AbstractRegeneration of Slender speedwell (Veronica filiformis) from small parts of stem without using stimulative agents was the focus of the investigation. Four different short fragments of shoot (main terminal, secondary terminal, nodal segment and internodal segment) were cultivated under greenhouse and natural conditions. All tested vegetative segments induced roots, rooted in a soil substrate and in a semi-natural lawn, survived winter, and flowered. Multiplication of clonal plants was confirmed for both the main and secondary terminal segments. These terminal segments had the best response in number of growing individuals, flowering stems, and weight of dried biomass. The manipulative experiment revealed that clonal success of speedwell is connected with possibilities to hive off of all tested above-ground segments. Establishment of plantlets from the segment in natural condition was not successful when accompanied by certain grass species.

We monitored an area that was revegetated with the goal of restoring a Eucalyptus tereticornis open forest on former agricultural land in central, eastern Queensland. Revegetation involved: (1) planting 60 ha of previously cleared and heavily grazed land with eight local trees species; and (2) removing cattle grazing to encourage natural regeneration in areas where some mature trees remained. We compared the revegetation site to native pasture that had also been previously cleared, with only scattered paddock trees remaining, and continued to be managed for livestock production (an area similar to the revegetation site, prior to planting) and a remnant forest (reference area). Nine years since revegetation began there was some evidence that the revegetated site was diverging from pasture in terms of understorey plant composition, sapling density and topsoil C and N. There was little divergence in terms of plant species richness (native, introduced, grass, forb and woody plant richness), herbaceous biomass and woody plant regeneration. Some monitoring plots were subject to fire (prescribed fire and or wildfire) over the period of monitoring. With increasing time since fire, the richness of native species, introduced species and grass species (both native and introduced) declined, and forb and grass species richness declined with increasing litter biomass, suggesting that the occurrence of fire and the associated removal of litter biomass has a positive influence on herbaceous diversity in this ecosystem. Woody plant regeneration persisted through lignotubers at the revegetation site and at the pasture, but this regeneration was stunted at the pasture presumably due to livestock grazing. Hence areas of former E. tereticornis forest showed promising regenerative capacity where mature trees remained and where livestock grazing was removed.

An efficient in vitro regeneration system was developed for Piper betle L. through direct and indirect organogenesis from nodal segment, leaf segment and petiole explants. Highest direct regeneration was recorded when nodal explants were cultured on MS with 1.0 mg/l BAP and 1.0 mg/l Kn where 80% explants produced multiple shoots and the average number of shoots per explants were 3.20. Remarkable results on callus induction and shoot initiation were observed when the explants cultured on MS + 2.0 mg/l BAP + 0.5 mg/l Kn + 1.0 mg/l IAA. It was observed that nodal explants were showed best response on shoot/explants 13.2 ± 4.5 after 8 weeks of callus culture on MS medium with 0.5 mg/l BAP. The best response towards root induction was observed on half strength of MS with 0.25 mg/l IBA. The well rooted plants were successfully acclimatized and transferred to soil.

This study reports on an improved protocol for callus induction and subsequent regeneration from nodal segment of wood apple (Aegle marmelos L.) Creamish friable competent callus was achieved from nodal segments on MS medium augmented with 4.0 mg1-1 2,4-D within two weeks of inoculation. The callus produced large number of shoots when cultured on MS medium fortified with 2.0 mgl-1 BAP+0.1 mgl-1 NAA within ten days of culture. In vitro raised shoots were rooted on half strength MS medium enriched with 1.0 mgl-1 IBA within fifteen days of culture. The rooted plantlets were successfully established with 80% survival. An effort was made to identify the in vitro responsiveness for callus induction in NaCl (salt stress) and subsequent plant regeneration of oats (Avena sativa L.). Callus induction was tested in different concentrations (0, 5, 10, 15 and 20 mg/l) of NaCl salt added in MS. The experiment was laid out in CRD considering 2,4-D growth regulator and NaCl salt as factors. The best response in callus induction was observed on MS containing 3 mg/l 2,4- D and 5 mg/l NaCl where the frequency of callus induction was 68.57%, callus weight was 0.36 g and callus diameter was 7.63 mm. During regeneration, the maximum frequency of regeneration (41.67%), regenerated shoots per callus (14.80), shoot length (12.14 cm), root number (10.90) and root length (3.76 cm) was observed from the callus derived from MS containing 3 mg/l 2,4-D and 5 mg/l NaCl. Plantlets that were regenerated from callus induced in MS with 3 mg/l 2,4-D and 5 mg/l NaCl showed higher rate of ex vitro survivability. Therefore, MS with 3 mg/l 2,4-D and 5 mg/l NaCl found better for in vitro callus induction and subsequent plant regeneration from mature seeds of oat. The response to the attack of Monilinia laxa, Polystigma rubrum and Stigmina carpophila in natural conditions of infection of 13 plum cultivars were evaluated during three years, in a commercial orchard located in North-western of Romania. The studied cultivars were: ‘Topfirst’, ‘Nectarina rosie’, ‘Tuleu timpuriu’, ‘Hangata’, ‘Toptaste’, ‘Tuleu gras’, ‘Vinete de Italia’, ‘Stanley’, ‘Vinete romanesti’, ‘Tophit’, ‘Jojo’,‘Anna Späth’, ‘Topend’. The highest degree of flower attack by Monilinia laxa was recorded for ‘Stanley’, while the lowest attacks were registered on ‘Topend’, ‘Jojo’, ‘Tophit’ and ‘Nectarina rosie’. The best response regarding the attack on fruits proved to be with ‘Topend’, ‘Jojo’, ‘Tophit’ and ‘Topfirst’. These cultivars registered small values of attack degree (0.7-3.3%). The cultivars with the lowest attack degree of red staining caused by Polystigma rubrum were ‘Top End’, ‘Jojo’, ‘Tophit’, ‘Toptaste’, ‘Nectarina rosie’ and ‘Topfirst’. A high degree of attack was calculated on ‘Anna Späth’, ‘Vinete romanesti’, ‘Vinete de Italia’ and ‘Tuleu timpuriu’. These cultivars prove to be very sensitive to the red staining disease in natural conditions of infections. Cultivars ‘Top End’, ‘Jojo’, ‘Tophit’, ‘Toptaste’, ‘Nectarina rosie’ and ‘Topfirst’ were slightly attacked by Stigmina carpophila (5.3-7.3% attack degree). The low infections levels recommend the mentioned cultivars as possible genitors in plum breeding programs, for creating new genotypes with good response to diseases attack.

Tissue culture studies ofCelosia cristatawere established from various explants and the effects of various hormones on morphogenesis of this species were examined. It was found that complete plant regeneration occurred at highest percentage on MS medium supplemented with 2.0 mg/L NAA and 1.5 mg/L BAP, with the best response showed by shoot explants.In vitroflowering was observed on MS basal medium after six weeks. The occurrence of somaclonal variation and changes in cellular behavior fromin vivoandin vitrogrown plants were investigated through cytological studies and image analysis. It was observed that Mitotic Index (MI), mean chromosome numbers, and mean nuclear to cell area ratio ofin vitroroot meristem cells were slightly higher compared toin vivovalues. However,in vitroplants produced lower mean cell areas but higher nuclear areas when compared toin vivoplants. Thus, no occurrence of somaclonal variation was detected, and this was supported by morphological features of thein vitroplants.

Abstract Passiflora setacea DC (Passifloraceae) is considered an important species in the genetic breeding of passion fruit. However, its use is limited due to low seed germination. This paper aimed to study the effect of cytokinins 6-benzyladenine (BA) and thidiazuron (TDZ) on the in vitro morphogenesis of P. setacea using three explants: hypocotyl, nodal segment, and root segment. The explants were induced to morphogenesis in MS medium modified and with different concentrations of BA and TDZ. After 55 days, the percentage of calluses and shoots were evaluated, and anatomical characterization was performed. The three explants used in the in vitro morphogenesis of P. setacea showed callus and shoots formation, but in greater numbers in the nodal segments treated with BA. TDZ isolated affected equal to or less than BA on callus and shoots formation for the three explants. Direct and indirect organogenesis was observed in the three types of explants. From the results obtained for plant regeneration via in vitro morphogenesis of P. setacea, it is recommended to use a nodal segment in MSM medium and supplemented with 2.22 μmol L-1 of BA.

Air, baik air tanah maupun air permukaan merupakan sumber kehidupan yang tidak dapat tergantikan oleh apa pun dan tanpa air, manusia, hewan dan tanaman tidak akan dapat hidup terutama sebagai air minum. Tetapi dalam pemanfaatannya, masih banyak yang tidak sesuai dengan peruntukannya ataupun mempergunakan sampai diluar batas, sehingga disatu sisi banyak yang terbuang dengan sia-sia tetapi dilain sisi banyak yang membutuhkan. Penggunaan air selama dasa warsa terakhir ini meningkat disebabkan oleh beberapa faktor, antara lain oleh kegiatan manusia yang semakin bervariasi, pertambahan jumlah penduduk yang meningkat, maka jumlah air untuk kebutuhan manusia juga meningkat dan hal ini berpengaruh terhadap kualitas air. Selain itu juga berpengaruh terhadap keterbatasan air dan kondisi alam. The effect of cytokinins on callus proliferation from cotyledons and plantlet development was studied in cotton. The frequency of callus induction was observed on MS medium enriched with a variety of cytokinins in different concentrations. With the increase of cytokinin concentration, the percentage of callus formation, percentage of shoot developing calli and number of shoots/calli were increased. Among the three different cytokinins studied, BA showed the highest performance. The highest percentage of callus (6.55%) and shoot developing calli (5.87%) was obtained on MS with 1.0 mg/l BA. Highest number of shoots (3.02) per calli was observed on MS media supplemented with 1.0 mg/l Kn. The rooting media composed of MS medium, 0.6% agar, sucrose and fortified with 2.0 mg/l NAA induced root development at the highest percentage (41.23%) with maximum number of roots (3.61) per cutting and length of root (3.62 cm) per culture. The plantlets were acclimatized in natural conditions.
