In the present study, authors propagated Sophora tonkinensisGagnep plants using stem nodal culture. The results indicated that on MS medium supplemented with 30 g/l sucrose, 5.5 g/l agar, 200 ml/l coconut water, 1 g/l activated charcoal, 0.75 mg/l TDZ shoots proliferated from stem segments have the best count and height of shoots. The most appropriate medium for multiplication of shoots was the MS medium supplemented with 30 g/l sucrose, 5.5 g/l agar, 200 ml/l coconut water, 1 g/l activated charcoal, 0.75 mg/l TDZ, 0.5 mg/l IBA, 2.0 g/l peptone, 30 g/l carrot puree, pH 5.5 with the results of 20.60 shoots/explant, shoot height of 3.75 cm and 4.6 leaves/shoot after 8 weeks of culture. Root formation of shoots carried out on the MS medium supplemented with 30 g/l sucrose, 5.5 g/l agar, 200 ml/l coconut water, 1 g/l activated charcoal, 1.0 mg/l αNAA gave the best result. In the nursery, a mixture of humus + coconut fiber powder (70:30 ratio) was regarded as the best substrate due to the high survival rate of plantlets (92%) and healthy plantlets (10.30 cm high with 7.2 leaves and 4.3 new roots/a plantlet) at 10 weeks after planting

Alcantarea nahoumii (Leme) J. R. Grant is a species native to the Atlantic Forest that stands out for ornamental purposes. The objective of this work was to evaluate the in vitro germination of A. nahoumii seeds and establish a micropropagation protocol for production of seedlings so as to minimize the effects of predatory extractivism and develop an in vitro conservation system. Mature seeds were disinfested, established in three culture media (MS, MS½ and MS⅓) and incubated at four temperatures (20, 25, 30 and 35ºC) in a germination chamber. In the micropropagation experiment, stem segments were introduced in MS medium supplemented with 0.5 μM of 1-naphthaleneacetic acid (NAA) and 0.0, 2.2, 4.4 and 6.6 μM of 6-benzylaminopurine (BAP). For the in vitro conservation, plantlets were established in MS or MS½ medium supplemented with 15 g L-1 or 30 g L-1 of sucrose. The plants were acclimated with commercial substrate. The highest seed germination percentages were promoted by temperature conditions of 20 and 25ºC, with MS culture medium. The highest multiplication rate of shoots was obtained from the treatment without addition of the growth regulator or when combined with 2.2 μM of BAP + 0.5 μM of NAA. The acclimation of the plants occurred with high survival rate. The species can be conserved in vitro under slow growth condition for 24 months when incubated in MS medium supplemented with 30 g L-1 of sucrose.

Tissue culture is an alternative form of producing healthy, vigorous and regular plants on a large scale. The purpose of this study was to evaluate the most efficient culture medium for in vitro plantlet germination and development of three Orchidaceae species. Seeds disinfested of three species were dispersed in distilled water and dripped into basic Murashige and Skoog (MS) medium. The experimental design was completely randomized in a factorial 3 x 4 (three species x four culture media), with 5 replications. Four treatments were established: (1) full-strength MS medium, with the full nutrient concentration (MSØ), (2) full-strength MS medium plus 0.3% activated charcoal (MSØ ACh), (3) half- strength MS medium (½ MS) and (4) half- strength MS medium with 0.3 % activated charcoal (½ MS ACh). Germination was evaluated after 15, 20, 25, 30, and 60 days. The shoot height, leaf number and length, root number and length of plantlets of the three studied species were assessed. In A. variegata, 73% germinated after 60 days in ½ MS ACh medium. In the same period, 100% of E. viparum and S. gloriosa seeds germinated in MSØ ACh medium. The plant height, leaf number and length, root number and length were significantly higher for the species A. variegata and E. viviparum in MSØ ACh medium. The culture media ½ MS and MSØ with addition of activated charcoal favored in vitro germination for the three orchid species of this study.

Chrysanthemum indicum plant is a valuable and beneficial herb for human health. In this paper, we present the results of in vitro propagation of this plant in Da Lat. The results showed that MS medium supplemented with 25 g/l sucrose, 9 g/l agar, pH 5.8 was the best for shoot regeneration and growth (shoot height was 2.41 - 2.47 cm, 1 shoot/explant). MS media supplemented with BA (0.1, 0.5, 1, 1.5, 2 mg/l), Kinetin (0.1, 0.5, 1, 1.5, 2 mg/l ) and TDZ (0.1, 0.5, 1 mg/l) were unsuitable for shoot regeneration and growth of C. indicum. The regeneration and growth of shoots on the medium supplemented with 1 g/l of activated charcoal was better than (plant height of 3.45 cm) medium without activated charcoal (plant height of 2.46 cm). MS medium supplemented with 0, 0.1, 0.5, 1 mg/l IBA, 25 g/l sucrose, 9 g/l agar, pH 5,8 were suitable for in vitro root regeneration, rate of root formation was 100%. Coconut fiber powder was the best substrate to transfer the plantlets to the greenhouse, with survial rate of 100%, plantlets grew well. Nitrophoska irrigation was the best for the growth and development of C. indicum cultivated on coconut fiber powder under greenhouse conditions (plant height of 61.70 cm, 100.80 flowers/tree, flower diameter of 1.68 cm, fresh weight of 0.376 g/flower). The results also show that seedlings derived from tissue culture can be used to cultivate C. indicum and C. indicum grew well in the climate of Da Lat - Lam Dong and flowered all year round.

The experiment was conducted to investigate the combined effect of different plant growth regulators and charcoal supplementation in MS medium on growth and development of plantlets regenerated from protocorm like bodies (PLBs) of hybrid orchid. The combination of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA at different concentrations with charcoal supplementation was studied. The result revealed that the use of different growth regulators had significant effect on different parameters studied. The maximum weight of PLBs (5.123 g) was obtained from the combination of BAP + IBA at 1.0 mg/l each. The highest shoot height (3.239 cm) and maximum number of rooted plantlets (4.473) was obtained from 1.0 mg/l each of BAP + NAA combination. The maximum number of leaves (3.490) and the maximum length of leaves (1.946 cm) were obtained from 1.0 mg/l each of BAP + IBA and the highest leaf width (1.166 cm) was obtained from 0.5 mg/l BAP +1.0 mg/l IBA combination. The highest root length was obtained from 0.5 mg/l each of BAP + IAA and the maximum number of regenerated plantlets (20) was obtained from 0.5 mg/l IAA + 1.0 mg/l IBA combination. However, the maximum fresh weight of single shoot (0.219 g) and the maximum number of roots per plantlet (6.300) was obtained from 1.0 mg/I each of IAA + IBA combination. Heritability is a measure that describes the magnitude of a genetic influence on a character rather than an environmental factor. The higher the heritability value of a character, the higher the genetic variability. The aim of the study was to know the interaction between the combination of growth regulators and chrysanthemum varieties that gave the best chrysanthemum growth in vitro and analyzed the heritability of various chrysanthemum characters. The research was carried out at the In vitro Laboratory of Agronomy Department, Faculty of Agriculture, Hasanuddin University, Makassar from May to September 2016. The study used a separate plot design in the group. The main plot was a combination of growth regulator consisting of 200 ml Coconut Water, 0.5 ppm BAP + Coconut Water 100 ml, BAP 1 ppm + Coconut Water 100 ml, 2,4-D 0.5 ppm + Coconut Water 200 ml, 2,4-D 0.5 ppm + BAP 0.5 ppm + Coconut Water 100 ml, 2,4-D 0.5 ppm + BAP 1 ppm + Coconut Water 100 ml. The subplots included 8 varieties of chrysanthemum (Fiji Pink, Marimar, Pasopati, Salzieta, Solida Pelangi, Limeron, Arosuka Pelangi, Yellow Tumohon). The basic media used was MS medium. The results showed that adding 200 ml young coconut to the culture media gave the best in-vitro chrysanthemum growth in terms of shoot and root formation, shoot count, root number, and shoot height in all chrysanthemum varieties; as well as all observed characters have high heritability and genetic variability

An efficient protocol for propagation of Dendrobium aggregatum using the axenic immature seeds, derived from green capsule, was developed. The immature embryos from 120 days old capsules after pollination were germinated on Murashige and Skoog (MS) medium supplement with various concentration of BAP alone or in combination with NAA along with coconut water, and the same media were used for induction, multiplication, elongation and rooting in vitro shoots. MS medium with the addition of 3% sucrose 1.5 mg L-1 Benzyl amino purine (BAP) and 15% coconut water (CW) favoured the higher rate of germination, more number of protocorm like bodies, production of maximum number of shoots, elongation of shoots, as well as root formation. During acclimatization, 95% of the plantlets survived after one month.

Tribulus (Tribulus terrestrisL.) is a medicinal plant with considerable implementation such as aphrodisiac and anti‑inflammation drugs. This research was conducted to study the effectivity of coconut water and IAA (Indole‑3‑acetic acid) application on various concentration as the growth regulator for tribulus growth on thein vitroand its effect toward observation variables. The material used were cotyledon from tribulus embryo and MS (Murashige and Skoog) medium. The research method used a Completely Randomized Design with first factor of coconut water concentration of 0 mL L-1, 50 mL L-1, 100 mL L-1, and 150 mL L-1and IAA concentration of 0 ppm, 0.15 ppm, 0.20 ppm, and 0.25 ppm as the second factor. Observed variables were shoots emergence time, number of shoots, shoot height, number of leaves, roots emergence time, number of roots and regression between observation variables. The result showed that the combination of 150 mL L-1coconut water and 0.25 ppm IAA gave the highest shoot height, roots emergence time, and number of roots. Coconut water treatment on 150 mL L-1concentration alone gave the best result on shoots emergence time, number of shoots, and number of leaves, while IAA concentration of 0.25 ppm independently gave the highest number of leaves. Regression analysis result indicate that the number of roots has a positive correlation with shoots height, number of shoots and number of leaves.

Two cultivars of sweetpotato, `Commensal' and `Salyboro', were subjected to salt stress using axillary bud cultures. The salt levels ranged from 0 to 10 g·L-1. After the initial experiments, levels of calcium in the medium were changed from 3 mm in the MS medium to 1.5, 6, and 12 mm. After 10 weeks of growth, plantlet shoot height, dry weight, number of nodes, levels of proline, soluble carbohydrate, and protein were measured. `Commensal' was tolerant to salt levels up to 4 g·L-1, but `Salyboro' was sensitive to concentrations of salt even at lower concentrations as evidenced by the growth and dry weight. Proline accumulation was higher in the shoot than in the root. The protein: carbohydrate ratios did not change much in `Commensal', but levels of carbohydrates increased in `Salyboro'. Reduction in calcium levels had a synergistic affect on salt-stressed cultivars. Enhanced levels of calcium reduced the inhibitory affects of salt stress. This was more pronounced in `Salyboro', which was susceptible. Proline levels were higher in plants subjected to salt stress and higher levels of calcium than controls, but lower than the plants subjected to salt stress. These and other metabolic changes suggest that calcium can reduce the adverse affects of salt stress in these two sweetpotato cultivars.

An efficient protocol is outlined for rapid and mass micropropagation of Ficus carica L. (fig). Shoot tips (5 mm) were obtained from mother plants stock grown on half strength Murashige and Skoog (½ MS) medium with the addition of 30 g/L sucrose. For shoot multiplication Benzyl amino purine (BAP) and kinetin produced differences number of new shoot per plant and shoot height. BAP at 0.4 mg/L in combination with 0.2 mg/L indole-3-butyric-acid (IBA) produce maximum in vitro propagation rate, with 4.2 shoots per ex-plant. Root initiation was experimented on MS medium containing different concentrations of mg/L, IBA, IAA (Indole-3-acetic-acid) (IAA) or Naphthalene acetic acid (NAA). Highest number of root (4.3) was resulted when 1.5 mg/L IBA was used. After acclimatization in a mixture of (1 soil: 1 perlite: 1 peat) survival rate of 80% was achieved. For in vitro conservation of F. carica was experimented as microshoots were stored for 40 weeks on MS medium containing different sucrose concentration. Medium supplemented with 3% sucrose gave the highest regrowth (89%) at 24 ± 2 °C. Culture grew slowly when transferred to new fresh medium after the storage periods.

An efficient protocol was established for Sedum spectabile Boreau propagation. Various leaf parts were used as explants to regenerate plantlets, the stem segments of which were cultured for shoot proliferation and plantlet multiplication. The results showed that the leaf base was the optimal explant, as compared to both the middle and the top of leaves, for shoot formation. The highest shoot induction of 88.9% was observed on MS medium supplemented with 0.6 mg/l TDZ and 0.1 mg/l NAA. Hyperhydric leaves obtained in primary culture developed first into abnormal somatic embryos 10 days after subculture, and then into hyperhydric plantlets after an additional 10 days. The hyperhydric plantlets reversed to normal plantlets when plant growth regulators were removed from culture medium. Further, stem segments from reversed plantlets were used for shoot regeneration and root induction. Optimal shoot regeneration was obtained in MS medium containing 0.6 mg/l TDZ with 0.1 mg/l NAA. Root induction and root mean number were all higher on auxin-free medium than on medium containing auxins.
