaginous choristoma tonsil. Hence, we suggest to continue the practice of routine histopathological analysis in adults. Conclusion We suggest continuing the practice of routine histopathological examination of all adult tonsillectomy specimens. How to cite this article Shoba K, Harikumar B, Jayaganesh P, Srinivasan K. Background: Illegal migration of pigs/piglets from Myanmar to Mizoram is a common practice to meet the local demands. The migrated animals are suspected as potential carrier of various microbial pathogens. The present study was conducted on isolation, identification and molecular characterization of major bacterial pathogens (Actinobacillus pleuropneumoniae, Bordetella bronchiseptica, Haemophilus parasuis, Mycoplasma hyopneumoniae and Pasteurella multocida) in pigs illegally migrated from Myanmar to Mizoram. Methods: A total of 209 rectal swabs and 209 nasal swabs were collected from apparently healthy migrated pigs during October 2018 to April, 2019. All the samples were processed for PCR based detection of target bacterial species followed by isolation and identification by bacteriological techniques. The bacterial species were further confirmed by BD Phoenix automated bacterial identification system and selected virulence genes of the bacterial species were determined by specific PCR assay. Result: By species specific PCR, 110 samples were found to be positive for selected bacterial species, of which 20 (9.57%), 1 (0.478%), 86 (41.15%), 2 (0.956%) and 1 (0.478%) were A. pleuropneumoniae, B. bronchiseptica, H. parasuis, M. hyopneumoniae and P. multocida, respectively. A total of 52 bacterial strains were isolated and identified, of which 11, 1, 39 and 1 were A. pleuropneumoniae, B. bronchiseptica, H. parasuis, M. hyopneumoniae and P. multocida, respectively. Virulence genes were detected in A. pleuropneumoniae and H. parasuis isolates. Based upon the published literatures, this is the first ever report of isolation and identification of pathogenic A. pleuropneumoniae and H. parasuis in pigs in India.

This study was carried out to evaluate the efficacy of vaccination of layer chickens with inactivated FC bacterin prepared from local Egyptian strains of Pasteurella multocida (P. multocida). A total of 200 layer chickens were divided into 5 equal groups, 40 for each. At the age of 6 weeks, chickens in groups (A) and (B) were vaccinated with P. multocida serotypes A:1 and A:3, respectively, booster doses were given after 3 weeks (9 weeks old) and challenge was done with virulent serotypes A:1 and A:3 at 2 weeks later (11 weeks old). Chickens in groups (C) and (D) were not vaccinated, only challenged with P. multocida serotype A:1 and A:3, respectively. Birds in group (E) were kept as non-vaccinated and non-challenged. Blood samples were collected weekly from all groups for humoral immune response. All the birds were kept under observation for signs, mortalities, lesions and re-isolation of challenging organism and for histopathological examination. Results of the mean Enzyme Linked Immuno-Sorbent Assay (ELISA) revealed that the highest level was at 5 weeks post vaccination as the titers reached to 3970 in group (A) and 3905 in group (B). The clinical signs, mortality rate and lesions were mild in the vaccinated birds while severe lesions were in non-vaccinated and challenged birds. The protection rates were 85 % and 80 % in groups (A) and (B); respectively, while 10 % and 20 % in groups (C) and (D); respectively. The re-isolation rates of P. multocida after challenge were 95 % and 90 % in non-vaccinated-challenged birds with P. multocida serotypes A:1 and A:3; respectively, while they were 25 % and 15 % in vaccinated-challenged groups with P. multocida serotypes A:1 and A:3; respectively. Histopathological examination of P. multocida vaccinated-challenged birds revealed mild to no microscopic lesions when compared with non-vaccinated challenged chickens. In conclusion, the prepared FC inactivated bacterin from the local Egyptian predominant P. multocida serovars proved efficacy and protection of layer chickens. Key words: Pasteurella multocida; chickens; immunization; protection; Egypt UČINKOVITOST CEPLJENJA KOKOŠI NESNIC Z INAKTIVIRANO BAKTERIJO KOLERE PERJADI, PRIPRAVLJENE IZ LOKALNIH EGIPTOVSKIH SEVOV BAKTERIJE Pasteurella multocida Povzetek: Raziskava je bila izvedena z namenom ocenitve učinkovitosti cepljenja kokoši nesnic z inaktivirano bakterijo FC, pripravljeno iz lokalnih egiptovskih sevov bakterije Pasteurella multocida (P. multocida). Skupno 200 kokoši nesnic je bilo razdeljenih v 5 enakih skupin. V vsaki skupini je bilo 40 kokoši. Pri 6 tednih smo kokoši v skupinah A in B cepili s serotipoma P. multocida A:1 in A:3, po 3 tednih, ko so bile živali stare 9 tednov, so dobile poživitvene doze cepiva. Po dveh tednih (v starosti 11 tednov) so bile kokoši okužene z virulentnima serotipoma A:1 in A:3. Piščanci v skupinah C in D niso bili cepljeni temveč samo okuženi s serotipoma A:1 in A:3. Kokoši v skupini E niso bile niti cepljene, niti okužene. Vzorci krvi so bili odvzeti pri vseh skupinah tedensko za preverjanje humoralnega imunskega odziva. Vse kokoši smo stalno opazovali in beležili prisotnost bolezenskih znakov, različnih ran in umiranje kokoši. Pri poginulih kokoših smo osamili bakterije ter opravili histopatološki pregled. Rezultati encimsko-imunskega testa (ELISA) so pokazali da je bila najvišja stopnja zaščite dosežena 5 tednov po cepljenju, saj so titri dosegli 3970 v skupini A in 3905 v skupini B. Klinični znaki, stopnja umrljivosti in rane so bili pri cepljenih kokoših blagi, hude rane pa so bile vidne pri necepljenih in okuženih kokoših. Stopnja zaščite je bila v skupinah A in B 85- oziroma 80-odstotna, v skupinah C in D pa 10- oziroma 20-odstotna. Stopnje ponovne izolacije P. multocida po okužbi so bile 90 in 95 odstotkov pri kokoših, ki niso bile cepljene, medtem, ko so bile v skupinah, ki so bile okužene s P. multocida serotipa A:1 in A:3 15- in 25-odstotkov. Histopatološki pregled cepljenih in okuženih kokoši je pokazal popolno odsotnost ali prisotnost blagih mikroskopskih poškodb, medtem ko so imele necepljene okužene kokoši bolj obsežne histopatološke poškodbe. Pripravljena inaktivirana bakterija FC iz lokalnih egiptovskih prevladujočih serovarov P. multocide se je izkazala za učinkovito zaščito kokoši nesnic.Ključne besede: Pasteurella multocida; kokoši; imunizacija; zaščita; Egipt
