Secretory trichomes have been described on johnsongrass leaves previously, but there is little knowledge of their ultrastructure or the nature of their secretion. Since johnsongrass is a pernicious weed, there is much interest in the nature of compounds found on its foliar surface, and the possible effect of these extrusions on herbicide efficiency. This report describes the results of an investigation into the ultrastructure of the secretory apparatus, and into the nature of the secretion.

ABSTRACT Sec20p is an essential endoplasmic reticulum (ER) membrane protein in yeasts, functioning as a tSNARE component in retrograde vesicle traffic. We show that Sec20p in the human fungal pathogen Candida albicans is extensively O mannosylated by protein mannosyltransferases (Pmt proteins). Surprisingly, Sec20p occurs at wild-type levels in a pmt6 mutant but at very low levels in pmt1 and pmt4 mutants and also after replacement of specific Ser/Thr residues in the lumenal domain of Sec20p. Pulse-chase experiments revealed rapid degradation of unmodified Sec20p (38.6 kDa) following its biosynthesis, while the stable O-glycosylated form (50 kDa) was not formed in a pmt1 mutant. These results suggest a novel function of O mannosylation in eukaryotes, in that modification by specific Pmt proteins will prevent degradation of ER-resident membrane proteins via ER-associated degradation or a proteasome-independent pathway.

The gypsy retroelement of Drosophila moves at high frequency in the germ line of the progeny of females carrying a mutation in the flamenco (flam) gene. This high rate of de novo insertion correlates with elevated accumulation of full-length gypsy RNA in the ovaries of these females, as well as the presence of an env-specific RNA. We have prepared monoclonal antibodies against the gypsy Pol and Env products and found that these proteins are expressed in the ovaries of flam females and processed in the manner characteristic of vertebrate retroviruses. The Pol proteins are expressed in both follicle and nurse cells, but they do not accumulate at detectable levels in the oocyte. The Env proteins are expressed exclusively in the follicle cells starting at stage 9 of oogenesis, where they accumulate in the secretory apparatus of the endoplasmic reticulum. They then migrate to the inner side of the cytoplasmic membrane where they assemble into viral particles. These particles can be observed in the perivitelline space starting at stage 10 by immunoelectron microscopy using anti-Env antibodies. We propose a model to explain flamenco-mediated induction of gypsy mobilization that involves the synthesis of gypsy viral particles in the follicle cells, from where they leave and infect the oocyte, thus explaining gypsy insertion into the germ line of the subsequent generation.

Gastric secretion in response to insulin hypoglycemia and electrical stimulation of the vagus was studied in 18 dogs who had bilateral vagosplanchnic anastomoses in the chest. In six dogs the pattern of gastric secretory response to insulin changed from negative in the early postoperative period to positive between 85 and 613 days postanastomosis. In two, apparent return of vagus function was confirmed by electrical stimulation of the vagi. One of five dogs in whom splanchnovagal nerve anastomoses were performed showed a return of response to insulin at 63 days which was abolished by excision of the anastomoses. On the basis of a review of some of the ideas regarding interpretation of cross nerve anastomoses and some of the conflicting opinions regarding the fiber content of the sympathetic splanchnic nerves, the authors conclude the most likely explanation for the observed phenomena is that there are preganglionic cholinergic fibers in the greater splanchnic nerves whose relationship to the gastric secretory apparatus is similar to that of cholinergic fibers in the vagus. The regenerating fibers of the vagus followed the sheaths of these degenerating fibers and re-established functional relationship with the gastric secretory apparatus.
