Recently, the commercial breeding of Dactylopius coccus in confined environments has been developed not to depend on seasonal production and ensure the supply chain. Previous studies have shown the technical viability of their breeding of i Opuntia in a repository also called "Nopaloteca". However, considering the genetic diversity of Opuntia in each region, it is necessary to evaluate alternative cultivars to maximize the efficiency of that production system. The evaluated cultivars were Esmeralda and Villanueva (Opuntia ficus-indica (L.) Mill.). Four height levels (m) within the “Nopaloteca” (N1: 0.5; N2: 1.0; N3: 1.5 and N4: 2.0) and two harvest indices (pre and post oviposition) were evaluated regarding their effects on the carminic acid concentration (CAC, %) and fresh weight (FW, g) of D. Coccus. The results showed no significant difference in the CAC of the D. coccus colonies in both cultivars (18 %); however, the highest FW was reached in the cv. Esmeralda with 6.3 g per cladode (p ≤ 0.05). The highest CAC was found in the N4 treatment (2.0 m) with 18.6 % (p ≤ 0.05); while the highest average FW values (p ≤ 0.05) were in the intermediate levels of the N2 (1.0 m) and N3 (1.5 m) treatments, with 6.4 and 6.1 g per cladode, each. The post oviposition phase harvest of D. coccus resulted in a higher CAC concentration (20.4 %); meanwhile, in the pre-oviposition phase, 15.6 % was harvested (p ≤ 0.05). Due to the productivity and quality obtained, the Esmeralda cultivar could be used as an alternative host for the intensive breeding of D. coccus in confinement, in north-central Mexico.

Introduction: Extensive genetic diversity of malaria parasites is a major draw back to ongoing control efforts. Population-specific investigation of genetic structure of the parasite is important for effective malaria intervention in endemic populations such as Nigeria where about one-third of the global burden of the disease is borne. This study describes the genetic diversity of Plasmodium falciparum isolates in the Niger River basins, North-Central Nigeria. Methodology: Parasite DNA w as extracted fr om finger -prick blood samples collected from eighty P. falciparum positive individuals. Polymerase Chain Reaction (PCR) genotyping was carried out to target K1, MAD20 and R033 allelic families of Merozoite Surface Protein (MSP) -1 gene and FC27 and 3D7 allelic families of MSP-2 gene. Results: Proportion of isolates with K1 family w as 28(70%) with two alleles in Idah and 16(40%) with two alleles in Ibaji. Proportion of isolates with MAD20 family was 8 (20%) and a total of two alleles were observed in Idah and 4(10%) with two alleles in Ibaji. RO33 proportion was 16 (40%) in Idah one allele and 8(20%) in Ibaji where the allelic family was also observed to be monomorphic. K1 was the most predominant MSP1 allele in the two parasite populations and the frequency of FC27 genotype was higher than 3D7 in both populations. Multiplicity of infection (Mol) with MSP-1 loci was higher in Ibaji (1.30) than Idah (1.05) while MoI with MSP-2 loci was lower in Ibaji (2.00) than Idah (2.13). However, there is no significant difference in the mean Mol between Idah and Ibaji (P > 0.05). The expected heterozygosity (HE) value was 0.56 for MSP-1 and 0.84 for MSP-2. Conclusion: Our findings revealed high levels of monoclonal infections with P. falciparum, suggesting low parasite diversity. This may be a pointer to a reduction in malaria transmission in the river basins.

AbstractCysticercus tenuicollis as metacestode of Taenia hydatigena is the most prevalent taeniid species in livestock. Eighty-eight C. tenuicollis samples were collected from sheep (n = 44) and goats (n = 44) of the northern Iran from 2015 to 2016. The isolated parasites were characterized by morphometric keys. The DNA of the larval stage was extracted, amplified and sequenced targeting mitochondrial 12S rRNA and Cox 1 markers. A significant difference in larval rostellar hook length was observed in 12S rRNA haplotypes. Analysis of molecular variance of 12S rRNA indicated a moderate genetic diversity in the C. tenuicollis isolates. The pairwise sequence distance of C. tenuicollis showed an intra-species diversity of 0.3–0.5% and identity of 99.5–100%. Using the 12S rRNA sequence data we found a moderate genetic difference (Fst; 0.05421) in C. tenucollis isolates collected from livestock of the northern and southeastern regions of Iran. We concluded that the genetic variants of C. tenuicollis are being undoubtedly distributing mostly in different parts of Iran. Further studies with a larger number of T. hydatigena isolates collected from various intermediate and definitive hosts are needed to study this evolutionary assumption and also to determine the apparent genetic differences observed in the studied regions.

The ascorbic acid (vitamin C) content of fresh imported field tomatoes and Nova Scotia greenhouse and field tomatoes was determined on a bi-weekly basis during the period of availability of each type of tomato to the Nova Scotia consumer in 1984. The average ascorbic acid contents of imported and Nova Scotia field and greenhouse tomatoes were 13.3, 16.7 and 17.7 mg 100 g−1 fresh weight, respectively. A study of nine recommended or promising field tomatoes for the Atlantic region showed significant differences in ascorbic acid content among the cultivars. The cultivar Quick Pick had the highest ascorbic acid content of 22.5 ± 1.5 mg 100 g−1, the cultivar Campbell 18 had the lowest content, 12.0 ± 2.9 mg 100 g−1. In Dombito greenhouse tomatoes the stage of maturity and the effect of cluster location were tested against ascorbic acid content. The lowest ascorbic acid content of 9.1 ± 1.0 mg 100 g−1 was found with the small green tomatoes while others from mature green to overripe contained 14.0–16.7 mg 100 g−1. Tomatoes from different cluster locations showed no significant difference in ascorbic acid content.Key words: Vitamin C, L-ascorbic acid, tomatoes

Environmental and infection variables may affect the genetic diversity of baculovirus populations. In this study, Helicoverpa armigera nucleopolyhedrovirus (HearNPV) was used as a model system for studying the effects of a key infection variable, inoculum dose, on the genetic diversity within nucleopolyhedrovirus populations. Diversity and equitability indices were calculated from DNA polymerase-specific denaturing gradient gel electrophoresis profiles obtained from individual H. armigera neonate larvae inoculated with either an LD5 or LD95 of HearNPV. Although the genetic diversity detected in larvae treated with an LD95 was not statistically different from the diversity detected in the HearNPV inoculum samples, there was a statistically significant difference in the genetic diversity detected in the LD5-inoculated larvae compared with the genetic diversity detected in the HearNPV samples used for the inoculations. The study suggests that inoculum dose needs to be considered carefully in experiments that evaluate HearNPV genetic diversity or in studies where differences in genetic diversity may have phenotypic consequences.

Pepper is the third most important solanaceous crop in the United States and fourth most important worldwide. To identify sources of resistance for commercial breeding, 170 pepper genotypes from five continents and 45 countries were evaluated for Phytophthora fruit rot resistance using two isolates of Phytophthora capsici. Genetic diversity and population structure were assessed on a subset of 157 genotypes using 23 polymorphic simple sequence repeats. Partial resistance and isolate-specific interactions were identified in the population at both 3 and 5 days postinoculation (dpi). Plant introductions (PIs) 640833 and 566811 were the most resistant lines evaluated at 5 dpi to isolates 12889 and OP97, with mean lesion areas less than Criollo de Morelos. Genetic diversity was moderate (0.44) in the population. The program STRUCTURE inferred four genetic clusters with moderate to very great differentiation among clusters. Most lines evaluated were susceptible or moderately susceptible at 5 dpi, and no lines evaluated were completely resistant to Phytophthora fruit rot. Significant population structure was detected when pepper varieties were grouped by predefined categories of disease resistance, continent, and country of origin. Moderately resistant or resistant PIs to both isolates of P. capsici at 5 dpi were in genetic clusters one and two.

Leucaena leucocephala leaves are nutritious and palatable for ruminants. However, the plant is poorly adapted on acid soil, water logged, and susceptible to psyllid (Heteropsylla cubana). Leucaena favours soils with pH >5. It performs best on calcareous soils. This study aimed to evaluate adaptability of several Leucaena species on acid soils. The study was conducted at the experimental field in Ciawi, Bogor, altitude 500 m above sea level, rainfall 1500-2000 mm year-1, on Latosol soil with pH 5.2. Leucaena species evaluated were L. leucocephala K28, L. leucocephala (local type), Leucaena KX2 hybrid, L. collinsii, L. trichandra, and L. diversifolia. Experiment was designed in a randomized complete block, four replications. All the seedlings were grown in the nursery for 2.5 months before being transplanted to the field. The plants were grown in row plots of 5 m x 1 m with planting distance 0.5 m and spacing between row plot was 3 m. One unit replicate was 5 m x 21 m. In wet season, the plants were pruned at 50 cm height every 3 months, whilst in dry season pruning was done every 5 months. Parameter measured were plant regrowth and forage production (edible part and non-edible part of the plant), as well as N and P content of the edible part. The result showed that Leucaena KX2 hybrid was superior to other leucaenas. Regrowth of KX2 hybrid was the fastest and produced highest forage. The highest fresh weight forage production for 1 year (four times harvests), i.e. 33.93 t ha-1 year-1 equal to 12.48 t ha- 1 year-1 of dry weight, was obtained from Leucaena KX2 hybrid, whereas the lowest was produced by L. diversifolia, i.e. 3.12 t ha-1 year-1 of fresh weight or 1.01 t ha-1 year-1 of dry weight. Production of non-edible part followed the pattern of production of edible part. N and P content in the edible part of all Leucaena species did not show any significant difference except for L. trichandra which contained the lowest. This study implies that Leucaena KX2 hybrid is adaptable to acid soil; therefore it can be grown in wider areas throughout Indonesia.

The field experiments with thirty genotypes were conducted during June to October month of kharif, 2018 and kharif, 2019, to assess extent of variability under aerobic condition. The genotypes were sown under dry direct seeded condition using randomized block design (RBD) with three replications. Each genotype was sown in single row of 5 m length with spacing of 20 cm between rows and 15 cm between plants. Data recorded for 22 characters including different morphological and quality traits from 5 randomly selected plants of each replication and mean data used for analysis. ANOVA revealed that the mean sum of squares were highly significant difference for most of the traits. The value of PCV was higher than GCV for all the twenty-two characters. However, maximum GCV and PCV were observed for root dry weight plant-1 (31.44% and 32.17%) followed grain yield plant-1 (29.97% and 31.03%), root volume (28.62% and 29.20%), root fresh weight plant-1 (28.51% and 29.08%), biological yield plant-1 (21.86% 22.50%) and number of grains panicle-1 (20.55% and 21.37%). Rest of the traits showed moderate and low GCV and PCV. High heritability and genetic advance were recorded for the traits viz., leaf length, number of tillers plant-1, number of grains panicle-1, 1000 seed weight, root length, root volume, root fresh weight plant-1, root dry weight plant-1, kernel length-breadth ratio, grain yield plant-1, biological yield plant-1 and harvest index. The information regarding different variability will provide direction to select high yielding genotypes under aerobic condition.
