Research relevance: fatty liver disease is one of the common worldwide disorders and is a public health problem, with obesity and other metabolic disorders. Research methods and materials: article is based on the publications review concerning fatty liver degeneration in cardiovascular diseases. Research objectives: to identify the course, etiology, and clinical picture of metabolic and immune changes in fatty liver degeneration. Research results: pathogenic factors associated with fatty liver disease are multifactorial and include inflammation, adipokines, intestinal dysbiosis, oxidative stress, which are established signs of cardiovascular disease. Conclusions: The accumulation of fat in the liver may be associated with ectopic adipose tissue, including myocardial fat and adipose tissue surrounding the heart, which is a central aspect.

AbstractNon-alcoholic fatty liver disease (NAFLD) is an increasingly prevalent condition that has been linked to high-fructose corn syrup consumption with induction of hepatic de novo lipogenesis (DNL) as the suggested central mechanism. Feeding diets very high in fructose (> 60%) rapidly induce several features of NAFLD in rodents, but similar diets have not yet been applied in larger animals, such as pigs. With the aim to develop a large animal NAFLD model, we analysed the effects of feeding a high-fructose (HF, 60% w/w) diet for four weeks to castrated male Danish Landrace-York-Duroc pigs. HF feeding upregulated expression of hepatic DNL proteins, but levels were low compared with adipose tissue. No steatosis or hepatocellular ballooning was seen on histopathological examination, and plasma levels of transaminases were similar between groups. Inflammatory infiltrates and the amount of connective tissue was slightly elevated in liver sections from fructose-fed pigs, which was corroborated by up-regulation of macrophage marker expression in liver homogenates. Supported by RNA-profiling, quantitative protein analysis, histopathological examination, and biochemistry, our data suggest that pigs, contrary to rodents and humans, are protected against fructose-induced steatosis by relying on adipose tissue rather than liver for DNL.

AbstractNonalcoholic fatty liver disease (NAFLD) bears serious economic consequences for the health care system worldwide and Ukraine, in particular. Cardiovascular diseases (CVD) are the main cause of mortality in NAFLD patients. Changes in the gut microbiota composition can be regarded as a potential mechanism of CVD in NAFLD patients.The purpose of this work was to investigate changes in major gut microbiota phylotypes, Bacteroidetes, Firmicutes and Actinobacteria with quantification of Firmicutes/Bacteroidetes in NAFLD patients with concomitant CVD.The author enrolled 120 NAFLD subjects (25 with concomitant arterial hypertension (AH) and 24 with coronary artery disease (CAD)). The gut microbiota composition was assessed by qPCR.Resultsthe author found a marked tendency towards an increase in the concentration of Bacteroidetes (by 37.11% and 21.30%, respectively) with a decrease in Firmicutes (by 7.38% and 7.77%, respectively) in both groups with comorbid CAD and AH with the identified changes not reaching a statistical significance. The author quantified a statistically significant decrease in the concentration of Actinobacteria in patients with NAFLD with concomitant CAD at 41.37% (p<0.05) as compared with those with an isolated NAFLD. In patients with concomitant AH, the content of Actinobacteria dropped by 12.35%, which was statistically insignificant.Conclusionsthe author established changes in the intestinal microbiota, namely decrease in Actinobacteria in patients with CAD, which requires further research.
