Elaeocarpus ganitrus (Rudrakshya) is an evergreen tree found in tropical and sub-tropical regions and has been commonly used in traditional medicine against various diseases. In the present study, phytochemical constituents, antimicrobial activity and antioxidant property of seed of Elaeocarpus ganitrus have been evaluated in hexane, ethyl acetate, acetone and methanol extracts. The phytochemical screening showed the presence of alkaloids, flavonoids, phenols, tannins, steroids, carbohydrates, saponins, terpenoids etc. The antibacterial test showed a significant zone of inhibition against Escherichia coli, Salmonella typhi and Staphylococcus aureus with the maximum zone of inhibition of 20 mm. The study showed that these extracts can be used as antimicrobial substances against various bacteria. Antioxidant activity of the methanol, acetone and ethyl acetate extract were studied by DPPH radical scavenging assay. The extracts showed significant antioxidant activity. Among the IC50 values of different extracts on DPPH assay for antioxidant determination, methanol seed extract showed the lowest value of 28.09 µg/mL. This means methanol extract of seed showed the significantly high antioxidant properties. Calculation of total phenolic content showed a significant amount of phenolic content in different solvent extracts. Methanol seed extract showed the highest total phenolic content of 174.6 mgGAE/g. Similarly, methanol seed extract showed the highest total flavonoid content of 107.13 mgQE/g. These results suggest that the seeds of the Elaeocarpus ganitrus (Rudrakshya) plant from the Ilam district of Nepal possess high antimicrobial and antioxidant properties and thus, the plant can be used in the discovery of new bioactive natural products.

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

Abstract Cadaba farinosa is used in traditional medicine for treatment of cancer, diabetes, and rheumatism. The research was aimed at evaluating the phytochemical and antioxidant activity of the extracts. The powdered stem bark was extracted successively with the aid of Soxhlet extractor using n-hexane, ethyl acetate, and methanol. The resulting extracts were concentrated on rotary evaporator and the percentage yields were calculated. The phytochemical and TLC profiles of the extracts were studied. The antioxidant activity of the extracts and ascorbic acid (standard) were determined using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical assay. The total phenolic content of the extracts and tannic acid (standard) were evaluated using Folin–Ciocalteu reagent. The percentage yields of n-hexane, ethyl acetate, and methanol extracts were found to be 1.19, 1.37, and 13.93%, respectively. The phytochemical screening revealed the presence of tannins, flavonoids, saponins, cardiac glycosides, alkaloids, and triterpenoids. The TLC profiles of the extracts revealed the presence of compounds as evidenced from their R f values. The total phenolic content of ethyl acetate and methanol extracts were found to be 135 and 112 mg, respectively. The free radical scavenging activity demonstrated by the extracts was comparable to ascorbic acid. The ethyl acetate extract had higher phenolic content and demonstrated the highest free radical scavenging with IC50 31.07 mg/mL. The results of research have provided strong preliminary evidence of antioxidant activity which may be because of phenolic compounds in the extracts.

PENENTUAN TOTAL FENOLIK DAN AKTIVITAS ANTIOKSIDAN FRAKSI ETIL ASETAT DARI TEPUNG PELEPAH AREN (Arenga pinnata) ABSTRAK Tujuan penelitian ini untuk menentukan total fenolik dan aktivitas antioksidan fraksi etil asetat dari tepung pelepah aren (Arenga pinnata). Tepung pelepah aren diekstraksi maserasi menggunakan etanol hasil redestilasi cap tikus kemudian di partisi menggunakan pelarut petroleum eter, etil asetat dan aquades. Penentuan kandungan total fenolik menggunakan metode Folin-Ciocalteu dan aktivitas antioksidan menggunakan metode serapan radikal 1,1-difenil-2-pikrilhidrazil (DPPH). Didapati kandungan total fenolik fraksi etil asetat sebesar 172.857 ± 4.762 mg/kg dan aktivitas penangkal radikal bebas fraksi etil asetat sebesar 90.573 µg/mL. Hasil ini menunjukan bahwa fraksi etil asetat berpotensi sebagai antioksidan alami. Aren’s midrib powder extracted with maceration method with cap tikus redistillated ethanol and then partitioned with solvents such as petroleum ether, ethyl acetate and distillate water. The determination of phenolic content was using Folin-Ciocalteu method and the antioxidant activity was done using diphenylpycrylhydrazyl (DPPH) radical scavenging. Total phenolic value of ethyl acetate fraction is 172.857 ± 4.762 and IC50 value of ethyl acetate fraction is 90.573 µg/mL. This result shows ethyl acetate fraction as natural antioxidant potential. Penelitian ini bertujuan untuk menentukan aktivitas antioksidan dan anti glikasi ekstraks fenolik bebas dan ekstrak fenolik terikat dari tongkol jagung. Tongkol jagung diekstraksi secara sekuensial berbantu ultrasonikasi dengan pelarut etanol dan aseton selama 60 menit yang untuk mendapatkan ekstrak fenolik bebas. Residu dari ekstraksi fenolik bebas dihidrolisis dengan NaOH 2 M dan dinetralkan serta diekstraksi dengan etil asetat untuk mendapatkan ekstrak fenolik terikat. Ketiga ekstrak tersebut dilakukan analisis kandungan total fenolik dan pengujian aktivitas antioksidan dan anti glikasi. Kandungan total fenolik ekstrak  ekstrak fenolik terikat (EFT), ekstrak fenolik bebas aseton (EFBA) dan ekstrak fenolik bebas etanol (EFBE) berturut-turut adalah 288,39; 100,32 dan 92,95 μg/mL yang dinyatakan sebagai ekuivalen asam galat. Ekstrak EFT menunjukkan aktivitas penangkalan radikal bebas DPPH dan total antioksidan lebih tinggi daripada ekstrak EFBA dan ekstrak EFBE. Sejalan dengan itu, aktivitas anti glikasi dari EFT, EFBA dan EFBE berturut-turut adalah 61,93; 64,42 dan 66,31%. Hasil penelitian ini menyimpulkan bahwa ekstrak EFT dari tongkol jagung mengandung senyawa yang memilki sifat antioksidan dan berpotensi sebagai anti glikasi. ABSTRACTThis objective of this research was to determine the antioxidant activity and inhibition of AGEs formation of free phenolic and bound phenolic extract from corn cobs. Corn cobs were extracted sequentially assisted by ultrasonication with ethanol and acetone solvents for 60 minutes to obtain free phenolic extract. The residue from free phenolic extraction was hydrolyzed with 2 M NaOH and neutralized and extracted with ethyl acetate to obtain bound phenolic extract. The three extracts were analyzed for total phenolic content and evaluated for antioxidant activity and inhibitory activity of AGEs formation. The total phenolic content of bound phenolic extract (EFT), acetone-free phenolic extract (EFBA) and ethanol-free phenolic extract (EFBE) were 288,39; 100,32 and 92,95 μg/mL expressed as gallic acid equivalents, respectively. EFT extract showed higher DPPH free radical scavenging activity and total antioxidants more than EFBA extract and EFBE extract. Anti-glication of EFT, EFBA and EFBE were 61,93; 64,42 dan 66,31%, respectively. These result concluded that the EFT extract from corn cobs contains compounds having antioxidant properties and potential as anti-glication.

AbstractIn this study, the extraction conditions extracted maximize amounts of phenolic and bioactive compounds from the fruit extract of Ficus auriculata by using optimized response surface methodology. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS as well as reducing power assays on total phenolic content (TPC). For the extraction purpose, the ultrasonic assisted extraction technique was employed. A second-order polynomial model satisfactorily fitted to the experimental findings concerning antioxidant activity (R2 = 0.968, P < 0.0001) and total phenolic content (R2 = 0.961, P < 0.0001), indicating a significant correlation between the experimental and expected value. The highest DPPH radical scavenging activity was achieved 85.20 ± 0.96% at the optimum extraction parameters of 52.5% ethanol (v/v), 40.0 °C temperature, and 22 min extraction time. Alternatively, the highest yield of total phenolic content was found 31.65 ± 0.94 mg GAE/g DF at the optimum extraction conditions. From the LC–ESI–MS profiling of the optimized extract, 18 bioactive compounds were tentatively identified, which may regulate the antioxidant activity of fruits of F. auriculata.

Antioxidant and free radical scavenging activity of methanol, ethanol, ethyl acetate and chloroform extracts of aerial parts of Lamium purpureum L. was determined by DPPH, ABTS, FRAP and TRP assays. Contents of flavonoids and phenols were also investigated. The total phenolic content in the extracts, determined using Folin?Ciocalteu assay, ranged between 8.57 to 128.00 mg GAE/g d.e. while concentrations of flavonoids in the extracts varied from 24.20 to 39.80 mg QuE/g d.e. The highest phenolic content was found in methanol extract (128.00 mg GAE/g d.e.). The highest content of total flavonoids was identified in the methanol extract (39.80 mg QuE/g d.e.) and the lowest was in the chloroform (24.30 mg QuE/g d.e.). DPPH scavenging of the extracts was determined and obtained IC50 values ranged from 0.12 to 3.12 mg/mL of solution. The values of ABTS radical scavenging activity ranged from 0.35 to 1.80 mg AA/g. The highest ABTS antiradical activity was registered for methanol extract. The FRAP value was found within the range 0.08 to 1.04 ?mol Fe/mg. The best radical scavenger was methanol (1.04 ?mol Fe/mg). In reducing power assay different extracts of L. purpureum showed increasing of activity with increased concentration, and all extracts possessed substantial dose dependent antioxidant activity. The best reducing capacity was obtained with methanol extract of L. purpureum (0.0132 mg AA/mL). The results in this study confirmed that L. purpureum possesses moderate antioxidant properties.

This work aims to determine antioxidant, total phenolic content and Fourier transform infrared spectroscopy (FTIR) analysis of breadfruit leaves which are essential in management of diabetes. The methanolic extracts of breadfruit leaves was used to analyze for total phenolic content and antioxidant. Total phenolic content of the extracts was measured using the Folin–Ciocalteu assay while the antioxidant activity of plant extracts was measured by the 1, 1-diphenyl-2-picryhyradrazyl (DPPH) radical scavenging assay. FTIR analysis was used to determine the chemical components in the leaves. Total phenolic content in fresh breadfruit leaves (144.16 mg/g ± 17.98) was comparable to those of green tea. The results showed the breadfruit leaves extracts exhibited potent antioxidant activity. The presence of OH group also suggests antioxidant capacity of breadfruit leaves to deactivate free radicals as glucose itself could react with hydrogen peroxide in the presence of iron and copper ions to form hydroxyl radical.

The total phenolic content and antioxidant activity of 6 Salvia spp. exudates were measured to find new potential sources of natural antioxidants. Total phenolic content was assessed by a modified Prussian blue method, and the antioxidant activity by two methods: 1,1- diphenyl-2-picryl- hydrazyl (DPPH) radical scavenging capacity assay and lipoxygenase inhibitory assay. The total phenolic content ranged between 1.3 μg/mg DW ( S. fallax) and 74.0 μg/mg DW ( S. cacaliaefolia). In the DPPH test, S. cacaliaefolia was more effective than BHT, while in the inhibition of lipid peroxidation all the extracts presented good antioxidant capacity.

Exudates (DE) secreted from two shiitake mushroom mycelia (strains 1358 and L5458) were evaluated for their antioxidative properties and phenolic content. 1358DE and L5458DE showed distinct antioxidant activity in different in vitro assays, including scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, hydroxyl radical, superoxide anions and hydrogen peroxide; the ability to chelate ferrous ions; reducing power; hemolysis inhibition activity in rat erythrocyte; and lipid peroxidation inhibition (IC50 values of 1358DE and L5458DE were 3.3 and 132.6; 44.5 and > 1000; 26.9 and 53.7; 153.6 and >175.0; 176.0 and 521.0; 26.7 and 746.4; 47.8 and 736.9; and 3.1 and > 1000 μg/mL, respectively). Their total phenolic content was 237.33 and 24.08 mg gallic acid equivalent (GAE)/g of dry DE, respectively. Overall, these results show that 1358DE generally possesses better antioxidant properties than L5458DE, possibly due to its larger total phenolic content. Shiitake mushroom mycelial exudates, particularly of 1358DE, could be a good source of natural antioxidants.

Antioxidant properties of crude extract, partition extract, and fermented medium from Dendrobium sabin (DS) flower were investigated. The oven-dried DS flower was extracted using 100% methanol (w/v), 100% ethanol (w/v), and 100% water (w/v). The 100% methanolic crude extract showed the highest total phenolic content (40.33 ± mg GAE/g extract) and the best antioxidant properties as shown by DPPH, ABTS, and FRAP assays. A correlation relationship between antioxidant activity and total phenolic content showed that phenolic compounds were the dominant antioxidant components in this flower extract. The microbial fermentation on DS flower medium showed a potential in increasing the phenolic content and DPPH scavenging activity. The TPC of final fermented medium showed approximately 18% increment, while the DPPH of fermented medium increased significantly to approximately 80% at the end of the fermentation. Dendrobium sabin (DS) flower showed very good potential properties of antioxidant in crude extract and partition extract as well as better antioxidant activity in the flower fermented medium.
