To protect themselves, plants can produce toxic secondary metabolites (phytotoxins) that appear with widely varying structures and negative effects. These phytotoxins often show similar properties as known aquatic micropollutants in terms of mobility, persistence, toxicity, and possibly also ecotoxicity. However, their occurrence in surface waters remains largely unknown, which is also due to unknown ability of available screening approaches to detect them. Therefore, we performed a target and suspect screening based on a persistence-mobility prioritization for phytotoxins in small Swiss creeks using high resolution mass spectrometry. In total, three of 26 targets were detected, three of 78 suspects tentatively identified, and six suspects fully confirmed by reference standards. To the best of our knowledge, it is the first time that three different plant secondary metabolite classes are detected in the same surface water sample. Estrogenic isoflavones were detected at 73% of the sites with formononetin as main toxin, which is in agreement with previous studies. Furthermore, pyrrolizidine alkaloids and the indole alkaloid gramine were detected. Especially pyrrolizidine alkaloids might be critical due to their production by various plants including the invasive Senecio inaequidens, and their known importance in food and feed safety. Based on these first screening results, different phytotoxin classes should be assessed for their ecotoxicological effects and considered in future water monitoring.

The objective of this quantitative survey (n=520) was to determine the awareness of the risks of mycotoxin contaminated food and feed throughout the Flemish population. The study highlighted gaps in food and feed safety knowledge regarding mould growth and consequently the presence of mycotoxins. Most people were aware of the negative effects of mould species in food or feed. Around 60% of the people picked out moulds as the producers of mycotoxins, but the majority did not have an idea about the toxicity of these toxins, especially towards plants. Respondents were also asked about possible food products where mould growth could occur and who is responsible for guarding the safety of food and feed with respect to moulds and mycotoxins. All answers and considerations are presented and discussed. Education on mycotoxins should be the focus of the whole population in the near future.

The Lateral Flow Immunoassay (LFIA) is by far one of the most successful analytical platforms to perform the on-site detection of target substances. LFIA can be considered as a sort of lab-in-a-hand and, together with other point-of-need tests, has represented a paradigm shift from sample-to-lab to lab-to-sample aiming to improve decision making and turnaround time. The features of LFIAs made them a very attractive tool in clinical diagnostic where they can improve patient care by enabling more prompt diagnosis and treatment decisions. The rapidity, simplicity, relative cost-effectiveness, and the possibility to be used by nonskilled personnel contributed to the wide acceptance of LFIAs. As a consequence, from the detection of molecules, organisms, and (bio)markers for clinical purposes, the LFIA application has been rapidly extended to other fields, including food and feed safety, veterinary medicine, environmental control, and many others. This review aims to provide readers with a 10-years overview of applications, outlining the trends for the main application fields and the relative compounded annual growth rates. Moreover, future perspectives and challenges are discussed.

Mycotoxins represent an assorted range of secondary fungal metabolites that extensively occur in numerous food and feed ingredients at any stage during pre- and post-harvest conditions. Zearalenone (ZEN), a mycotoxin categorized as a xenoestrogen poses structural similarity with natural estrogens that enables its binding to the estrogen receptors leading to hormonal misbalance and numerous reproductive diseases. ZEN is mainly found in crops belonging to temperate regions, primarily in maize and other cereal crops that form an important part of various food and feed. Because of the significant adverse effects of ZEN on both human and animal, there is an alarming need for effective detection, mitigation, and management strategies to assure food and feed safety and security. The present review tends to provide an updated overview of the different sources, occurrence and biosynthetic mechanisms of ZEN in various food and feed. It also provides insight to its harmful effects on human health and agriculture along with its effective detection, management, and control strategies.

Recent studies on multiple continents indicate members of the Fusarium tricinctum species complex (FTSC) are emerging as prevalent pathogens of small-grain cereals, pulses, and other economically important crops. These understudied fusaria produce structurally diverse mycotoxins, among which enniatins (ENNs) and moniliformin (MON) are the most frequent and of greatest concern to food and feed safety. Herein a large survey of fusaria in the Fusarium Research Center and Agricultural Research Service culture collections was undertaken to assess species diversity and mycotoxin potential within the FTSC. A 151-strain collection originating from diverse hosts and substrates from different agroclimatic regions throughout the world was selected from 460 FTSC strains to represent the breadth of FTSC phylogenetic diversity. Evolutionary relationships inferred from a 5-locus dataset, using maximum likelihood and parsimony, resolved the 151 strains as 24 phylogenetically distinct species, including nine that are new to science. Of the five genes analyzed, nearly full-length phosphate permease sequences contained the most phylogenetically informative characters, establishing its suitability for species-level phylogenetics within the FTSC. Fifteen of the species produced ENNs, MON, the sphingosine analog 2-amino-14,16- dimethyloctadecan-3-ol (AOD), and the toxic pigment aurofusarin (AUR) on a cracked corn kernel substrate. Interestingly, the five earliest diverging species in the FTSC phylogeny (i.e., F. iranicum, F. flocciferum, F. torulosum, Fusarium spp. FTSC 8 and 24) failed to produce AOD and MON, but synthesized ENNs and/or AUR. Moreover, our reassessment of nine published phylogenetic studies on the FTSC identified 11 additional novel taxa, suggesting this complex comprises at least 36 species.

Mycotoxins are a group of naturally occurring toxins that are produced by different filamentous fungi genera such as Aspergillus, Penicillium, Fusarium, etc. The word mycotoxin literally is derived from Greek word “myke” meaning fungus and “toxicum” meaning toxin. These contaminants can develop on different food and feed commodities during different stages including pre-harvest, harvest, and storage. Mycotoxins are of concern because their outbreak result in animal and human diseases and economic losses. It has been estimated that global post-harvest losses are approximately at 50%. Human exposure to mycotoxins is typically through consumption of contaminated agricultural products or indirectly by consumption of animal products containing mycotoxins or their metabolites. The chapter provides the latest information on mycotoxin issues and challenges related to food and feed safety.
