A Bacteriological Survey of Raw Ground Beef1

The microbiological quality of 150 units of raw ground beef obtained from a local retail store was determined. The range of aerobic plate counts was from 6.9 × 104 to 8.3 × 107/g. By using the most probable number method 96.7% of the 150 units were positive for coliforms, 94.7% for Escherichia coli and 61.3% for Staphylococcus aureus. By the plate methods, 99.3% of the units were positive for fecal streptococci and 56% were positive for Clostridium perfringens. No salmonellae were isolated. Aerobic and anaerobic organisms were isolated and identified. E. coli was the most frequently isolated aerobe followed by organisms in the Klebsiella-Enterobacter group. Among the anaerobic isolates, C. perfringens was the organism most frequently encountered.

The consumption of lamb meat in Brazil has increased in the last years but little information about the microbiological quality of this product is available. To evaluate the hygienic-sanitary conditions of lamb carcasses, the quantification of microorganism populations indicators (mesophiles and psychrotrophs; total and thermotolerant coliforms; Escherichia coli; moulds and yeasts) and the pathogenic microorganisms indentification (Salmonella sp. and Listeria spp.) were performed. A total of 60 lamb carcasses were sampled from one abattoir in São Paulo. Swab samples were collected from three points (forequarter, back and hindquarter) on the muscle surface after carcasses final washing. Statistical analysis consisted of descriptive evaluation of the results whose counts were grouped by intervals of microorganism populations. Counts ranged from 1.0 x 10¹ to 8.0 x 10(4) colony-forming unit cm-2 (CFU cm-2) for mesophiles; 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for psychrotrophs; < 1.0 x 10(0) to 4.4 x 10(4)CFU cm-2 for moulds and yeasts; < 0.3 to > 32.0 most probable number/cm² (MPN cm-2) for total and thermotolerant coliforms and Escherichia coli. Salmonella sp. and Listeria spp. were not found in any of the carcasses. Most carcasses presented low counts for all microorganisms. Overall results may be explained by the small size of the industry where the study was taken. Results suggest that good microbiological quality lamb meat is possible to be obtained, but improvement in hygienic-sanitary conditions is still required.

Majority of the population of Bangladesh depend on tap or surface water as their source of water supply. This study was carried out to examine the microbial quality of both water and soil collected from different places using the multiple tube fermentation technique to determine coliform count by the most probable number (MPN) method in brilliant green lactose broth (BGLB) media.Inoculum from positive tubes of the presumptive test were further transferred on eosinemethylene blue (EMB) and MacConkey agar.The organisms isolated were further characterized using biochemical tests. Out of 93 water samples, 30 (32.26%) indicated the presence of lactose fermenter and gas producer in all 3 tubes of dilution series using inoculum quantities of 1.0, 0.1 and 0.01 ml, whereas out of 85 soil samples, 45 (52.94%) showed acid and gas production in all 3 tubes of dilution series.Among 85 soil samples, 40 samples that contained at least one positive in each dilution series and among 93 water samples, 31 samples that contained at least one positive in each dilution series were further re-identified with biochemical tests.This study showed 30.59% soil isolates and 26.88% water isolates were Escherichia coli which highlighted the fact that both water and soil act as a major reservoir of E.coli, which indicates possible fecal contamination as well as presence of potentially pathogenic E. coli. During visits to 20 federally inspected establishments producing meat ravioli, 577 production line samples and 480 finished product units were collected for bacteriological analyses. Four types of finished, packaged ravioli were encountered: (a) whole ravioli boiled at least 5 min; (b) raw pasta stuffed with a cooked filling; (c) only the meat component cooked; and (d) uncooked ravioli. The microbiological quality of frozen ravioli was affected more by the filling than the pasta. Slow freezing resulted in increased bacterial levels in the finished product. For the boiled ravioli, 100% of the finished product sets contained less than 50 coliforms per g, four of five sets had less than one Escherichia coli per g, and 100% had fewer than one Staphylococcus aureus per g. Four of five sets of packaged boiled ravioli had aerobic plate counts (APC) of less than 10,000 per g. For the raw pasta with a cooked filling-type ravioli, the geometric means of 9 sets were: coliforms, 47 per g; E. coli, 6.7 per g; S. aureus, 10 per g; and APC, 170,000 per g. For the ravioli with only the meat component cooked, the geometric means of 27 sets were: coliforms, 190 per g; E. coli, 1.8 per g; S. aureus, 3.9 per g; and APC, 300,000 per g. For uncooked ravioli, the geometric means of 7 sets were: coliforms, 490 per g; E. coli, 19 per g; S. aureus, 5.7 per g; and APC, 690,000 per g. Only one finished ravioli unit in 480 contained Salmonella.

ABSTRACT Cronobacter is a bacterial genus that includes seven species, and the species Cronobacter sakazakii is most related to meningitis and septicemia in infants associated with powdered infant formula (PIF). The objectives of this study were to evaluate the presence of C. sakazakii and to determine the microbiological quality of PIF for infant consumption. To do this, a total of 128 PIF samples were analyzed in four brands and countries (Chile, Mexico, Holland, and Brazil), considering three types of PIF: premature (PIF1), infant (PIF2), and follow-up (PIF3). Aerobic plate counts (APC) and Enterobacteriaceae (ENT) were assessed in accordance with Chilean official standards. The outer membrane protein A (ompA) gene was amplified to detect Cronobacter spp. and the fusA gene was amplified to identify C. sakazakii by using the PubMLST Web site and BLAST (NCBI). The antibiotic resistance profile was performed according to the Clinical and Laboratory Standards Institute standards. The pathogen was quantified by the most probable number (MPN). The results showed that APC median values for PIF1, PIF2, and PIF3 were 3.2, 4.9, and 4.8 log CFU g−1, respectively. The APC were higher in PIF2 (P &lt; 0.01) from Holland (P &lt; 0.01) in the commercial brand 4 (P &lt; 0.01). The ENT median values in PIF1, PIF2, and PIF3 were 1.8, 1.5, and 1.7 log CFU g−1, respectively. Five strains of C. sakazakii and one strain of Cronobacter malonaticus were identified as having values between 0.023 and 2.3 MPN/g. All strains (100%) harbored the ompA, plasminogen activator (cpa), and hemolysin (hly) virulence genes. To conclude, C. sakazakii was found in four PIF samples from four Chilean products and one from Mexico, which is distributed throughout America. C. sakazakii strains exhibit virulence factors and resistance to ampicillin, thus posing a risk when PIFs are consumed by infants. HIGHLIGHTS

The microbiological quality of four frozen and seven fresh seafood products (597 units in total) obtained from a local retail store were analyzed. Aerobic plate count means (geometric) ranged from 3.5 × 103/g to 9.3 × 104/g for the frozen products and from 7.8 × 104/g to 2.7 × 108/g for fresh products. Average (geometric) coliform Most Probable Number (MPN) values ranged from 1.0 to 7. 7/g for the frozen items and from 7.8/g to 4.8 × 103/g for the fresh seafoods. Employing the MPN method, 4.7% of the 597 units analyzed were positive for Escherichia coli. while 7.9% were positive for Staphylococcus aureus. Two percent of the samples contained Clostridium perfringens. Neither salmonellae nor Vibrio parahaemolyticus was isolated in any of the 597 units.

In Bangladesh, food sold at local markets are usually not kept under hygienic conditions leading to contamination with different microorganisms and deterioration of food quality. This study was carried out to examine the microbial quality of prawns sold at local markets. Prawn samples collected from different markets were assessed for their bacteriological quality using the multiple tube fermentation technique to determine coliform count using the most probable number method in Brilliant Green Lactose Broth (BGLB) media. Positive tubes of the presumptive test were further cultured on Eosine Methylene Blue(EMB) and Mac Conkey agar media. The organisms isolated were further characterized using biochemical tests. Out of the 65 samples, 47 (72.3%) showed positive results in all 3 tubes of dilution series using inoculum quantities of 1, 0.1 and 0.01 g.Among 65 samples 57 samples that contained at least one positive in each dilution series were further re-identified with biochemical tests.This study showed 56.14%isolates were Escherichia coli which conformed to expected biochemical reactions, formed round, small,elevated colonies with pink pigmentation on Mac Conkey agar media and round, small colonies with metallic green sheen pigmentation on EMB agar media. Other bacteria which presumptively appeared to be enterics and were isolated from BGLB were identified as Klebsiellapneumoniae(29.82%),Staphylococcus aureus(8.7%), Enterobacter aeroginosa(3.5%) and Salmonellatyphimurium (1.75%).Presumptive identification of E. coli in prawn in order to determine fecal contamination was able to identify ¾ of BGLB tubes with actual occurrence of E. coli. From this study it has been found that 97.14% bacteria were sensitive to Co-Trimoxazole, compared to other antibiotics used in this test whereas only 37.14% bacteria were sensitive to nitrofurantoin. This study also highlighted the fact that prawns act as a major source of E. coli which indicates possible fecal contamination as well as presence of potentially pathogenic E. coli and these bacteria are resistant at a great percentage to almost all of these antibiotics used in this study. Fecal coliforms (FC) counts were compared with Escherichia coli counts in differently contaminated freshwater samples (n = 166). FC were enumerated by plate count on triphenyl 2,3,5-tetrazolium chloride Tergitol medium. Escherichia coli were enumerated by the most probable number microplate method based on the detection of glucuronidase activity. FC and E. coli counts were highly correlated; an average E. coli/FC ratio equal to 0.77 was found, meaning that on average, 77% of FC were E. coli. Knowing the E. coli/FC ratio allows us to convert the historical microbiological quality data expressed in FC counts into E. coli abundance and thus to compare with present and future monitoring data that are (or will be) based on E. coli enumeration.

Over the last few years, ready-to-eat (RTE) foods have become popular in grocery stores all over the world. This study was conducted to evaluate the microbiological safety and shelf life of some RTE salads sold in supermarkets in Sharjah Emirate, United Arab Emirates. Samples of four RTE salads, tabbouleh, hummus, Greek salad, and coleslaw, were obtained from supermarkets and examined for aerobic bacteria, Escherichia coli, Staphylococcus aureus, Salmonella, and Listeria monocytogenes. The results indicated significant differences (P ≤ 0.05) for the aerobic bacteria count among the RTE salad types. In tabbouleh, hummus, Greek salad, and coleslaw, E. coli counts of 0.9, 0.50, 0.27, and 0.25 log most probable number (MPN)/g and total aerobic bacterial plate counts of 3.57, 2.71, 2.76, and 2.52 log CFU/g, respectively, were found after preparation (day 0). At day 0, all RTE salads tested except tabbouleh contained an acceptable count of total bacteria, but the aerobic bacteria count for all the RTE salads increased rapidly during storage from day 2 to day 6, regardless of the storage temperature (5, 25, and 40°C). Twenty percent of all samples contained E. coli, although the numbers were as low as 1 log MPN/g. No S. aureus, L. monocytogenes, or Salmonella serotypes were detected in any of the RTE salads tested in this study.

A study was undertaken to assess the microbiological quality of sprouts marketed in Mumbai and its suburbs. A total of 124 sprout samples of four different legumes—mung (Phaseolus aureus), matki (Phaseolus aconitifolius), chana (Cicer arietinum), and vatana (Pisum sativum)—were analyzed over a period of 12 months for aerobic plate counts, coliforms, yeast and mold counts, staphylococci, Salmonella, Listeria monocytogenes, Escherichia coli, E. coli O157:H7, and coagulase-positive Staphylococcus aureus. Aerobic plate counts ranged from 7.6 to 8.9 log CFU/g, coliform counts ranged from 5.4 to 7.9 log CFU/g, yeast and mold counts ranged from 3.6 to 7.3 log CFU/g, and staphylococci counts ranged from 3.3 to 6.6 log CFU/g. Nonpathogenic E. coli was detected in 13% of the mung, 26% of the matki, 40% of the chana, and 19% of the vatana samples. Salmonella Typhimurium was detected in 21% of the mung, 40% of the matki, and 4% of the chana samples. Salmonella Dublin was detected in 2% of the mung samples, and Salmonella Washington was detected in 4% of the matki samples. L. monocytogenes and E. coli O157:H7 were not detected in any of the samples examined. Coagulase-positive S. aureus was detected in 4% of the mung, 11% of the matki, and 4% of the chana samples. The results indicated that the marketed sprouts were of poor microbiological quality; therefore, further processing, such as radiation processing, is needed to ensure their safety.

Quality of drinking water and juice is very important because if the quality deteriorates by the contamination of faecally originated microorganisms, it may cause serious diarrhea associated problems leading to death. In overpopulated countries like Bangladesh, this is a common scenario to experience diarrheal diseases due to drinking non-potable water as well as contaminated fresh juices. Present study was conducted to determine the quality of drinking water and juice by detection of indicator bacteria Escherichia coli by MPN (Most Probable Number) method which was performed by three consecutive steps including presumptive test, confirmed test and completed test. Other gram negative bacteria were also identified by biochemical methods. The indicator bacterium Escherichia coli was detected in two water samples out of 15 samples and one juice sample out of fifteen samples respectively during the MPN test method. Other Gram negative bacteria found in both water and juice samples included Klebsiella spp., Alcaligenes spp., Pseudomonas spp. and Proteus spp. The quality of drinking water and juice was found to be good in Bangladesh but proper hygiene should be maintained more strictly to avoid the contamination by other gram negative bacteria which are also capable of causing disease.
