The crossbreeding of Swamp and River type buffalo breeds is practiced for the improvement of milk yield and reproductive performance in swamp buffalo herds. This study aimed to modify the Ovsynch synchronization protocol (GPG) and improve the fixed-timed artificial insemination (FTAI) for better reproductive performance of crossbred buffaloes. Comparison of four conventional synchronization protocols [pregnant mare gonadotropin-prostaglandin F2α-gonadotropin-releasing hormone (PmPG), gonadotropin-releasing hormone-prostaglandin F2α-gonadotropin-releasing hormone (GPG), prostaglandin F2α-gonadotropin-releasing hormone-prostaglandin F2α-estradiol benzoate (PGPE), and progesterone-pregnant mare gonadotropin-prostaglandin F2α-gonadotropin-releasing hormone (P4PmPG)] in crossbred buffaloes showed that the GPG protocol treated buffaloes displayed higher (P < 0.05) estrus response with an increasing tendency in ovulation (84.6%) and pregnancy rates (30.8%) than PmPG, PGPE, and P4PmPG treated buffaloes. Buffaloes treated with a dose of 0.4 (mg/kg) mifepristone combined with GPG, exhibited higher (P < 0.05) estrous response (82.4%), ovulation (94.1%), and pregnancy (47.1%) rates compared with other doses (0, 0.3, or 0.5 mg/kg) groups. Injection of mifepristone along second GnRH injection in buffaloes improved (P < 0.05) pregnancy rate (35.3%) when compared to before or after the second GnRH of GPG protocol. Single AI after 24 h of mifepristone or second GnRH injection seems the best time to enhance the pregnancy rates in buffaloes compared to double or other single AI times in the modified GPGMH protocol. In comparison, GPGMH reduced the follicular cyst incidence (P < 0.05) with increasing ovulation (P > 0.05) and pregnancy rates (P > 0.05) than the P4GPG and GPG protocols in crossbred buffaloes. The current study supported that new synchronization protocol (modified of GPG protocol; GPGMH) by the inclusion of mifepristone (with a dose of 0.4 mg/kg along second GnRH), AI after 24 h of mifepristone or second GnRH, and human chorionic gonadotropin (hCG at day 5 of AI) enhance the ovulation and pregnancy rates in crossbred buffaloes.

Timed AI (TAI) programs have been developed to increase the service risk. However, when Ovsynch is used as a synchronization protocol, lack of regression of the corpus luteum after prostaglandin F2α (PGF) administration has been demonstrated, leading to reductions in fertility. In this way, a second PGF administration might increase the frequency of achieving complete luteolysis. This study aimed to evaluate the effect of an additional treatment with PGF on fertility when an Ovsynch synchronization protocol was applied at different postpartum intervals. Multiparous high-production dairy cows (n=471) of a commercial farm were stratified by postpartum intervals [<120 (n=136), 120-150 (n=131), 150-180 (n=92), 180-210 (n=52), and >210 days (n=60)] and randomly allocated to receive either the Ovsynch (n=108) or Ovsynch with second PGF protocol (n=243). Cows inseminated 12h after observed heat detection (AI) served as a control (n=120). The Ovsynch protocol consisted of an injection of gonadotropin-releasing hormone (100 µg; Fertagyl®, Intervet, Millsboro, MD, USA) on Day 0, an injection of PGF (0.5 mg; Estrumate®, Schering-Plough Animal Health, Montréal, QC, Canada) on Day 7, another injection of gonadotropin-releasing hormone (100µg) on Day 9, and timed insemination on Day 10. The Ovsynch with second PGF protocol consisted of the same hormone injection schedule as the Ovsynch protocol, but a second injection of PGF (0.5 mg; Estrumate®) was administered 24h after the first PGF. Data were analysed using the chi-square test. The level of significance was taken as P<0.05. When data were pooled at 120 to 150 days of postpartum interval, pregnancy rate was higher, but not significantly different (P>0.05), for cows that received the Ovsynch (37.5%, n=32) or Ovsynch with second PGF protocol (33.8%, n=77) compared to nonsynchronized cows (22.8%, n=22). After 180 days of postpartum interval, Ovsynch with second PGF protocol triggered pregnancy rates similar to those of nonsynchronized cows (33.3%, n=21 and 28.6%, n=14), whereas the Ovsynch protocol resulted in lower percentages (11.8%, n=17; P>0.05). Similar pregnancy rates (P>0.05) were observed when TAI protocols were administered before 120 days (26.5 and 30.2% for Ovsynch and Ovsynch with second PGF, respectively) or between 150 and 180 postpartum days (31.8 and 33.9% for Ovsynch and Ovsynch with second PGF, respectively) compared with AI cows (25.7 and 28.8% for <120 days and between 150 and 180 postpartum days, respectively). These preliminary results showed a tendency of a better pregnancy rate when both TAI synchronization protocols were applied between 120 and 150 postpartum days when compared with AI. Also, the application of a second dose of PGF seems recommendable when the Ovsynch protocol is used to synchronize cows of >180 postpartum days. Ongoing research will allow an increase in the number of the samples in each postpartum interval in order to confirm these results.

A comparative hormonal study of prostaglandin, gonadotropin releasing hormone and their combination was conducted on 30 randomly selected cross-bred dairy cows of SEMEX project for artificial insemination in Hillat Kuku, Khartoum North, Sudan. The study attempts to determine the response of the animals to hormonal induction and synchronization of estrous as well as fertility following fixed time artificial insemination. Three treatment groups of 10 cows each were undertaken. Groups 1, 2 and 3 involved administration of prostaglandin F2α (PGF2α), gonadotropin releasing hormone (GnRH) and a combination of PGF2α + GnRH, respectively. Descriptive statistical analysis was performed. The results revealed that all the three protocols induced and synchronized estrous were almost synchronous in all the treated groups. However, protocol 3 revealed a significantly (p<0.05) better result of 70% pregnancy rate compared to other two protocols that equally provided 50% pregnancy rates. The study resolves that protocol 3 be highly recommended to change the mindset of farmers about the spread of reproductive technology in Sudan. Protocols 1 and 2 provided acceptable pregnancy rates that can be enhanced with the improvement of management.

Lactating crossbred Holstein-Friesian dairy cows (n = 331) were started on an Ovsynch regimen 68 ± 8.2 days after calving; 200 µg GnRH intramuscularly (i.m.) on Days 0 and 9, and 35 mg prostaglandin F2a i.m. on Day 7. Thirty-eight and 31 cows (11.5 and 9.4%, respectively) were in oestrus on Days 0 to 6 and 7 to 8, respectively, and inseminated, and the remainder were fixed-time inseminated (on Day 10). For these three groups, pregnancy rates (60-65 days after breeding) were 31.6, 38.7 and 34.0%, respectively (P = 0.82) and calving rates were 100, 100 and 89.9% (P = 0.23). In a preliminary trial, twelve lactating cows (45 to 60 days postpartum) with inactive ovaries were given 1500 IU eCG i.m.; 10 were in oestrus within 10 days after treatment (and inseminated) and eight of these were pregnant (30 days after breeding). The Ovsynch program resulted in acceptable reproductive performance in cyclic cows and eCG treatment has considerable promise for inducing oestrus in anoestrous cows.

An oestradiol-based protocol (named J-Synch) has been developed recently with the aim of providing for a longer proestrus, enhancing the development of the dominant follicle before ovulation, and increasing pregnancy rates following fixed-time AI (FTAI) in heifers (de la Matta and Bo 2012 Taurus 55, 17–23). Two experiments were performed in Uruguay with 1953 Angus × Hereford crossbred heifers to compare pregnancy rates obtained with the J-Synch protocol with the conventional 7-day oestradiol-based protocol. A secondary objective was to determine the effects of the time of FTAI and the addition of eCG at the time progestin device removal on pregnancy rates. In Experiment 1, 966 heifers received a DIB device (0.5 g of progesterone, Syntex, Buenos Aires, Argentina) plus 2 mg of oestradiol benzoate IM (Syntex) on Day 0. Heifers in the conventional treatment group (n = 485) received cloprostenol (500 μg, Ciclase DL, Syntex) and oestradiol cypionate (0.5 mg, Cipiosyn, Syntex) IM and had their DIB removed on Day 7 a.m. Heifers in this group were then subdivided to be FTAI on Day 9 a.m. or p.m. (i.e. 48 or 56 h after DIB removal). Heifers in the J-Synch treatment group (n = 481) received cloprostenol IM and DIB removal on Day 6 p.m. and received gonadotropin-releasing hormone (100 μg gonadorelin acetate, Gonasyn Gdr, Syntex) on Day 9 a.m.; heifers were then FTAI on Day 9 a.m. or p.m. (i.e. 60 or 72 h after DIB removal). All heifers in this experiment were also treated with 300 IU of eCG (Novormon, Syntex) at DIB removal. In Experiment 2, 987 heifers were treated with the J-Synch protocol as described in Experiment 1. At device removal (Day 6 p.m.), heifers were divided to receive (n = 488) or not (n = 499) 300 IU of eCG IM at that time and were further subdivided to receive gonadotropin-releasing hormone and FTAI on Day 9 a.m. or p.m. (i.e. 60 or 72 h after DIB removal). Pregnancy rates were determined by ultrasonography 30 days after FTAI. Data were analysed using logistic regression, and results are shown in Table 1. J-Synch-treated heifers tended (P < 0.1) to have higher pregnancy rates following FTAI, whereas time of FTAI only affected pregnancy rates following the conventional 7-day treatment (P < 0.05). However, removal of eCG from the J-Synch protocol in Experiment 2 resulted in reduced pregnancy rates when inseminations were done on Day 9 p.m. (P < 0.05). In summary, the addition of eCG to the J-Synch protocol provided for a wider window of insemination times facilitating FTAI in large groups of beef heifers.
