The basic moments of a process of obtaining apple tetraploids as donors of diploid gametes for apple breeding with polyploidy using are demonstrated. In industrial terms, triploids are of the greatest importance. The manifested effect of heterosis leads to the improvement of many characteristics - higher resistance to diseases, pests and adverse environmental conditions, greater autogamy than in diploids, less pronounced periodicity of fruiting, larger fruits and a convenient crown for harvesting. Triploids are developed as a result of crosses 2n × 3n or 2n × 4n. Tetraploids are necessary for more successful apple breeding with polyploidy using. For industry they are not of great importance but they are of interest as donors of diploid non-reduced gametes and allow to make the selection process more directional. One of the methods of experimental polyploidy is the induction of polyploids using mutagenes. The germs were treated with colchicines at concentrations 0.1% - 0.4% during 24 and 48 hours. According to the morphology the obtained plants were divided into five groups. Colchicine concentrations 0.3% and 0.4% during 48 hours of the treatment had a disastrous impact on the development of germs. As a result of the cytological analysis, tetraploids and chimeras were revealed, which were obtained from the seeds from the open pollination of Orlik and Svezhest (treatment variants – 0,1% colchicines solution and 24 and 48 hours of exposition), as well as from the seeds obtained as a result of the Svezhest × Bolotovskoye crossing. Tetraploids had a normal growth but they differed in large leaves, while chimeras were of low size with normal leaves and internodes.

Abstract A reciprocal chromosomal transposition was identified in several annual oilseed Brassica napus genotypes used as parents in crosses to biennial genotypes for genetic mapping studies. The transposition involved an exchange of interstitial homeologous regions on linkage groups N7 and N16, and its detection was made possible by the use of segregating populations of doubled haploid lines and codominant RFLP markers. RFLP probes detected pairs of homeologous loci on N7 and N16 for which the annual and biennial parents had identical alleles in regions expected to be homeologous. The existence of an interstitial reciprocal transposition was confirmed by cytological analysis of synaptonemal complexes of annual × biennial F1 hybrids. Although it included approximately one-third of the physical length of the N7 and N16 chromosomes, few recombination events within the region were recovered in the progenies of the hybrids. Significantly higher seed yields were associated with the parental configurations of the rearrangement in segregating progenies. These progenies contained complete complements of homeologous chromosomes from the diploid progenitors of B. napus, and thus their higher seed yields provide evidence for the selective advantage of allopolyploidy through the fixation of intergenomic heterozygosity.

The OBJECTIVE was to perform a comparative cytological analysis of the wound exudate in healing wounds during autodermoplasty on a granulating burn wound with and without vacuum therapy.METHODS AND MATERIALS. The article presents the results of the cytological study substantiated the use of vacuum therapy in combustiology. The comparative analysis was performed in 2 groups of patients. The first group consisted of patients with granulating burn wounds, who underwent surgical treatment with vacuum therapy. The second group consisted of patients who underwent surgical treatment without vacuum therapy. In both groups, the complex of surgical treatment included autodermoplasty on a granulating burn wound. All the patients were treated in the Burn Department of the «Scientific research institute – Ochapovsky regional clinic hospital № 1».RESULTS. The application of vacuum-associated dressings in the surgical treatment of patients in burn hospitals helps to reduce the time of graft retention in the plasty of granulating burn wound, decrease the number of dressings and the amount of dressing material, reduce the time of treatment of patients.CONCLUSION. The obtained cytological results allow us to justify the application of vacuum therapy in the surgical treatment of granulating burn wounds.

Both hexaploid bread wheat (AABBDD) (Triticum aestivum L.) and tetraploid durum wheat (AABB) (T. turgidum spp. durum) are highly significant global food crops. Crossing these two wheats with different ploidy levels results in pentaploid (AABBD) F1 lines. This study investigated the differences in the retention of D chromosomes between different hexaploid × tetraploid crosses in subsequent generations by using molecular and cytological techniques. Significant differences (P < 0.05) were observed in the retention of D chromosomes in the F2 generation depending on the parents of the original cross. One of the crosses, 2WE25 × 950329, retained at least one copy of each D chromosome in 48% of its F2 lines. For this cross, the retention or elimination of D chromosomes was determined through several subsequent self-fertilised generations. Cytological analysis indicated that D chromosomes were still being eliminated at the F5 generation, suggesting that in some hexaploid × tetraploid crosses, D chromosomes are unstable for many generations. This study provides information on the variation in D chromosome retention in different hexaploid × tetraploid wheat crosses and suggests efficient strategies for utilising D genome retention or elimination to improve bread and durum wheat, respectively.

The purpose of this research was to evaluate the growth phase of orchid Vanda hybrid (Vanda limbata Blume X Vanda tricolor Lindl. var. suavis) response with colchicines treatment, to determine precise effective concentration of colchicines and duration of treatment to induce polyploidy. This research used variation of colchicines concentration which were 0.01%; 0.05%; 0.1%; 0.5%; 1% and 0% and incubation for 6 hours; 12 hours; 18 hours; 24 hours; and  4 days. Treatment was done in aseptic condition using protocorm aged 7 week after sowing. After treatments, plants were planted on Vacint and Went (VW) medium supplemented with of 150 ml/l coconut water. After four months, they were subcultured in medium VW + 150 ml/l + 150g/l banana extract. The results showed that the orchid Vanda hybrid was intolerant to colchicine concentration of 0.1; 0.5 and 1% by death protocorm was more than 50%. Concentration of 0,5% with 6 hours soaking time is the most effective to induced tetraploid in orchid Vanda hybrid. Morphological characters of tetraploid plantlets had lower average number and length of roots, the number, length, and width of the leaves than those of control. Anatomical characters of the tetraploid plantlets revealed the highest stomatal size and had smaller of stomatal index than controls. This evident indicated a negative correlation between stomatal index and ploidy level. Cytological analysis with flow cytometry revealed that tetraploid plantlets (2n=4x=76) stained with DAPI showed the chromosome number more than those of control (2n=2x=38).Key words      : polyploidization, colchicine, vanda orchid, chromosome number
