Bioserotypes and Virulence Markers of Yersinia enterocolitica Strains Isolated from Mallards (Anas platyrhynchos) and Pheasants (Phasianus colchicus)

Yersinia enterocolitica is the causative agent of yersiniosis in different animal species and in humans. Food contaminated with Y. enterocolitica is the main source of infection for humans, and swine plays a major role in the transmission of the disease. There are a limited number of reports of the prevalence of Y. enterocolitica in wild animals and birds. This study characterized virulence markers associated with Y. enterocolitica isolates recovered from mallards and pheasants. Y. enterocolitica strains were isolated from 5 (11.11%) of 45 mallards originating from a cold culture (peptone, sorbitol, and bile salts medium) belonging to biotype 1A. Serotyping showed that three of these five serotypes represented serotype O:8, one belonged to serotype O:5, and one did not agglutinate with any of the sera and was classified as nonidentified. Molecular analysis for virulence markers detected the ystB gene, which encodes an enterotoxin, in five isolates. Y. enterocolitica was not detected in any of the 16 examined pheasants.

     Yersiniosis is a zoonotic disease which infects many animal species. A preliminary study was done to detect the incidence of Yersinia enterocolitica in sheep. One thousand and two hundred fecal samples were collected randomly from sheep in four governorates in the southern region of Iraq: Thi-Qar, Al-Muthana, Messan and Basrah, in the period from July 2016 to June-2017. Enrichment Yersinia broth at 4°C for 48 hours was used for isolation of Yersinia enterocolitica. Selective Cefsulodin Irgasan Novobiocin and non selective media were used for bacterial culture. The identification of Yersinia enterocolitica was based on colony morphology and biochemical characters, API 20E and VITEK2 compact systems were used for the same purpose also. Data were analyzed by using SAS, Version 9-1. Chi- square test was used for comparison. The total isolation rate of Y. enterocolitica was 5.16% (62). High and low rates of isolation were recorded in Al-Muthana (6. 01%) and Basrah (3.86%). Similarly a significant high isolation rate was recorded in diarrheic sheep (17.4%), moreover the infection rate increased significantly (9.5%) in the young sheep (1day to 6 month's age). Furthermore, the recovery rate of Y. enterocolitica  increased significantly during the cold months (12%) as compared with temperate and hot months (spring and summer), at the same instance, non significant variation among sex difference was detected. Fever, diarrhea and mild to moderate degree of dehydration were the most common clinical manifestations observed on the infected animals. Conclusively sheep were considered a source of infection to other species including human being, and the spread of microorganism increased markedly in the cold and wet environment.

AbstractFree-living animals are an important environmental reservoir of pathogens dangerous for other animal species and humans. One of those is Yersinia (Y.) enterocolitica, the causative agent of yersiniosis - foodborne, enzootic disease, significant for public health. The purpose of the study was to identify bioserotypes and virulence markers of Y. enterocolitica strains isolated from roe deer (Capreolus capreolus) and red deer (Cervus elaphus) obtained during the 2010/2011 hunting season in north-eastern Poland. From among 48 rectal swabs obtained from 24 roe deer, two strains of Y. enterocolitica from one animal were isolated. Although both belonged to biotype 1A they were identified as different serotypes. The strain obtained from cold culture (PSB) belonged to serotype O:5, while the strain isolated from warm culture (ITC) was regarded as nonidentified (NI), what may suggest mixed infection in that animal. The presence of ystB gene, coding for YstB enterotoxin, directly related to Y. enterocolitica pathogenicity was detected in both strains using triplex PCR. The effect of the examination of 32 swabs obtained from 16 red deer was the isolation of two Y. enterocolitica strains from two different animals. Both belonged to biotype 1A with NI serotype, but were originated from different types of culture. They gave positive results in case of products of a size corresponding to the ystB gene. No amplicons corresponding to ail and ystA genes were found. Roe deer and red deer may carry and shed Y. enterocolitica, what seems to be important in aspect of an environmental reservoir of this pathogen. The Y. enterocolitica strains isolated from wild ruminants had the amplicons of the ystB gene, what suggest they can be potential source of Y. enterocolitica infection for humans

ABSTRACT Yersinia enterocolitica is an enteric pathogen that exploits diverse means to survive in the human host. Upon Y. enterocolitica entry into the human host, bacteria sense and respond to variety of signals, one of which is the temperature. Temperature in particular has a profound impact on Y. enterocolitica gene expression, as most of its virulence factors are expressed exclusively at 37°C. These include two outer membrane proteins, YadA and Ail, that function as adhesins and complement resistance (CR) factors. Both YadA and Ail bind the functionally active complement alternative pathway regulator factor H (FH). In this study, we characterized regions on both proteins involved in CR and the interaction with FH. Twenty-eight mutants having short (7 to 41 amino acids) internal deletions within the neck and stalk of YadA and two complement-sensitive site-directed Ail mutants were constructed to map the CR and FH binding regions of YadA and Ail. Functional analysis of the YadA mutants revealed that the stalk of YadA is required for both CR and FH binding and that FH appears to target several conformational and discontinuous sites of the YadA stalk. On the other hand, the complement-sensitive Ail mutants were not affected in FH binding. Our results also suggested that Ail- and YadA-mediated CR does not depend solely on FH binding.

This paper reports on the serological and biochemical characteristics of 24 human isolates of Yersinia enterocolitica submitted to the California Department of Health from 1968 through 1975. Nine different serotypes were represented. The majority of strains were serotype O:8 (six strains) and serotype O:5 (five strains). Sources of the isolates included feces (12 cases), blood (3), sputum or throat (3), bile or bowel drainage (2), wounds (2), breast abscess (1), and skin abscess (1). Clinical histories indicated a number of different syndromes. Underlying medical conditions existed in 13 cases. Results of selected biochemical tests and antimicrobial susceptibility tests on the strains indicated grouping compatible with the O serotypes of the organisms.

A rare ocular dirofilariasis case along with the clinical characteristics, treatment, and outcome is reported. A whitish roundworm (10.6 cm long and 0.5 mm width) emerged from the pterygium, a triangular tissue growth on the cornea of the eye, of a male patient. The worm had a rounded anterior part, mouth without lips, smooth cuticular surface, and short rounded posterior tail with spicules: these features suggested that it was a male Dirofilaria sp. Molecular identification confirmed that the worm belonged to Dirofilaria immitis. This is the first molecular confirmation that D. immitis is a causative agent of ocular dirofilariasis in Thailand: dirofilariasis is a newly emerging zoonotic disease. Physicians should be alert to zoonotic filarial worms and knowledgeable about treatment of this disease.

Monitoring infectious diseases is one of the most important pillars of preventative veterinary medicine in zoological collections. The zoo environment offers a great variety of different animal species living in proximity and in contact with small wild animals and vectors (e.g., ticks and mosquitos). In this context, tick-borne encephalitis virus (TBEV), Usutu virus (USUV), and West Nile virus (WNV) causing vector-borne diseases are emerging pathogens that raise concern. The aim of the study was to detect antibodies to selected flaviviruses in various animal species in the Ljubljana Zoo, Slovenia. In total, 874 sera from 96 animal species were tested for antibodies to TBEV by enzyme-linked immunosorbent assays (ELISA); positive samples were confirmed by a virus neutralization test (VNT) using TBEV, WNV, and USUV antigens. Antibodies to TBEV were detected by ELISA in 3.9% (34/874) of zoo animals, with 4% (30/753) in mammals and 5% (4/86) in birds; the sera of reptiles (n = 34) and amphibians (n = 1) were negative. Antibodies to TBEV were confirmed by VNT in 11 mammals; one bird was positive for both WNV and USUV. The mixture of exotic animal species and their contact with wild animals and vectors such as ticks and mosquitos suggest that screening of infectious diseases in zoo animals might provide good insight into the epizootological situation of the area. This is the first survey of TBEV, WNV, and USUV in a zoological collection in Slovenia.
