Abstract Total phenolic content, total flavonoid content, antioxidant activity and individual phenolic compounds were assessed in full fat and defatted walnut kernel. For quantification of phenolic fraction of walnut kernels, two different solvents (methanol and ethanol) and two methods of extraction (ultrasonic-assisted extraction and Soxhlet extraction) were tested. Total phenolics, flavonoid content and antioxidant capacity of alcoholic extracts varied depending on the solvent used and extraction methods. Seventeen phenolic compounds were detected and the study provides evidence on high phenolic contents and high antioxidant potential of full fat walnut kernel and defatted walnut kernel. The Soxhlet extraction is the best in terms of the amounts of total phenolic content (2,089.2 mg gallic acid equivalent/100 g dry matter), while the ultrasonic assisted extraction is a fast method but resulted in significantly lower phenolic content (667.3–1,426.8 mg gallic acid equivalent /100 g dry matter). The concentrations of phenolics (especially (+)-catechin hydrate, juglone, vanillic acid, caffeic acid, ferulic acid, sinapic acid, salicylic acid and ellagic acid) are many fold lower in ultrasonic-assisted extraction as compare to the Soxhlet method using the same extraction solvent. The results of this study provide evidence on high phenolic contents and high antioxidant potential of full fat and defatted walnut kernel.

Hawthorn leaves are a rich source of phenolic compounds that possess beneficial activities for human health. Ultrasonic-assisted extraction (UAE) is an extraction technique frequently used for the isolation of phenolic compounds in plants. Thus, in this study, a Box–Behnken design was used to optimize UAE conditions such as the percentage of acetone, the extraction time and solvent-to-solid ratio (v/w) in order to obtain the maximum content of total compounds by Folin–Ciocalteu and the maximum in vitro antioxidant activity by DPPH, ABTS and FRAP assays in Crataegus monogyna leaves. The optimum conditions to obtain the highest total phenolic content and antioxidant activities were 50% acetone, 55 min and 1/1000 (w/v). A total of 30 phenolic compounds were identified and quantified in C. monogyna leaf extract obtained at these optimum UAE conditions. HPLC-MS allows the identification and quantification of 19 phenolic compounds and NP-HPLC-FLD analyses showed the presence of 11 proanthocyanidins. According to the results, the most concentrated phenolic compounds in C. monogyna leaf extract obtained at optimum UAE conditions were phenolic acid derivatives such as protocatechuic acid-glucoside, dihydroxy benzoic acid pentoside and chlorogenic acid, flavones such as 2″-O-rhamnosyl-C-hexosyl-apigenin, flavonols such as hyperoside and isoquercetin and proanthocyanidins such as monomer and dimer. As a result, the optimized UAE conditions could be used to obtain an extract of C. monogyna leaves enriched with phenolic compounds.

Green leafy vegetables are the most underexploited class of vegetables despite high nutritional value. The current study has been focused on the evaluation of anti-oxidant status of fresh and dehydrated under- utilized green leafy vegetable namely Mukia maderaspatana (L.) (Family: Cucurbitaceae), an indigenous plant; traditionally it is used as an ingredient of various cocktail preparations for the management of severe inflammatory disorders in Indian system of medicine. The total phenolic content (TPC), total flavonoid content (TFC) and total antioxidant activities were evaluated for the ethanolic extract of leaves to assess the in vitro antioxidant activities. The results showed that there exist a linear correlation between polyphenol content and antioxidant property. The ethanolic extract of dehydrated Mukia leaves showed the highest phenolic content (269.34 ± 0.78mg GAE/g), and total antioxidant activity (543±46 µmol Trolox/100 g). HPTLC analysis has revealed the presence of significant quantity of Quercitin (26.52%), an important flavonoid of tremendous antioxidant, anticancer and ant inflammatory properties in both the fresh and dehydrated leaves which might be the chief bioactive principle in Mukia.

The bioactive compounds, as well as the in vitro antioxidant activity of two Savoy cabbage cultivars, Dama and Leticia, grown in west of Spain under similar conditions were identified, quantified and compared. We found that cv. Dama presented in general betters results when compared with cv. Leticia. Cv. Dama presented higher concentrations of chlorophyll a (2.26 mg/100 g fresh weight), total phenolic content (102.71 mg of chlorogenic acid equivalent/100 g fresh weight) and total intact glucosinolates (195.22 µmol of sinigrin equivalent/100 g fresh weight). Thus, cv. Dama exhibited higher values of in vitro antioxidant activity.

This paper studies the optimization of ultrasonic-assisted extraction of polysaccharides from Oenothera biennis L. leaves using Box-Behnken design, and their in vitro antioxidant activity and structure. It is demonstrated that the optimum extraction conditions are: the ultrasonic power at 400 W, solvent-tosample ratio of 23:1 (mL/g), ultrasonication temperature at 53 °C, and time for 36 min, under which the yield of polysaccharides from Oenothera biennis L. leaves, namely 3.54%, remains close to the predicted value in the model. The assays of the scavenging effect on DPPH and hydroxyl free radicals, and of the reducing power indicate that polysaccharides from Oenothera biennis L. leaves possess strong antioxidant activity and certain relationship exists between their dose and effect in the concentration range of the experiment; Fourier IR spectroscopy reveals that characteristic absorption peak of polysaccharides can be seen from the sample; the DE-52 cellulose column chromatography, HPLC profiles, and GC-MS chromatogram identify the obtained OCP-2 as heteropolysaccharides, whose major components, including arabinose, mannose, galactose, and talose, have an average molecular weight of 5435 Da.
