AbstractMulberry leaves are used in traditional Chinese medicine and contain numerous active substances that are known to be beneficial for human health. The aim of this study was to investigate the phenolic compositions and antioxidant activities of the leaves from 23 mulberry cultivars. Qualitative LC-ESI-QTOF analysis revealed the presence of 11 phenolic compounds in the free phenolic extracts and 10 phenolic compounds in the bound fractions. Chlorogenic acid and caffeic acid were the major components in the free and bound fractions, respectively. The results revealed that the changguosang cultivar from Taiwan contained the greatest content of phenolic compounds as well as the highest antioxidant activity among the 23 cultivars examined, as determined using three separate antioxidant assays. The isoquercitrin, chlorogenic acid, and rutin contents of the free phenolic extracts displayed significant correlations with the antioxidant activities, while syringic acid and rutin were the main contributors to the antioxidant activities of the bound phenolic fractions. The obtained results demonstrate that mulberry leaves contain a variety of beneficial phenolic substances and may be suitable for further development as a herbal medicine.

One of the most common approaches for control of skin pigmentation involves the inhibition of tyrosinase, a copper-containing enzyme which catalyzes the key steps of melanogenesis. This review focuses on the tyrosinase inhibition properties of a series of natural and synthetic, bioinspired phenolic compounds that have appeared in the literature in the last five years. Both mushroom and human tyrosinase inhibitors have been considered. Among the first class, flavonoids, in particular chalcones, occupy a prominent role as natural inhibitors, followed by hydroxystilbenes (mainly resveratrol derivatives). A series of more complex phenolic compounds from a variety of sources, first of all belonging to the Moraceae family, have also been described as potent tyrosinase inhibitors. As to the synthetic compounds, hydroxycinnamic acids and chalcones again appear as the most exploited scaffolds. Several inhibition mechanisms have been reported for the described inhibitors, pointing to copper chelating and/or hydrophobic moieties as key structural requirements to achieve good inhibition properties. Emerging trends in the search for novel skin depigmenting agents, including the development of assays that could distinguish between inhibitors and potentially toxic substrates of the enzyme as well as of formulations aimed at improving the bioavailability and hence the effectiveness of well-known inhibitors, have also been addressed.

AbstractBioactivity of mulberry has been widely described, but mostly related to its fruits, while studies with leaves are scarce. Herein, in this study leaves of two mulberry species (Morus alba L. and Morus nigra L.) were characterized, compared, and evaluated in terms of their phenolic composition and antioxidant activity. Aiming to valorize mulberry leaves extracts which can be included in modern diet four different extracts (infusions, decoctions, tinctures and hydromethanolic) were examined. The chemical characterization was done by quantifying total phenolics (TPC), flavonoids (TFC) and phenolic acids (TPAC) content, L( +)ascorbic acid (ASA) and individual phenolic compounds in the extracts by HPLC technique. Moreover, DPPH and FRAP assays were used to assess the antioxidant activity of white and black mulberry leaves. Findings of these studies revealed that black mulberry leaves were richer in TFC and TPAC, while white mulberry leaves contained higher levels of individual phenolic compounds in water extracts. Gallic acid was found in the highest concentrations in two mulberry leave extracts. Correlation analysis showed strong relationships between antioxidant activity and TPC and TFC. This fact suggests the crucial role of phenolic compounds as antioxidant agents in white and black mulberry leaves. The results obtained in this study demonstrate that leaves of both white and black mulberry can be used as a valuable source of phenolic compounds with bioactive potential, which can be applied in the food sector, as foods and as promising source of natural ingredients.

Phenolic compounds in mulberry leaves harvested from three cultivars (Buriram 60, BR 60; Sakonnakhon, SK; and Khunphai, KH) at different leaf ages (tips, young, and old leaves) were identified and quantified using HPLC-DAD and HPLC-ESI/MS. A total of 13 phenolic compounds, which were mainly as caffeoylquinic acids and flavonol glycosides, were detectable. Predominant phenolic compounds were 5-O-caffeoylquinic acid (3.5–13.1 mg/g dry weight), 4-O-caffeoylquinic acid (1.3–2.4 mg/g dry weight), and quercetin-3-O-rutinoside (1.0–4.4 mg/g dry weight). Qualitative and quantitative differences in phenolic compounds in mulberry leaves were investigated among cultivars and leaf ages. Principal component analysis and hierarchical cluster analysis were used for classification of the mulberry leaves. Based on the similarity of phenolic compounds, mulberry leaves were clustered into three groups: (1) tips of leaves from all cultivars; (2) young and old leaves of mulberry cv. BR 60; (3) young and old leaves of mulberry cv. SK and KH. Therefore, according to phenolic compounds in mulberry leaves, tips of leaves from all cultivars should be intended for production of functional healthy foods.

The anti-melanogenic bioactivities of phytophenolic compounds have been well recognized. Riceberry rice contains a rich source of phenolic compounds that act as melanin inhibitors through their antioxidant and anti-tyrosinase properties. Germination has been shown to be an effective process to improve targeted phenolic compounds. In this study, germinated riceberry rice extract was tested for antioxidant activity. Total phenolic content was determined while the tyrosinase inhibitory effect was screened by the in vitro mushroom tyrosinase assay. Cytotoxicity of germinated riceberry rice extract was investigated in B16 cells before evaluating its activities on cellular tyrosinase, melanogenesis, melanin excretion, morphological appearance, and cellular oxidants. Germinated riceberry rice extract showed increased potency of antioxidants and was also twice as effective for mushroom tyrosinase inhibition when compared with ungerminated riceberry rice extract. In B16 cells, the extract inhibited cellular tyrosinase, melanogenesis, and cellular oxidants in a dose-dependent manner when compared with untreated cells. Germinated riceberry rice extract also displayed an effect on B16 cells morphology by reducing the number of melanin- containing cells and their dendriticity. Additionally, the germination of riceberry rice dominantly enhanced two phenolic acids, protocatechuic acid and vanillic acid, which have the potential for antioxidant-associated hyperpigmentation control. Thus, the restricted germination of riceberry rice tended to promote protocatechuic acid and vanillic acid, which dominantly displayed antioxidants and tyrosinase-related melanogenic inhibition.
