We isolated several strains of human and animal origin, focusing on the genera Lactobacillus and Bifidobacterium. Samples of cow colostrum, feces and saliva of calves and piglets, and infant feces were acquired, bacterial strains were isolated and identified, and twenty strains of lactobacilli and bifidobacteria were shortlisted for further in vitro analysis of adhesion capacity to human cells, surface hydrophobicity, and auto-aggregation. Adhesion capacity was evaluated using a mixture of Caco-2 and HT-29-MTX cells and hydrophobicity was measured using the microbial adhesion to the hydrocarbon method. Lactobacillus reuteri was the most frequently isolated species, followed by L. casei subsp. paracasei and L. gasseri; all strains were isolated from infant feces. Bifidobacteria were represented by the species B. longum (infant feces), and B. thermophilum and B. pseudolongum (calves, piglets). All twenty strains showed good adhesion capacities to the mixed cell culture (17.7–37.2%), particularly, L. reuteri isolates K7 and K14 (37.2% and 35.5%, respectively). In hydrophobicity and auto-aggregation assays, strain-specific differences irrespective of the origin or taxonomic group were observed. Hydrophobicity values varied considerably (from 6.1% to 87.4%), whereas auto-aggregation ability ranged from 21.7% to 69.7%. No relation was observed between hydrophobicity and adhesion capacity; instead, auto-aggregation was apparently related with adhesion.

OBJECTIVE: To evaluate and compare the influence of three local anesthetic dental formulations manufactured in France (Septodont), Korea (Medicaine) and Pakistan (HD-Caine) in terms of cytotoxicity and anti-bacterial activities. METHODOLOGY: 90 commercially available local anesthetic cartridges of similar composition (2% lidocaine with epinephrine 100,000) viz. Septodont, Medicaine and HD-Caine were randomly collected from three different Pakistani cities and were assigned as Group S, Group M and Group H, respectively. The cartridges were further divided into three sub-groups each consisting of 10 cartridges to first evaluate cytotoxicity on Mesenchymal Stem Cells (MSCs) using a flow cytometer and secondly to investigate anti-bacterial activity by measuring zones of inhibition and through Broth Dilution Method against five bacterial strains. RESULTS: The results indicated that Septodont (94.5±0.1) and Medicaine (94.7±0.0) showed the highest viability percentage with no significant difference when the two were compared (P=0.6). HD-Caine (93.9±0.0) showed the least, being significantly (P<0.01) different from Septodont and Medicane. A statistically significant (P<0.05) difference was identified between the three study groups regarding the anti-bacterial activity. HD-Caine showed the highest anti-bacterial potential, followed by Medicaine and Septodont. CONCLUSION: Mild toxicity was observed by all the three groups in human MSCs, justifying their safe use in clinical practice. Additionally, Medicane and HD-Caine showed significant anti-bacterial activity indicating their possible use as sterile irrigants. A detailed understanding of the mechanism of protein synthesis will ultimately depend on knowledge of the native structure of the ribosome. Towards this end we have investigated the low resolution structure of the eukaryotic ribosome embedded in frozen buffer, making use of a system in which the ribosomes crystallize naturally.The ribosomes in the cells of early chicken embryos form crystalline arrays when the embryos are cooled at 4°C. We have developed methods to isolate the stable unit of these arrays, the ribosome tetramer, and have determined conditions for the growth of two-dimensional crystals in vitro, Analysis of the proteins in the crystals by 2-D gel electrophoresis demonstrates the presence of all ribosomal proteins normally found in polysomes. There are in addition, four proteins which may facilitate crystallization. The crystals are built from two oppositely facing P4 layers and the predominant crystal form, accounting for >80% of the crystals, has the tetragonal space group P4212, X-ray diffraction of crystal pellets demonstrates that crystalline order extends to ~ 60Å.

SummaryThe variability of the platelet GP Ia/IIa density has been associated with the 807 C/T polymorphism (Phe 224) of the GP Ia gene in American Caucasian population. We have investigated the genotype and allelic frequencies of this polymorphism in Spanish Caucasians. The T allele was found in 35% of the 284 blood donors analyzed. We confirmed in 159 healthy subjects a significant association between the 807 C/T polymorphism and the platelet GP Ia density. The T allele correlated with high number of GP Ia molecules on platelet surface. In addition, we observed a similar association of this polymorphism with the expression of this protein in other blood cell types. The platelet responsiveness to collagen was determined by “in vitro” analysis of the platelet activation and aggregation response. We found no significant differences in these functional platelet parameters according to the 807 C/T genotype. Finally, results from 3 case/control studies involving 302 consecutive patients (101 with coronary heart disease, 104 with cerebrovascular disease and 97 with deep venous thrombosis) determined that the 807 C/T polymorphism of the GP Ia gene does not represent a risk factor for arterial or venous thrombosis.
