The aim of the present ex vivo study was to assess and compare coronal discoloration induced by four endodontic biomaterials used in regenerative endodontic procedures (REPs). Root resection was executed horizontally, 2 mm apical to the cementoenamel junction, in all fifty-four teeth. After accessing the pulp chamber, specimens were randomly divided in groups and filled with either saline solution or blood, followed by calcium silicate-based cements (CSCs) placement: ProRoot mineral trioxide aggregate (MTA) (Dentsply Sirona), Biodentine (Septodont), TotalFill BC (FKG), or pulp capping material (PCM) (Coltène). Color change (ΔE) was assessed using the L* a* b* system at five different timepoints (before and immediately after biomaterial application, 72 h, 7 days, and 6 months). The significance level for statistical analysis was set at p < 0.05. There are statistically significant differences regarding ΔE over time (p < 0.001). Statistical differences are found considering material (p < 0.001), treatment (p = 0.007), or both (p = 0.002). If solely the material or treatment is considered, regardless of time, statistically significant differences are detected (p < 0.001). After a six-month period of evaluation, blood exposure might be a critical factor in biomaterials’ color variation. Biodentine presents the lowest discoloration potential, followed by TotalFill and PCM, albeit without statistically significant differences. MTA exhibited the greatest color variation. The selection of biomaterial should consider the material’s discoloration potential.

Aim: The purpose of the present study is to assess the color stability of two calcium silicate-based cements (CSCs) used in regenerative endodontic procedures (REPs). Methods: A total of 40 acrylic single-rooted transparent teeth, with immature apex, were used. Root canals were filled up to 3 mm below the level of the cementoenamel junction, with either saline solution (Mineral Trioxide Aggregate (MTA)/saline and Biodentine/saline) or blood (MTA/blood and Biodentine/blood). Subsequently, ProRoot MTA® or BiodentineTM was placed in the root canal to create a cervical barrier. Color measurement was carried out at four different evaluation periods (3 h, 72 h, 7 days, and 6 months). Shade analysis within the L* a* b* color space was performed and color variation (∆E) calculated. The significance level for statistical analysis was set at p < 0.05. Results: The four groups showed a significant decrease in L* values over time. The ΔE value increased over time for all groups but was not statistically significant for the Biodentine/blood group. Two-way ANOVA showed no interaction between the CSC and treatment (contact with saline solution or blood). CSC used was the factor responsible for ΔE over time, inducing statistically significant color variations from T3H to T7D (p = 0.04) and T3H to T6M (p < 0.01). After 6 months, MTA/saline had 5.08 (p = 0.001) higher ΔE than Biodentine/Saline and the MTA/blood had 3.65 (p = 0.009) higher than Biodentine/blood. Conclusions: After 6 months, regardless of blood exposure, Biodentine exhibits superior color stability compared to MTA. Biodentine might be a suitable alternative to MTA as a cervical barrier material in REPs.

ABSTRACT Objective: The aim of the present ex-vivo study was to evaluate the effect of the vertical placement angle of mini-implants on primary stability by analyzing maximum insertion torque (MIT). Methods: Mini-implants were placed in 30 human cadavers, inserted at either a 90° or 60° angle to the buccal surface of the maxillary first molar. Out of 60 self-drilling mini-implants used, half were of the cylindrical type and half were of the conical type. Primary stability was assessed by means of measuring the MIT. Data were subjected to analysis of variance (ANOVA) and Newman-Keuls tests. A significance level of 5% was adopted. Results: The MIT was higher for both mini-implant types when they were placed at a 90° angle (17.27 and 14.40 Ncm) compared with those placed at a 60° angle (14.13 and 11.40 Ncm). Conclusions: MIT values were differed according to the vertical mini-implant placement angle in the maxillary posterior area. Regardless of the type of mini-implant used, placement at a 90° angle resulted in a higher MIT.

The present study evaluated the effectiveness of XP-Endo Finisher (XPF) associated with XP-Endo Shaper (XPS) or Reciproc Blue (RB) files in the reduction of Enterococcus faecalis in oval-shaped root canals. Eighty mandibular incisors with single oval-shaped root canals were contaminated with E. faecalis for 10 days at 37°C with centrifugation on alternate days. The teeth were randomly assigned to eight experimental groups (n=10) as follows: G1 – XP-Endo Shaper, G2 – XP-Endo Shaper + XP-Endo Finisher, G3 – Reciproc Blue, and G4 – Reciproc Blue + XP-Endo Finisher. Chemomechanical preparation was performed with saline solution (NaCl) or 2.5% sodium hypochlorite (NaOCl). Microbial reduction was assessed by colony forming units (CFU/mL) count before (S1) and after (S2) CMP by using sterile paper points. ANOVA, Tukey’s test and Bonferroni’s post-hoc tests were used at 5% significance level. Bacteria were present in all initial samples (P>.05). All instrumentation techniques reduced bacteria, irrespective of the irrigating solution (P<.05). With the use of NaCl, Reciproc Blue was more effective than XP-Endo Shaper (P=.035). The association of XP-Endo Finisher improved the cleaning efficacy of both systems (P=.239). With the use of NaOCl, XP-Endo Shaper and Reciproc Blue presented similar effectiveness (P=.779). XP-Endo Finisher enhanced the bacterial reduction of both systems tested (P<.05). The use of NaOCl improved the CMP, irrespective of the instrumentation technique used (P<.05).

Aim: The present study aimed to assess, in vitro, the effect of chlorhexidine on the potentiation of polycrystalline ceramic bracket staining. Methods: Seventy-two polycrystalline ceramic brackets of upper right central incisors were divided into six groups (n=12) according to immersion solution. The groups were G1 - distilled water (control); G2 - chlorhexidine; G3 - coffee; G4 - red wine; G5 - chlorhexidine associated with coffee; and G6 - chlorhexidine associated with red wine. The samples were analyzed by means of a spectrophotometer according to the CIEL*a*b* system, and color change (ΔE*) was calculated. The readings were performed at the following times: T0 - After package removal and T1 - After staining. The data were analyzed by Kruskal Wallis and t tests (p<0.05) at 5% significance level. Results: The total color variation (ΔE*) was greater in the group that received chlorhexidine associated with red wine (p<0.05) and lower in the groups that received distilled water. All other groups showed greater value variations when compared to G1 and G2. Group G6 showed a greater color change due to the potentiation of chlorhexidine with the dye substance. Conclusion: It is concluded that chlorhexidine potentiates the staining caused by red wine in polycrystalline ceramic brackets.
