An effort was made to identify the in vitro responsiveness for callus induction in NaCl (salt stress) and subsequent plant regeneration of oats (Avena sativa L.). Callus induction was tested in different concentrations (0, 5, 10, 15 and 20 mg/l) of NaCl salt added in MS. The experiment was laid out in CRD considering 2,4-D growth regulator and NaCl salt as factors. The best response in callus induction was observed on MS containing 3 mg/l 2,4- D and 5 mg/l NaCl where the frequency of callus induction was 68.57%, callus weight was 0.36 g and callus diameter was 7.63 mm. During regeneration, the maximum frequency of regeneration (41.67%), regenerated shoots per callus (14.80), shoot length (12.14 cm), root number (10.90) and root length (3.76 cm) was observed from the callus derived from MS containing 3 mg/l 2,4-D and 5 mg/l NaCl. Plantlets that were regenerated from callus induced in MS with 3 mg/l 2,4-D and 5 mg/l NaCl showed higher rate of ex vitro survivability. Therefore, MS with 3 mg/l 2,4-D and 5 mg/l NaCl found better for in vitro callus induction and subsequent plant regeneration from mature seeds of oat. A suitable plant regeneration system has been established using 3-4 weeks old calli derived from immature and mature seeds of four wheat varieties viz. Pavon 76, Akbar, Barkat, and Kanchan. As plant growth regulators, various auxins (2,4-D, BAP and IAA) either single or in combination were used in MS medium. The variety Pavon 76 showed maximum (72.25%) callus induction and Akbar exhibited the lowest (37.78%) from calli derived from immature seeds. Hormonal effects on callus induction were evaluated and significant results were found in case of genotypes at P <0.01. Out of four genotypes, the highest frequency of plant regeneration was recorded in Pavon 76 (67.00%) and lowest in Kanchan (43.10%) when 1.5 mg/l BAP and 0.5 mg/l IAA was added in the medium. It was observed that Pavon 76 produced highest number of green plants than others. For mature seeds all of the mentioned genotypes showed significant difference with maximum frequency of callusing in Pavon 76 (69.57%) in MS + 2.5 mg/l 2,4-D followed by Kanchan (60.84%), Barkat (52.73%), and Akbar (47.19%). For plant regeneration, Pavon 76 also showed best performance (64.36%) in MS + 2.0 BAP + 1.0 mg/l IAA, using calli derived from mature seeds. The other genotypes Barkat, Kanchan and Akbar exhibited 59.44, 52.71 and 52.32% regeneration in the same medium respectively. Here, the lowest regeneration (40.63%) was found in Akbar. In this case, it was aimed to establish a suitable protocol for in vitro callus induction and regeneration for advance biotechnological research on wheat in Bangladesh.

Lychnis cognate Maxim and Lychnis fulgens Fish. Ex Spreng are two valued ornamental plants in Korea. Soaking of seeds in GA3 solution remarkably promoted germination up to 60%, but the control (0 mg/l) was not effective (> 5%). To select an adequate temperature for seed germination, seeds, previously soaked in a 1000 mg/l GA3 for 24 hrs, were incubated at 15, 20, 25, and 30°C. Seed germination of over 20% was obtained at 15, 20, and 25°C, but only 10% at 30°C. These results indicate that the seeds of L. cognate and L. fulgens are in a such dormant state that they hardly germinate even by dormancy breaker (GA3) and low (15 ? 25°C) temperature treatment. The highest callus induction was observed in the leaf explants of the seedlings on MS containing specific concentrations of 3.0 mg/l BA and 1.0 mg/l NAA. The adventitious shoot was formed < 90% of calli on 1/2 WPM medium. The height of in vitro propagated plantlet was no different media used for regeneration. The experiment was conducted to investigate the performance of three different genotypes (BARI Sarisha-6, BARI Sarisha-8, and BARI Sarisha-11) in two different media viz., MS and B5 with different concentrations of phytohormone (2, 4-D) for callus induction from uninucleate stage anthers of Brassica and subsequent plant regeneration in MS media with different concentrations of phytohormone (BAP and NAA). Among the genotypes, BARI Sarisha-8 showed the best performance for all the parameters of callus induction. The performance of BARI Sarisha-6 was poor compared to others. Maximum rate of callus induction (%) was observed in MS + 0.5 mg/L 2, 4-D followed by B5 + 0.5 mg/L 2,4-D. The media combination MS + 1.0 mg/L BAP 0.3 mg/L 2,4-D showed the best performance for maintenance of calli. Significant variations were observed among the genotypes and media composition for shoot regeneration. Among the genotypes, BARI Sarisha-8 showed the best performance for shoot regeneration followed by BARJ Sarisha-l1. The genotype BARI Sarisha-8 produced higher percent of shoots/calli and required minimum days for shoot initiation. Higher percent calli without shoot were produced by the genotype BARI Sarisha-6. The media combination MS + 2.0 mg/L BAP + 0.5 mg/L NAA showed the best performance for shoot regeneration and required maximum days for shoot initiation. Salinity is a major problem affecting crop production all over the world. Excessive soil salinity can reduce the productivity of many agricultural crops including many vegetables and spices. Onion is one of the most important spices in the Asiatic region which is now in high demand. The experiment was conducted to observe in vitro regeneration of onion (Allium cepa L.) under salt stress condition from September 2016 to July 2017. The experiment was conducted as two factorial (genotype and treatment) Completely Randomized Design (CRD) with 3 replications for each treatment. Shoot tip segments of three genotypes namely Faridpuri, Taherpuri and Pusa red (Indian) were cultured in MS (Murashige and skoog, 1962) media supplemented with 25, 50, 75 and 100 mM NaCl. The genotype Faridpuri gave maximum salt tolerance upto 100 mM salinity level with 10.60 cm shoot length and 1.94 cm root length having the highest relative shoot and root growth. Pusa red was found to be salinity sensitive genotype which showing lowest shoot length of 7.03 cm and root length of 0.96 cm at 100 mM NaCl treatment. However, Taherpuri was tolerant up to 100 mM salinity level with 8.14 cm shoot length and 1.25 cm root length. Both the highest fresh weight of root (54.77 mg) and dry weight of root (41.36 mg) was from the genotype Faridpuri with 25 mM NaCl treatment. However, a convenient in vitro regeneration protocol of onion genotypes under different salinity level has been developed and the genotype Faridpuri can be used for further investigation in field condition to evaluate its performance at various salinity levels.

ABSTRACT Indonesia is rich in rice genetic diversity in the form of javanica rice. Subspecies javanica have some superiority characters, among others a strong plant tissue, but in generally they have low productivity. Weaknesses of javanica rice can be repaired by biotechnology through genetic transformation techniques from callus of plant. Auxin types and genotypes are important factors for callus induction and plant regeneration. Therefore, this research held for know callus introduction and plant regeneration of three varieties of Javanica rice (Pendok, Genjah Arum, and Menthik Wangi Susu) combined with type of auxin (2,4-D and pychloram). The experimental design of RALs two factorials, there are three varieties of rice (Pendok, Genjah Arum, and Menthik Wangi Susu) and two types of auxin (2,4-D and Pychloram) and analyzed by DMRT α5%. The results of this study is Mentik Wangi Susu have highest regeneration than Pendok and Genjah Arum. The use of 2,4-D resulted in a higher callus induction and plant regeneration than pychloram in the three rice varieties used. While the best combination of treatments for callus induction and regeneration is Menthik Wangi Susu and 2,4-D. Keyword: auxin, javanica, callus induction, plant regeneration. ABSTRAK Indonesia kaya akan keragaman genetik padi berupa padi javanica. Padi sub spesies javanica memiliki beberapa keunggulan diantaranya jaringan tanaman yang kuat, namun pada umumnya mempunyai tingkat produktivitas gabah yang rendah. Kelemahan pada padi javanica dapat diperbaiki dengan bioteknologi melalui teknik transformasi genetik menggunakan jaringan kalus. Genotip dan jenis auksin merupakan faktor penting dalam induksi kalus dan regenerasi tanaman padi secara in vitro. Oleh karena itu, penelitian ini dilaksanakan untuk mengetahui daya pembentukan kalus dan regenerasi pada tiga varietas padi javanica, yaitu Pendok, Genjah Arum, dan Menthik Wangi Susu yang dikombinasikan dengan perlakuan jenis auksin berupa penggunaan 2,4-D dan pikloram. Rancangan percobaan yang digunakan adalah RAL dua faktorial yaitu varietas tanaman padi dan jenis auksin  dengan pengujian  menggunakan DMRT α5%. Hasil penelitian menunjukkan bahwa dari ketiga varietas padi yang digunakan, varietas Mentik Wangi Susu menunjukan daya regenerasi yang tertinggi dibandingkan varietas Pendok dan Genjah Arum.  Penggunaan 2,4-D menghasilkan tingkat pengkalusan dan regenerasi yang tinggi dibandingkan pikloram pada tiga varietas padi yang digunakan. Sedangkan kombinasi perlakuan yang terbaik untuk induksi kalus dan daya regenerasi adalah penggunaan varietas Menthik Wangi Susu dan auksin dalam bentuk 2,4-D.  Kata Kunci: auksin, javanica, induksi kalus, regenerasi tanaman.

AbstractThis study examines the effects of light emitting diodes (LEDs) on tissue culture proliferation of Acacia melanoxylon plantlets among five different clones (FM1, FM2, FM4, FM5, and FM10). Shoot bud apex cuttings were transplanted onto Murashige and Skoog basal medium containing 0.1 mg L-1 6-benzyladenine and 0.5 mg L-1 naphthalene acetic acid and cultured in vitro for 40 days. Root growth was studied under different light intensities and photoperiods ex vitro. The bud proliferation coefficient was greatest under a light intensity of 45 μmol m-2 s-1 photosynthetic photon flux and photoperiod of 16 h light, but decreased as the light intensity increased. However, the greatest light intensity was beneficial for the growth of robust plantlets. Plantlets exposed to red and blue LED combinations grew tall and green, with a small number of roots. Plantlets also grew taller and some roots expanded under the longer photoperiod. Increased light intensity had positive effects on root number and rooting rate, and prolonged light greatly increased root number. Therefore, lower light intensity and a short photoperiod were beneficial for bud proliferation, while red/blue LED combinations, increased light intensity, and longer light illumination were beneficial for plantlet growth and root growth of Acacia melanoxylon.

Anther of five varieties of Brassica species, namely BARI Shariaha-7, Tori-7, Agrani, Daulat and Safal were cultured in vitro to observe their regeneration potentiality. Different concentrations and combinations of growth regulators were supplemented in MS medium. The range of callus induction was 12.50-87.50 %. Maximum callus induction (75.00%) was observed on MS +4 mg/L 2, 4-D + 1.0 mg/L BAP. Among the genotypes, BARI Sharisha-7 showed the highest percentage of callus induction (60.42%). Among the treatments, highest percentage of shoot regeneration (75.00%) was observed on MS + 4 mg/L BAP + 1.0 mg/L NAA. BARI Sharisha-7 also showed the highest rate of plant regeneration (66.67%). Root induction was highest (75%) on half strength MS medium supplemented with 1.0 mg/L IBA and 0.5 mg/L NAA. The plantlets with sufficient roots thus obtained were transferred successfully to plastic pots and subsequently to the field. BARI Sharisha-7 and Tori-7 survived easily in the pots as well as in the field but Safal was very poor in survivability both in the pots and in the field. This study reports on an improved protocol for callus induction and subsequent regeneration from nodal segment of wood apple (Aegle marmelos L.) Creamish friable competent callus was achieved from nodal segments on MS medium augmented with 4.0 mg1-1 2,4-D within two weeks of inoculation. The callus produced large number of shoots when cultured on MS medium fortified with 2.0 mgl-1 BAP+0.1 mgl-1 NAA within ten days of culture. In vitro raised shoots were rooted on half strength MS medium enriched with 1.0 mgl-1 IBA within fifteen days of culture. The rooted plantlets were successfully established with 80% survival.
