Background: High fat diets are known to cause a positive fat balance and consequently to the accumulation of adipose mass, this diet does not seem to stimulate fat oxidation in the same way in obese and lean subjects. HFD was an inducing factor for ICAM-1 expression in the aorta of Wistar rats. HFD effect on ICAM-1 seems to be time dependent. ICAM-1 is one of the first events in the formation of atherosclerotic lesions. HFD up-regulated Cav-1, regulated expression other biomarker in HFD is eNOS. Recent studies showed that E. longifolia Jack protected HFD animal model from atherosclerosis based on the reduce atherosclerotic plaque size and formation HFD-rats treated with E. longifolia Jack.Objective: To prove that Eurycoma longifolia has anti inflammatory effect on endothelial cell blood vessels of Sprague Dawley rat with high fat diet.Method: Study design was experimental study, by used Randomized Post Test only Control Group Design with Kruskal-Wallis test was used to analyze the differences among groups and followed by a Mann Whitney test. Treatment is ethanolic or water extract of Eurycoma longifolia Jack, and out come are sICAM-1 and eNOS levels. Thirty Sprague Dawley (SD) Rat, were divided into 6 groups. C(-) was SD group, C(+) was group with HFD, X1 (SD treated with EL dosage 10 mg/kg), X2 (SD treated with EL dosage 15 mg/kg), X3 (HFD treated with EL dosage 10 mg/kg), X4 (HFD treated with EL dosage 15 mg/kg).

Background: Treatment with buceng combination of Eurycoma longifolia Jack and Pimpinella alpine Molk has been proven to increase testosterone level, decrease apoptosis and caspase3 expression. Bcl2 is an antiapoptotic protein found in cytoplasm which inhibits cells apoptosis. This study was aimed to investigate the effect of buceng on Bcl2 expression on penile and prostate tissues of the rats. Methods: In this experimental study, 24 male Sprague Dawley rats of 90 days old, weighing ± 300 grams, were randomly assigned into four groups. Group A, normal rats. Group B, castrated rats and treated with buceng 100 mg/day, per oral (Cast-Bcg); Group C, castrated rats and treated with 2 ml of water as placebo against buceng (Cast-Plac). Group D, castrated rats, treated with mesterolone 6.75 mg/day, per oral, as exogenous testosterone (Cast-Mest). All rats were treated for 30 days. Manova test was used to analyze the different expression of Bcl2 among groups with significance level at p ≤ 0.05. Results: Castration was associated with significant decrease of Bcl2 expression in the penile and prostate tissues (53.0 and 50.9%, respectively) compared to normal rats (82.6 and 84.2%, respectively, p < 0.001). Treatment with mesterolone reversed Bcl2 expression (77.1 and 78.1%) to a near normal level. The same level of Bcl2 expression was also observed with buceng treatment (73.8 and 78.2%).Conclusion: The treatment with buceng could enhance Bcl2 expression in penile and prostate tissues, comparable to normal rats and mesterolone treated rats.

Doxorubicin chemotherapy agents cause serious side effects in normal cells, especially in the liver and heart caused by free radicals. RAS is an important component of the signal transduction pathway that controls cell proliferation, differentiation and survival. The pasak bumi root (Eurycoma longifolia, Jack) has been reported to have acted as an antioxidant and hepatoprotector. This study aims to determine the effect of giving ethanol extract of pasak bumi root on HRAS expression in healthy mice given doxorubicin. This research was conducted by dividing the test animals into 7 groups, namely doxorubicin control group 4.67 mg / kgBB, group II control ethanol extract of pasak bumi roots 200 mg / kgBB, group III, IV and V were the treatment groups given doxorubicin 4.67 mg / kgBW with ethanol extract of the pasak bumi root, each 50 mg / kgBB; 100 mg / kgBB; 200 mg / kgBW, group VI solvent control and group VII healthy control. The study was continued by painting using an immunohistochemical method to see the expression of HRAS in each experimental group. Data analysis was performed by calculating per cent expressions, then proceed with statistical tests using SPSS. The results of statistical tests showed that there were significant differences between the doxorubicin control group (18.56 � 1.85) and the treatment of group 5 (10.43 � 1.71) and there were no significant differences between the doxorubicin control group (18.56 � 1, 85) with group 3 treatment (17.80 � 4.23) and group 4 treatment (16.70 � 1.97). From the results of the study concluded that the ethanol extract of pasak bumi roots at a dose of 200 mg / KgBB can reduce the expression of HRAS in the administration of doxorubicin

Akar pasak bumi mengandung senyawa kuasinoid diduga mempunyai efek penghambatan karsinogenesis, antiulcer, dan antimalaria. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian eksktrak etanol akar pasak bumi terhadap ekspresi p53 pada kanker payudara tikus betina yang diinduksi DMBA.Tikus dibagi menjadi 6 kelompok. Kelompok I, II dan III diberi ekstrak etanol akar pasak bumi dosis berturut-turut 100; 200 dan 400 mg/kg BB. Kelompok IV diberi larutan DMBA 20 mg/kg BB. Kelompok V diberi corn oil. Kelompok VI sebagai baseline. Tikus yang mengalami kanker, dihitung jumlah nodul tumornya dan pada minggu ke-23 semua tikus dikorbankan dan diambil jaringannya untuk pengamatan secara mikroskopik menggunakan metode HE dan imunohistokimia. Hasil penelitian menunjukkan bahwa rerata ekpresi p53 mutant pada pemberian ekstrak berturut-turut 100, 200 dan 400 mg/kg BB adalah 6,375 ± 4,07; 11,25 ± 16,53; dan 0,875 ± 1,75; kelompok  IV (DMBA) sebesar 13,125 ± 14,4; kelompok V (corn oil) adalah 1,375 ± 1,55; kelompok VI (baseline) adalah 0,375 ± 0,75. Hasil penelitian disimpulkan bahwa ekstrak etanol akar pasak bumi (Eurycoma longifolia Jack) dosis 400 mg/kg BB yang diberikan sebelum dan selama induksi DMBA mampu menurunkan ekspresi protein p53 mutant (proapotosis) dan mampu menghambat pertumbuhan kanker payudara tikus betina Sprague Dawley yang diinduksi 7,12-dimetilbenz(α)antrasen (DMBA).Kata kunci:  akar pasak bumi (Eurycoma longifolia Jack), kemopreventif, imunohistokimia, ekspresi p53

Kanker payudara dapat terjadi karena paparan dari senyawa 7,12-dimethylbenz (a) antrasene (DMBA) yang bersifat mutagenik dan karsinogenik.  Drug of choice untuk kanker payudara adalah Doxorubicin, namun menimbulkan efek samping dan toksik pada sel normal maka diperlukan penelitian ko-kemoterapi dalam hal ini digunakan fraksi etil asetat akar pasak bumi yang dapat bersifat sebagai antiproliferasi.  penelitian ini adalah untuk menelusuri mekanisme ko-kemoterapi akar pasak bumi dan Doxorubicin terhadap aktivitas proliferasi  protein Ki-67 pada Tikus SD yang diinduksi DMBA. Akar pasak bumi (Eurycoma longifolia Jack) diekstraksi menggunakan pelarut etanol 70 % dan difraksinasi menggunakan etil asetat. Fraksi etil asetat akar pasak bumi diuji mekanisme molekulernya secara in vivo terhadap 5  Kelompok tikus  galur Sprague Dawley terdiri atas Kelompok 1 hanya diberi pakan dan minum, Kelompok 2 diberikan DMBA dosis 20 mg/kgBB, Kelompok 3 diberikan DMBA 20 mg/kgBB + Doxorubicin 1,17 mg/kgBB, Kelompok 4 diberikan DMBA 20 mg/kgBB + fraksi etil asetat akar pasak bumi 100 mg/kgBB, Kelompok 5 diberikan DMBA 20 mg/kgBB + Doxorubicin 1,17 mg/kgBB + fraksi etil asetat akar pasak bumi 100 mg/kgBB kemudian dilakukan pembedahan dan uji imunohistokimia  analisis data Kolmogorof  Smirnof, uji Levene dan uji Kruskal Wallis. Hasil nilai rata-rata % ekspresi protein Ki-67 juga terjadi penurunan pada kelompok ko-kemoterapi DMBA + Doxorubicin + APB (8,89±3,20)% dibandingkan kelompok DMBA+Doxorubicin (12,73±3,37)% dan kelompok DMBA+APB  (11,37±5,16)%, dan terdapat perbedaan yang bermakna dengan kelompok DMBA+Doxorubicin  dengan nilai signifikansi 0,020 (p<0,05) namun tidak berdeda bermakna dengan DMBA+APB  dengan nilai signifikansi 0,114 (p<0,05). Kesimpulan dari penelitian ini adalah pemberian ko-kemoterapi fraksi etil asetat akar pasak bumi dapat menurunkan ekpresi protein Ki-67 pada tikus model kanker payudara yang diinduksi DMBA

Pasak bumi root (Eurycoma longifolia Jack) contains quasinoid. Quasinoid potentially has antitumor/anticancer, anti-parasitic, and immunostimulatory activity. The aim of this research was to study the lymphocyte proliferation activity MTT-test of ethanol extract of Eurycoma longifolia Jack root on induced by 7,12-dimethylbenz[a] antracene (DMBA) female Sprague Dawley rat. The test was done at 6 groups of 10 SD rat each. Each groups was administered orally with DMBA (Group I) dose of 20 mg/kg bw, corn oil (Group II), baseline (Group III), and  ethanol extract of pasak bumi root dose 12.6; 25.2; and 50.4 mg/kg bw (Group IV,V,and VI). Ethanol extract is given for 5 weeks. At the second week five rats were sacrificed to isolate the lymphocytes of spleen then rats are injected orally with DMBA 2 times a week for 5 weeks. At the eight until twenty fourth week all groups were just administered by aquades. Measurement of spleen relative weight and isolation of the lymphocyte spleen were done at second week and the last of experiment. Lymphocyte proliferation activity were measured by MTT-reduction method. Data were analyzed statistically by analyzes of variance (anova) continued by LSD test and Kruskall Wallis continued by Mann Whitney test. The result showed that limphocyte proliferation activity MTT of ethanol extract of E. longifolia root doses of 12.6; 25.2; and 50.4 mg/kg bw for 14 days prior to DMBA-induced are  339.35 ± 8.43;61.20 ± 6.27;310.28 ±  6.81   and  dose 50,4 mg/kg bw after  DMBA-induced is 122.108 ±  11.426 .Keyword: E. longifolia root, spleen,  lymphocyte cell, 7,12-Dimetilbenz (a) antracene (DMBA)

Eurycoma longifolia (EL) is a species that have economical benefit as medicinal ingredients for human health. This species grows in tropical forest areas with high rainfall. Research on the ecology of EL has been done a lot, but research related to ethnotaxonomic, bioecology that includes morphology, ecology, and the association of EL with other plants hasn’t been done much. This study aim to analyze the bioecology of EL plant. The method is literature study, which is looking for literature on the results of research that has been carried out related to EL. The results showed that EL has long been known by the communities of Indonesia and other countries in Southeast Asia with several local names. This plant is also associated with other plants around where it grows, one of which is kempas (Koompassia malaccensis). Based on the results of literature review, research on natural regeneration of EL hasn’t been done much. Therefore, there is a need for research related to the natural regeneration of EL. In addition, it is necessary to conduct research related to the characteristics of the place to grow for optimal cultivation of EL. Thus, the preservation of EL in the future can be maintained.
