Bioassay guided fractionation led to the isolation of a new triterpene glycoside, holothurinogenin B (1) along with three known compounds, holothurin B (2), holothurin A (3), and holothurin A2 (4), from the methanol extract of the Vietnamese sea cucumber Holothuria scabra. Their structures were deduced from the spectral analysis (1D-NMR, 2D-NMR, MS) and chemical evidences.

The unusual non-holostane triterpene glycoside, colochiroside E (1) was isolated from the sea cucumber Colochirus robustus (Cucumariidae, Dendrochirotida). The structure of 1 was established by analysis of ID, 2D NMR and HRESI MS data. Colochiroside E (1) belongs to a rare group of glycosylated 9β-H-lanosta-18(16)-lactones and has an unprecedented sulfated trisaccharide carbohydrate chain consisting of two glucose and one xylose units. In contrast with (9β-H)-7(8)-unsaturated holostane glycosides, the 7(8)-double bond in the having (9β-H)-configuration aglycone of colochiroside E is not capable of migration into the 8(9)- and then into the 9(11)-position on treatment with HC1. The formation of a chlorine derivative of 1 was observed under these conditions.

<strong>Extraction and Oral Suspension Formulation of Sea Cucumber Holothuria scabra as Source of Antioxidants.</strong> Sea cucumbers have long been utilized in the food and medicine for Asia and Middle East communities. Indonesia is one of the main suppliers in the trade of sea cucumbers in Asia. Sea cucumber utilization in Indonesia is generally done only in the form of raw materials. Holothuria scabra is one of the sea cucumber that can be explored as a potential source of antioxidants. This study aims to determine the antioxidant activity of sea cucumber H.scabra in extract and suspension. Sea cucumber was extracted with methanol 96% and ethanol 96%, then both antioxidant activity was tested. Extract with higher antioxidant activity were made into oral suspension preparation for further evaluation of the stability (organoleptic, pH, microorganisms) and antioxidant testing. The results showed that ethanol extracts had higher antioxidant activity of methanol extract, but still lower than ascorbic acid. Based on the calculation, IC<sub>50 </sub>of methanol extract was 316.85 ppm and ethanol extract was 130.54 ppm, while ascorbic acid showed IC<sub>50</sub> 34.06 ppm. The most stable formula (F1a) shows that there is still an antioxidant activity that is equal to 42.11 %.

Silica gel column chromatography of the ethyl acetate fraction of methanol extract of Vincetoxicum stocksii resulted in the separation of three new rarely occurring natural products; [4-(4-(methoxycarbonyl)benzyl)phenyl] carbamic acid (1), bis[di-p-phenylmethane]ethyl carbamate (2), methyl 2-hydroxy-3-(2-hydroxy-5-(3-methylbut-2-enyl)phenyl)-2-(4-hydroxyphenyl) propanoate, stocksiloate(3), along with five known compounds; 1-(4-hydroxy-3-methoxyphenyl)-1,2,3,-propanetriol (4), feruloyl-6-O-β-D-glucopyranoside (5), 4-hydroxy-3,5-dimethoxybenzoic acid (6), apocynin (7) and vincetomine (8). The structures of compounds 1 and 2 were established with help of 1D, 2D-NMR techniques and high resolution mass spectrometry, whereas, compound 3 could only be characterized tentatively by 1D, 2D-NMR techniques. Compounds 1 is new compound while 2 is synthetically known but never been reported from natural source. The known compounds were identified due to 1D NMR analysis and in comparison with the literature values. Compounds 1-3 were found inactive in an anti-urease assay.

Background: Holothuria scabra is one of the most commercially important species found in the Pacific region. The sea cucumber extracts have been widely reported to have beneficial health effects. The aim of this study was to determine the nutritional compositions of H. scabra, and compare its important nutritional contents with that of other species.Methods: The sea cucumbers were dissected, sliced into small pieces, and then freeze-dried. The nutritional compositions, including proximate composition, amino acids, fatty acids, collagen, GABA, Vitamin A, C, and E of the whole body and body wall of H. scabra, were analyzed.Results: H. scabra contained a high quantity of protein (22.50% in whole body and 55.18% in body wall) and very low lipids (1.55% in whole body and 1.02% in body wall). The three most abundant amino acids found in both the whole body and body wall were glycine, glutamic acid, and proline. The main fatty acids found in the whole body were stearic acid and nervonic acid, and in the body wall were arachidonic acid and stearic acid. The whole body and body wall also contained high levels of essential amino acids, essential fatty acids, and collagen, in addition to moderate amounts of vitamin E and low amounts of GABA and vitamin C.Conclusions: The sea cucumber, H. scabra, contained high quantity of protein and very low lipid. It contained high essential amino acids, essential fatty acids, nervonic and arachidonic acids, and collagen, which also contained GABA, vitamin C, and vitamin E.Keywords: sea cucumber; Holothuria scabra; nutrition components; functional food

This research is conducted in the coastal waters of Bahoi Village, West Likupang Sub-district, North Minahasa District with the aims to know the types of sea-related environmental factors in the research site and know the community structure of the sea cucumber including the density, Index of diversity, dominance index, and distribution patterns. Data collection was done by using the line transect method, and sampling technique by using quadrat. The results of the study found 5 species of sea cucumber i.e. Holothuria atra,  Holothuria scabra,  Bohadschia marmorata, Actinopyga echinites, and  Bohadschia vitiensis. The individual density of all species is 0.12 ind/m2, and the diversity index (H ') 0.950. The value of the similarity index/compatibility obtained is e = 0.918. The result of the calculated dominance index of sea cucumber species is 1.0 and the pattern of spreading species of sea cucumber in the research site shows a random spread pattern because the value of dispersion index  (I) approaches 1 (one). Keywords: community; Sea cucumber; Bahoi; Dominance; Diversity.AbstrakPenelitian ini dilakukan di perairan pantai desa Bahoi, Kecamatan Likupang Barat, Kabupaten Minahasa Utara dengan tujuan untuk mengetahui jenis-jenis teripang yang ada di lokasi penelitian dan mengetahui struktur komunitas teripang yang meliputi kepadatan, indeks keanekaragaman, indeks dominasi, dan pola penyebaran. Pengambilan data dilakukan dengan metode line transek, dan teknik pengambilan sampel dengan mengunakan kuadrat. Hasil penelitian ditemukan 5 spesies teripang yaitu Holothuria atra, Holothuria scabra, Bohadschia marmorata, Actinopyga echinites dan Bohadschia vitiensis. Kepadatan individu dari seluruh spesies yaitu 0,12 ind/m2, indeks keanekaragaman (H’) 0,950. Nilai indeks kemerataan/keserasian diperoleh nilai indeks e = 0,918. nilai indeks dominasi spesies teripang diperoleh 1,0 dan pola penyebaran spesies teripang dilokasi penelitian menunjukkan pola sebaran acak (random) dikarenakan nilai indeks dispersi (I) mendekati 1 (satu).Kata Kunci : Komunitas; Teripang; Perairan Bahoi; Dominasi; Keanekaragaman
