<strong>Extraction and Oral Suspension Formulation of Sea Cucumber Holothuria scabra as Source of Antioxidants.</strong> Sea cucumbers have long been utilized in the food and medicine for Asia and Middle East communities. Indonesia is one of the main suppliers in the trade of sea cucumbers in Asia. Sea cucumber utilization in Indonesia is generally done only in the form of raw materials. Holothuria scabra is one of the sea cucumber that can be explored as a potential source of antioxidants. This study aims to determine the antioxidant activity of sea cucumber H.scabra in extract and suspension. Sea cucumber was extracted with methanol 96% and ethanol 96%, then both antioxidant activity was tested. Extract with higher antioxidant activity were made into oral suspension preparation for further evaluation of the stability (organoleptic, pH, microorganisms) and antioxidant testing. The results showed that ethanol extracts had higher antioxidant activity of methanol extract, but still lower than ascorbic acid. Based on the calculation, IC<sub>50 </sub>of methanol extract was 316.85 ppm and ethanol extract was 130.54 ppm, while ascorbic acid showed IC<sub>50</sub> 34.06 ppm. The most stable formula (F1a) shows that there is still an antioxidant activity that is equal to 42.11 %.

ABSTRACTUtilization of marine life is not just consumptive but can be used as raw material for medicines, one of which is sea cucumber. Sea cucumbers (H.atra) are marine invertebrates belonging to the Echinoderms phylum which have been studied containing antioxidants. Therefore, a study was conducted to determine the antioxidant activity of sea cucumber (H.atra) extract taken from the waters of Manado Bay using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. Sea cucumber samples were extracted using the maceration method. The ethanol extract of sea cucumber showed high antioxidant activity at a concentration of 8 μg / ml with 77.83% inhibition percentage. Keywords: H.atra, Antioxidant, DPPH (1,1-diphenyl-2-picrylhydrazyl).  ABSTRAKPemanfaatan biota laut bukan hanya sekedar konsumtif saja tapi dapat berpotensi sebagai bahan baku obat-obatan salah satunya teripang. Sampel teripang diekstraksi menggunakan metode maserasi. Holothuria scabra is one of high-value sea cucumber. The beneficial aspect of sea cucumbers is determined by the source of raw materials. The extraction process is a process that happens before sea cucumbers are used. In this process, the use of a various solvent is not only to facilitate the production process but also to maintain the active material. Also, the amount of yield obtained in the extraction process becomes a consideration in the production process. One of the benefits that are often claimed is antioxidants. This study aims to determine the yield of sea cucumbers after the extraction process by maceration with various types of solvents then antioxidant testing was carried out with DPPH followed by BSLT test. It was commonly used and easy to do. Besides, BSLT test was carried out to determine the initial toxicity and potency to be developed as antibacterial, antifungal and anticancer candidates. Results showed that the highest yield was 8.98% produced by dry sea cucumber samples extracted with ethanol PA 70% ethanol (code T5). In the results of the antioxidant tests, the best IC50 comes from dried sea cucumber extracted using ethyl acetate (code T2 and T3) with 1140 ± 0.00 ppm and1140 ± 10 ppm respectively. In the BSLT test results, the lowest LC50 was shown by extract using 100% methanol PA solvent (T4 code), with 108 ppm.

Plant natural products have a long history of being a rich source of neuroactive drugs.The total phenolic and flavonoid content, and antioxidant activity of ethanol, chloroform and aqueous extracts of AlgerianMedicagoSativaL, leaves were measured to discover new potential sources of Algerian natural antioxidants and neuroactives. All extracts were non-neurotoxic up to 1 mg/ ml for 24 h exposure. The total phenolcontent of M.Sativa extracts ranged between 38 and 69 mg GAE per g extracts. All of the extracts displayed antioxidant capacityassessed by two methods, but at different levels depending on the concentration and the extraction solvent used. The overall antioxidant potency was in the order water > chloroform> ethanol extracts, and the phenolic content exhibited a positive correlation with the antioxidant activity of M. Sativa extracts.Despite a weaker antioxidant activity, the ethanol extract elicited a calcium spike and suppressed the KCl-induced depolarisation response in neurons, indicating effect on ligand-and/or voltage gated calcium channels. In contrast, the chloroform and water extractsdisplayed no apparent direct functional effects. In conclusion, the antioxidant and functional properties related to phenolic content of Algerian Medicago Sativa L. in water and ethanol extracts, respectively.

ABSTRACTHabitat of sponge Stylissa sp., were under the sea and these sponges contain active compounds, which are more active than the compounds produced by terrestrial plants. Antioxidants are inhibitors of oxidation reactions due to the free radicals, which can cause weak damage to unsaturated cells, cell wall membranes, blood vessels, DNA bases, and lipid tissue, that causing disease. The study was conducted to determine the antioxidant activity of ethanol extracts  of sponge Stylissa sp., which is located on the Bangka Island, Likupang District, North Minahasa Regency. Sponge Stylissa sp., was extracted by maceration with using ethanol. Testing of antioxidant activity was carried out by the DPPH method measured by a UV-Vis spectrophotometer. The results of the study showed that ethanol extracts of sponge Stylissa sp., has antioxidant activity in each concentration and the highest at a concentration of 100 mg / L. The conclusion is a ethanol extract of sponge Stylissa sp. have high antioxidant activity with a concentration of 25 mg/L (77,40%), concentration of 50 mg/L (85,63%), concentration of 75 mg/L (88,66%), and concentration 100 mg/L (88,96%). Key words: Stylissa sp. Antioksidan adalah zat penghambat reaksi oksidasi akibat radikal bebas yang dapat menyebabkan kerusakan lemah tak jenuh, membran dinding sel, pembuluh darah, basa DNA, dan jaringan lipid sehingga menimbulkan penyakit. Penelitian dilakukan untuk mengetahui aktivitas antioksidan ekstrak etanol biota laut spons Stylissa sp. yang terdapat di pulau Bangka, Kecamatan Likupang, Kabupaten Minahasa Utara. Spons Stylissa sp. ini diekstrak dengan metode maserasi menggunakan etanol. Pengujian aktivitas antioksidan ini dilakukan dengan metode DPPH yang diukur dengan alat spektrofotometer UV-Vis. Hasil dari penelitian menunjukan bahwa ekstrak etanol spons Stylissa sp. memiliki aktivitas antioksidan disetiap konsentrasi dan yang tertinggi pada konsentrasi 100 mg/L. Kesimpulan yang didapat bahwa ekstrak etanol spons Stylissa sp. memiliki aktivitas antioksidan yang tinggi dengan konsentrasi 25 mg/L (77,40%), konsentrasi 50 mg/L (85,63%), konsentrasi 75 mg/L (88,66%), dan konsentrasi 100 mg/L (88,96%). Kata Kunci : Spons Stylissa sp., Antioksidan, DPPH (1,1-difenil-2-pikrilhidrazil)

ABSTRACTDayak onion plant (Eleutherine americana Merr.) or commonly called forest onion is a typical plant of Central Kalimantan that has been used by local peoples for cure a various types of diseases. This study aims to determine the activity of antioxidant compounds from Dayak bulb ethanol extracts from Kotamobagu City. This research method is an experimental laboratory. Initially the Dayak bulb sample was extracted maceration with 96% ethanol solvent then evaporated until a thick extract was obtained. As a parameter, the DPPH method (1,1-diphenyl-2-picrylhydrazyl) was used to test the antioxidant activity of the samples at concentrations of 50, 60, 70, 80 and 90 mg / L and as a comparison standard used Vitamin C with concentrations of 2, 4, 6, 8 and 10 mg / L. The results obtained showed antioxidant activity in Dayak bulb ethanol extracts with IC50 values of 41.46 mg / L and on vitamin C IC50 values of 1.04 mg / L. Based on these results it can be concluded that the onion bulb ethanol extract has very strong antioxidant activity. Keywords: Dayak Onion bulbs, Activity, Antioxidant, DPPH  ABSTRAK Tanaman bawang Dayak (Eleutherine americana Merr.) atau biasa disebut bawang hutan merupakan tanaman khas Kalimantan Tengah yang sudah dipergunakan masyarakat lokal sebagai obat berbagai jenis penyakit. Penelitian ini bertujuan untuk mengetahui aktivitas senyawa antioksidan dari ekstrak etanol bulbus bawang Dayak yang berasal dari Kota Kotamobagu. Metode penelitian ini adalah eksperimental laboratorium. Awalnya sampel bulbus bawang Dayak diekstraksi maserasi dengan pelarut etanol 96% kemudian dievaporasi hingga didapatkan ekstrak kental. Sebagai parameter, digunakan metode DPPH (1,1-diphenyl-2-picrylhydrazyl) untuk pengujian aktivitas antioksidan sampel pada konsentrasi 50, 60, 70, 80 dan 90 mg/L dan sebagai standar pembanding digunakan Vitamin C dengan konsentrasi 2, 4, 6, 8 dan 10 mg/L. Hasil penelitian yang didapat menunjukkan aktivitas antioksidan pada ekstrak etanol bulbus bawang Dayak dengan nilai IC50 sebesar 41,46 mg/L dan pada vitamin C nilai IC50 sebesar 1,04 mg/L. Berdasarkan hasil tersebut dapat disimpulkan bahwa ekstrak etanol bulbus bawang Dayak memiliki aktivitas antioksidan yang sangat kuat. Kata kunci: Bulbus Bawang Dayak, Aktivitas, Antioksidan, DPPH

ABSTRACT Sponges are a component of coral reef biota that has bioactive compounds with a greater percentage of activity compared to compounds produced by terrestrial plants. This study aims to determine the presence of antioxidant activity in the ethanol extracts of sponge Ianthella basta from the waters of Tumbak Village. Ianthella basta sponge was extracted using maceration method with 96% ethanol solvent. Antioxidant activity testing was carried out using the DPPH method with a concentration of 100 µg / mL, 75 μg / mL, 50 μg / mL, and 25 μg / mL and Vitamin C p.a as positive control. Each sample was made three repetitions of the test. Testing using a UV-Vis spectrophotometer. The results showed that the ethanol extract of the Ianthella basta sponge had antioxidant activity with a percentage of 48.73% at a concentration of 100 µg / mL. Keywords: Ianthella basta sponge, Antioxidants, Extraction, DPPH  ABSTRAK Spons merupakan salah satu komponen biota penyusun terumbu karang yang mempunyai senyawa bioaktif dengan persentase keaktifan lebih besar dibandingkan dengan senyawa-senyawa yang dihasilkan oleh tumbuhan darat. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antioksidan di dalam ekstrak etanol Spons Ianthella basta dari perairan Desa Tumbak. Spons Ianthella basta diekstraksi menggunakan metode maserasi dengan pelarut etanol 96%. Pengujian aktivitas antioksidan dilakukan dengan menggunakan metode DPPH dengan konsentrasi 100 µg/mL, 75 µg/mL, 50 µg/mL, dan 25 µg/mL dan Vitamin C p.a sebagai kontrol positif. Masing-masing sampel dibuat tiga kali pengulangan uji. Pengujian menggunakan alat spektrofotometer UV-Vis. Background: Binjai (Mangifera caesia) is a herb derived from South Kalimantan possessing antioxidant properties which promote wound healing inhibiting oxidation radicals. The natural antioxidants present in binjai leaves can be extracted by fractionation. Purpose: This study aimed to analyze the antioxidant activity of ethyl acetate fraction in 96% ethanol extract of binjai leaf. Methods: The study constituted a pure experimental study incorporating a post-test design with only random sampling technique consisting of two groups, namely; an ethyl acetate fraction as the treatment group and ascorbic acid as the positive control group. The leaves were treated in accordance with the soxhlet method and subsequently fractionated to extract ethyl acetate fraction. This was used to measure antioxidant activity with DPPH radical damping method using a UV-Vis spectrophotometer. A linear regression calculation was performed with a standard curve to quantify the IC50 value, before the ethyl acetate fraction underwent a qualitative test of secondary metabolite. Results: An independent t-test indicated significant differences between groups, an average value of IC50 in ascorbic acid of 13.812 ppm with 0.996 linearity and a fraction of ethyl acetate 38.526 ppm with a linearity of 0.999. In contrast, at this linearity value ascorbic acid and ethyl fraction acetate demonstrate a very high linear connection between concentration and inhibition. A secondary metabolite test conducted on the ethyl acetate fraction produced positive results for flavonoid, tannins, and phenol. Conclusion: Based on the IC50 parameters, the fraction of ethyl acetate in 96% ethanol extract of binjai leaf produces very strong antioxidant activity in the content of the compounds in the fraction, namely: flavonoid, tannins and phenol.

The effect of Cu2+ and Mg2+ metal ions on the antioxidant activity of anthocyanins of ethanol extract from the red dragon skin fruit (Hylocereus polyrhizus) has been done. Anthocyanin pigments from ethanol extract the red dragon skin fruit, content and antioxidant activity of anthocyanins from the red dragon fruit skin with ethanol solvent, and the effect of Cu2+ and Mg2+ metal ions on the antioxidant activity of anthocyanins. Identification of anthocyanin was done by identification of color test, using spectrophotometer analysis of UV-Vis and FTIR. The anthocyanin content was determined using pH difference method, and determined effect of metal ions extract was done by adding 50 ppm, 100 ppm, and 150 ppm of metal ions Cu2+ and Mg2+ into the anthocyanin extract that was been acidified by HCl , using spectrophotometer analysis of FTIR. Antioxidant activity test using DPPH method. The results show that ethanol extracts the red dragon anthocyanin by content is 12.5241 mg / L and antioxidant activity (IC50) is 0.478 μg / mL and with addition of2+ and Mg2+ metal ions can be increased antioxidant activity (IC50) of anthocyanins to be 0.2259 μg / mL for Cu2+ at concentration 50 ppm and 0.3351 μg / mL for Mg2+ at concentration 50 ppm.

AbstractAquacultivated sea cucumbers often suffer from SKin Ulceration Diseases (SKUDs). SKUDs have been observed in six holothuroid species from nine countries. All SKUDs present a similar symptom—the skin ulceration—and can be induced by bacteria, viruses, or abiotic factors. We here provide an update on SKUDs in holothuroids and analyse the case of the SKUD observed in Holothuria scabra in Madagascar. Field observations revealed a seasonality of the disease (i.e. wintertime maximum peak). Morphological analyses of integument ulcers showed that sea cucumbers react by forming a collagen fibre plug. Metagenomic analyses revealed a higher proportion of Vibrionaceae (Gammaproteobacteria) in ulcers in comparison to the healthy integument of the same individuals. Experimental infection assays were performed with ulcer crude extracts and bacteria isolated from these extracts (e.g. Vibrio parahaemolyticus) but did not significantly induce skin ulceration. Our results suggest that the disease is not induced by a pathogen or, at the very least, that the pathogen is not found within the ulcers as the disease is not transmissible by contact. An initial cause of the SKUD in Madagascar might be the repeated and prolonged exposures to cold temperatures. Opportunistic bacteria could settle in the dermis of ulcerated individuals and promote the ulcer extension. We propose a general nomenclature for SKUDs based on the acronym of the disease, the affected sea cucumber species (e.g. Hs for Holothuria scabra), the concerned region using an ISO code 3166-2 (e.g. MG for Madagascar), the description date (e.g. 20 for the year 2020), and, when known, the inducing agent (first letter of the general taxon, b for bacteria, v for virus in currently known cases; a a if it is an abiotic inducing parameter; nothing if the inducing cause has not been precisely identified). The disease described in this work will be designated under the name SKUD Hs-MG-20.

Purpose This paper aims to evaluate the antimicrobial and antioxidant activity of ethanol, methanol and aqueous extracts of the jujube fruit. Design/methodology/approach The fruit samples were harvested from Ziziphus lotus L. in three regions of Morocco: Aïn Chifae (Immouzer), Saiss (Fez) and Guercif. The fruit extracts were obtained by Soxhlet method using ethanol, methanol and water, and then a phytochemical screening was done for each extract. Total phenolic and total flavonoids contents were also determined. Afterward, the antimicrobial activity of the studied extracts was evaluated using the broth microdilution method. To estimate the total antioxidant effect of these extracts, the DPPH (2,2-diphenyl-1-picrylhydrazyl) scavenging and phosphomolybdate tests were performed. Findings Results showed that the highest content of polyphenols and flavonoids was found for ethanol and methanol extracts, while the lowest content was found in the aqueous extracts for all populations studied. Thus, results showed that the highest content of phenolic compounds was recorded in the population of Fez. Methanol extract of this population was the richest in polyphenols (30.36 mg/g) and in flavonoids (13.03 mg/g). The antimicrobial tests showed that Enterococcus faecalis and Pseudomonas aeruginosa were the most sensitive (6.25 mg/ml), compared to the other tested strains. Based on the minimal bactericidal concentrations/minimal fungicidal concentration or MFC/MIC ratio, it seems that ethanol extracts showed a bactericidal effect against Escherichia coli, Staphylococcus aureus and Bacillus subtillis, and a bacteriostatic effect against Enterococcus faecalis. On the other hand, the methanol extract showed bacteriostatic effect against Enterococcus faecalis and Pseudomonas aeruginosa. In addition, methanol extracts of Ziziphus lotus have significant antioxidant potent. Originality/value Methanol and ethanol extracts of Ziziphus lotus fruit have demonstrated strong antimicrobial effect. Moreover, the authors were able to show that the extract of Ziziphus lotus fruit has a very important antioxidant power.
