Explants of nodal and root segments of bitter gourd were cultured on MS supplemented with various concentrations of BAP in combination with either 2,4-D or NAA. Nodal segments produced the highest percentage (93.75) of callus in MS supplemented with 1.0 mg/l 2,4-D and 1.0 mg/l BAP whereas, root segments produced the highest (85.00%) callus in 0.6 mg/l NAA and 2.5 mg/l BAP combination. A combination of 1.0 mg/l 2,4-D and 1.0 mg/l BAP exhibited 75.00% shoot regeneration from nodal segments. The highest shoot length (5.15 cm) was recorded with 2.5 mg/l BAP and 0.2 mg/l IAA from nodal segments. No sign of regeneration of shoot was found from root segments in any of the combinations. Indirect regeneration of plant was obtained through organogenesis in leaf callus cultures of Ionidium suffruticosum. Leaf explants were found to be best suited for callus induction on MS with 2, 4-D (0.5 and 1.0 mg/l). Maximum shoot regeneration was obtained in MS supplemented with Kn (4.0 mg/l) alone and NAA (0.4 mg/l) with Kn (2.0 m/l).  The in vitro shoots thus obtained were successfully rooted in MS supplemented with Kn (4.0 mg/l) alone and with NAA (2.0 mg/l) and Kn (0.2  mg/l).  Seventy per cent of the rooted plants survived and they were successfully acclimated in soil. Key words: Ionidium suffruticosum, micropropagation, Medicinal herb D.O.I. 10.3329/ptcb.v19i2.5431 Plant Tissue Cult. & Biotech. 19(2): 143-150, 2009 (December)

Nodal explants of 12 accessions from four species of yam (Dioscorea spp.) were cultured for six weeks on MS to evaluate the influence of IAA, Kn, NAA and BAP on the production of leaves and microtubers. Four Dioscorea polystachya Turcz., three each of D. bulbifera L. and D. sansibarensis Pax. and two D. japonica Thunb. accessions were used. Five and 10 mg/l of Kn along with IAA and sucrose, and 0.2 and 0.5 mg/l of NAA, sucrose and with or without BAP were used in four treatments. The accessions Yam 23 and Yam 25 of D. sansibarensis failed to initiate any leaf under four treatments. The remaining accessions produced 0.11 to 1.76 leaves per explant. The medium containing IAA with higher concentration of Kn (10 mg/l) and 3% sucrose was found to be best for in vitro production of leaf (0.71/explant) and the most productive species was D. japonica (1.36), followed by D. polystachya (1.19/explant). At the same culture period, Yam 16 of D. bulbifera failed to initiate any microtuber at IAA with Kn, and NAA with or without BAP. The remaining accessions produced 0.09 to 1.15 microtubers per explant. Lower concentration of Kn (5 mg/l) with IAA and sucrose was favourable for producing microtubers (0.61/explant on an average), the best species being D. sansibarensis (1.27) followed by D. japonica (0.59/ explant). Shoot tip explants from in vitro germinated seedlings of Solanum capsicoides All. inoculated on MS containing 2 mg/l BA produced maximum shoot induction response (26 shoots per explant). Rooting of the microshoots (19.4 roots per explant) was obtained better in half strength of MS supplemented with NAA (0.5 mg/l). Effects of different concentrations of 2-ip and IBA in WPM basal medium for Blomidon blueberry in vitro propagation and four different rooting agents at the early stage after transplantation showed that 15 mg/l of 2-ip is the best concentration to induce shoots. In Costa Rica Albizia guachapele, Cedrela odorata, Platymiscium pinnatum and Guaiacum sanctum are important plant species in both economic and ecological terms and their wood is precious and reported to be highly resistant material. This research has evaluated the in vitro micropropagation as a technology focused to conserve these species. Maximum shoot regeneration (87.85%) and number of shoots per explant in the epicotyl segments having longitudinal cut in Citrus reshni var. Cleopatra were obtained on MT medium supplemented with BAP (2 mg/l) + NAA (0.2 mg/l). Maximum shoot length (5.10 cm), average leaf number per shoot (12), leaf length (2.12 cm) and width (1.5 cm) in Cleopatra were obtained on MT medium supplemented with BAP (2 mg/l) + NAA (0.2 mg/l). The maximum in vitro rooting (78.88 %) in the excised shoots occurred in liquid medium on MS supplemented with BAP (0.5 mg/l) + NAA (0.5 mg/l). Plant tissue culture techniques have been used to evaluate the effects of many different substances and/ or conditions in plant growth and development. It provides information of great value about problems related to basic and applied aspects of plant as well as contributed to understanding of factors responsible for growth, metabolism, synthesis of secondary compounds, stress response. Considering all this wide range of applications and as all plant tissue culture techniques are undergone under axenic and controlled conditions (culture medium composition, light and temperature, for instance), it seems to be a value model for Ultra High Diluted (UHD) studies. Lippia alba is a Brazilian plant that tissue cultures protocols and in vitro essential oil production have already been described in scientific literature. None of all scientific papers evaluated the effects of UHD substances on in vitro development or secondary metabolic production. The main goal was to evaluate the use of plant tissue culture to investigate the effects of UHD benzilaminopurine (BA) on Lippia alba shoot culture. Nodal segments obtained from plants growth in vitro was subcultured to Murashigue & Skoog semi-solid medium added with 2ml of these different solutions: BA 3µmol, BA 12CH (10-24), water 12CH and water (no dilution and succussion). Weekly 1 ml of solutions were added to cultures. The experiment was repeated twice and each one consisted in 3 culture vessel with 5 nodal segments per treatment (n=30). All plants were maintained in growth room under controlled temperature (25°C), light and photoperiod (16L/8D). The tested substances were prepared according to the method of stepwise dilution and succussion as describe in Brazilian Homeopathic Pharmacopoeia. The experiment was blinded all the time. After 60d, plantlets were evaluated for number of shoots, shoot length, rooted plants (%), callus development (%) and fresh biomass. Data were submitted to ANOVA following by Duncan’s and t-test. Plants from water 12CH and BA 12CH increased the number of new shoots and promoted the highest shoot length. By adding BA 3µmol the organogenetic response was inhibited since neither shoot nor root were developed. However, it was observed a significant basal callus development. Plant tissue culture could be adapted for UHD studies. More studies are being conducted in way to analyze other experimental conditions and biochemical/phytochemical parameters.

The aim of this study was to identify the primary materials for in vitro propagation of the Petunia hybrida L. The sterilisation method and the medium in different stages: shoot regeneration, rapid shoot multiplication, and the part of the in vitro plant used for rapid multiplication were identified. The study results showed that using nodal segments with axillary buds as the primary material and sterilising by HgCl2 0.1% in 10 minutes gave the best disinfection effect (survival rate reached 60%). Murashige and Skoog (MS) medium supplemented with 1 mg of BAP/l + 6.5 g agar/l + 30 g sacarose/l was reported as the best one for shoot regeneration (100% of shoot formed and number shoot per sample was 2.73), meanwhile, the mixture of MS + 1 mgBA/l + 0.2 mg IBA/l + 6.5 g agar/l + 30 g sacarose/l was considered as the best medium for rapid shoot multiplication (average shoots/sample reached 19.53). In vitro,leaf tissue is the most suitable vegetative organ for rapid multiplication, 100% of in vitro shoots regenerate through the form of callus, and the ratio of shoots/sample reached 21.53.
