Tabat barito atau Ficus deltoidea (FD) merupakan tanaman obat yang telah lama digunakan masyarakat lokal Indonesia terutama di pulau Sumatera dan Kalimantan, namun belum sepopuler di Malaysia. Beberapa kendala dalam pengembangan tumbuhan sebagai obat tradisional adalah ketebatasn informasinya. Penulisan artikel ini didasarkan pada kajian literature yang terbit secara on line maupun ofline tentang manfaat dan bioaktivitas FD. Data yang diperoleh disintesakan sehingga diperoleh informasi yang konfrehensif. Secara etnobotani FD digunakan untuk mengatasi luka, diare, diabetes mellitus, tonik pasca melahirkan, penambah stamina dan obat kanker. Ficus deltoidea memiliki bioaktivitas sebagai anti diabetes mellitus, anti kanker, anti mikroba, aprosidiak, anti inflamasi, antioksidan dan anti obesitas. Aktivitas FD berhubungan dengan kandungan metabolit sekundernya terutama asam ursolik, fenolat, flavonoid, isoviteksin, moretenol, betulin, lupenone, dan lupeol. Isoviteksin memiliki aktivitas sebagai antiangiogenik dan anti kanker. Bioaktivitas FD sebagai anti kanker dan obesitas perlu dilanjutkan sehingga dapat dikembangkan sebagai obat alternatif.

The present study aimed to evaluate the potential of standardized methanolic extracts from seven Ficus deltoidea varieties in inhibiting the formation of AGEs, protein oxidation, and their antioxidant effects. The antiglycation activity was analyzed based on the inhibition of AGEs, fructosamine, and thiol groups level followed by the inhibition of protein carbonyl formation. The antioxidant activity (DPPH radical scavenging activity and reducing power assay) and total phenolic contents were evaluated. After 28 days of induction, all varieties of Ficus deltoidea extracts significantly restrained the formation of fluorescence AGEs by 4.55–5.14 fold. The extracts also reduced the fructosamine levels by 47.0–86.5%, increased the thiol group levels by 64.3–83.7%, and inhibited the formation of protein carbonyl by 1.36–1.76 fold. DPPH radical scavenging activity showed an IC50 value of 66.81–288.04 μg/ml and reducing power activity depicted at 0.02–0.24 μg/ml. The extent of phenolic compounds present in the extracts ranged from 70.90 to 299.78 mg·GAE/g. Apart from that, correlation studies between the activities were observed. This study revealed that seven varieties of Ficus deltoidea have the potential to inhibit AGEs formation and possess antioxidant activity that might be attributed to the presence of phenolic compounds.

Objective: To isolate and elucidate a cytotoxic principle against breast tumor MCF-7 cells of the Indonesian terrestrial plant Ficus deltoidea Jack leaves.Methods: F. deltoidea leaves collected at National Park of mount Gede-Pangrango, Indonesia have been subjected to chemical and biological work. F. deltoidea leaves were extracted with 96% aqueous ethanol (EtOH) and was then partitioned into three layers n-hexane, dichloromethane (CH2Cl2), and n-butanol (n-BuOH). All layers were checked for their activity against breast tumor MCF-7 cells using MTT assay method. A portion of the most active layer was purified using open column chromatography to give fraction that has toxicity against zebra fish embryos. Based on the assay-guided isolation, compound 1 was isolated. The chemical structure of 1 was elucidated using nuclear magnetic resonance (NMR) and high-resolution mass spectrometry (HRMS) data as well as comparing data with literature.Results: The CH2Cl2 layer of F. deltoidea leaves was found to inhibit breast tumor MCF-7 cells with IC50 10 µg/ml which was the most toxic among the layers. A portion of the most active layer was purified using open column chromatography to give 7 fractions. The fraction 5 showed toxicity against zebrafish embryos (LC50 35 µg/ml, 48 hpf). This fraction was purified using high performance liquid chromatography (HPLC) octadecylsilyl (ODS) column with gradient elution 70% aqueous acetonitrile (MeCN) to 100% MeCN (linear gradient) for 40 min with UV detection at 254 nm (tR = 30.99 min) to give compound 1. The chemical structure of 1 was revealed as a chlorin-type compound named methyl 10-epi-pheophorbide A.Conclusion: Methyl 10-epi-pheophorbide A was isolated for the first time from the active fraction of the Indonesian F. deltoidea leaves or tabat barito. The chemical structure including absolute stereo chemistry was elucidated using NMR and HRMS data as well as by comparison with the literature values. The 13C NMR data has been added to complete the previous report.
