Investigation of the Essential Oil of Ormenis africana from Morocco: Revision of Chemical Composition and Antibacterial Activity

Abstract- The essential oil composition from the aerial parts of Ormenis africana (Asteraceae), an endemic species from Morocco, has been investigated by GC/MS. A total of 31 compounds were identified, representing 77%. After fractionation by column chromatography, the main compound was isolated and its structure elucidated by NMR spectroscopy. The essential oil was dominated by oxygenated compounds with spathulenol (45.8%) followed by camphor (7.1%), -cadinol (5.9%) and -bisabolol (5.9%) as the main compounds. This oil can be classified as spathulenol-type according to its spathulenol content. In vitro the antibacterial activity of the whole essential oil against three Gram positive (Bacillus cereus, Enterococcus faecalis, Streptococcus C) bacteria and three Gram negative (Proteus vulgaris, Escherichia coli, Pseudomonas aeroginosa) bacteria, showed significant results. The essential oils from fresh aerial parts of Monticalia greenmaniana (Hieron) C. Jeffrey (Asteraceae) collected in March, were analyzed by GC/MS. Oil yields (w/v) of 0.1% (flowers), 0.07%, (stems) and 0.1% (leaves) were obtained by hydrodistillation. Thirteen, sixteen and eighteen components, respectively, were identified by comparison of their mass spectra with those in the Wiley GC-MS Library data base. The major components of the flower and stem oils were 1-nonane (38.8% flowers; 33.5% stems), α-pinene (29.0% flowers; 14.8% stems) and germacrene D (15.6% flowers; 18.6% stems). However, in the leaf oil, germacrene D was observed at 50.7%, followed by β-cedrene at 8.4 %. The leaf essential oil showed a broad spectrum of antibacterial activity against the important human pathogenic Gram-positive and Gram-negative bacteria Staphylococcus aureus (ATCC 25923), Enterococcus faecalis (ATCC 19433), Escherichia coli (ATCC 25922), Pseudomonas aeruginosa (ATCC 27853) and Klebsiella pneumoniae (ATCC 25955) with MIC values ranging from 75 to 6000 ppm.

The aerial parts of Satureja spicigera were collected at full flowering stage at Gazvin, Iran. The essential oil was isolated by hydrodistillation and analyzed by a combination of capillary GC and GC-MS. Fourteen compounds were identified, of which carvacrol (53.74%) and thymol (36.03%) were the main constituents, representing 99.12% of the total oil. The in vitro antibacterial activity of the essential oil was determined against six ATCC standard bacterial strains (Bacillus subtilis, Enterococcus faecalis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa) using disc diffusion as well as measurement of minimum inhibitory concentrations. The disc diffusion results and MIC values indicated high inhibitory activity against the test bacteria. The most susceptible organisms were the Gram-positive B. subtilis and S. aureus followed by E. faecalis, usually resistant to most common antibiotics. Among the Gram-negative bacteria, E. coli and K. pneumoniae were highly sensitive to the different oil concentrations in the disc diffusion method. Finally, P. aeruginosa, a highly resistant organism to most antibiotics, showed moderate susceptibility to Satureja spicigera essential oil.

The essential oil of the aerial parts of Centaurea sempervirens L. (Asteraceae), synonym : Cheirolophus sempervirens (L.) Pomel, was obtained by steam distillation and analyzed by GC-FID and GC-MS. 30 components were identified corresponding to 78.5% of the total oil. Among the identified constituents, oxygenated compounds represented 33.4%, from which 21.2% were hydrocarbons, 10.7% were sesquiterpenes. The non oxygenated compounds were hydrocarbons (9.8%). Phthalates represented 35.3% of the total oil. The major components were 6,10,14-trimethylpentadecan-2-one (12.4%) and epi-torilenol (5.1%). This is the first report on the chemical composition of the essential oil of this species.

The aerial parts of Ocimum gratissimum L. (Lamiaceae) harvested in Togo was steam-distilled and investigated for essential oil composition (GC and GC/MS) and in vitro antifungal activities. Thymol (31.79 %), p-cymene (15.57 %) and γ-terpinene (12.34 %) and were the major components of the oil. Other notable components identified in this oil were myrcene (6.94 %) and α-thujene (6.11 %).The in vitro antifungal activity was recorded with the minimum inhibitory concentrations (MICs) ranging from 80 to 150 µl.l-1, 150 to 500 µl.l-1  and from 100 to 150 µl.l-1 respectively on dermatophytes, imperfect filamentous fungi and pathogenic yeasts. Likewise, on tested fungi the minimum fungicidal concentration (MFC) varied from 300 µl.l-1 to 500 µl.l-1, 500 to 700 µl.l-1 and from 250 to 300 µl.l-1, respectively on dermatophytes, imperfect filamentous fungi and pathogenic yeasts. Hyptis colombiana is an aromatic shrub native to the Colombian and Venezuelan Andes. Aerial parts were collected in Mérida State at about 3100 m above sea level in February 2005, and May and October 2006. The essential oil was found to contain germacrene D and β-caryophyllene as main constituents (about 50%). The February 2005 and October 2006 oils were found to have antibacterial activity against Staphylococcus aureus, but not the May 2006 oil, probably due to the lack of some minor constituent.

The chemical composition of the essential oil of Ballota nigra L. ssp foetida obtained from the flowering aerial parts was analyzed by GC/MS. From the 37 identified constituents of the oil, β-caryophyllene (20.0%), germacrene D (18.0%) and caryophyllene oxide (15.0%) were the major components. The oil was active against both Gram-negative and Gram-positive bacteria as well as against three Candida species.

A water-distilled essential oil (EO) from the aerial parts of Pimpinella cypria Boiss. (Apiaceae), an endemic species in northern Cyprus, was analyzed by GC-FID and GC-MS. Forty-five compounds were identified in the oil, which comprised 81.7% of the total composition. The compound classes in the oil were oxygenated sesquiterpenes (33.9%), sesquiterpenes (22.0%), monoterpenes (11.4%), oxygenated monoterpenes (2.6%), and phenylpropanoids (7.5%). The main components of the oil were ( Z)-β-farnesene (6.0%), spathulenol (5.9%), ar-curcumene (4.3%), and 1,5-epoxy-salvial(4)14-ene (3.8%). The P. cypria EO deterred yellow fever mosquitoes (Aedes aegypti) from biting at a concentration of 10 μg/cm2 in in vitro bioassays. The oil was tested for repellency in assays using human volunteers. The oil had a minimum effective dosage (MED) for repellency of 47 ± 41 μg/cm2 against Ae. aegypti, which was less efficacious than the positive control N,N-diethyl-3-methylbenzamide (DEET). In larval bioassays, P. cypria EO showed an LC50 value of 28.3 ppm against 1st instar Ae. aegypti larvae. P. cypria EO demonstrated dose dependent repellency against nymphs of the lone star tick, Amblyomma americanum. Between 45.0% and 85.0% repellency was observed at concentrations ranging from 26 to 208 μg/cm2. However, P. cypria EO was less effective compared with DEET in the tick bioassays. Cytotoxicity assays showed that the P. cypria EO did not exhibit significant effects up to the maximum treatment concentration of 50 μg/mL on HEK293, PC3, U87MG, and MCF cells. P. cypria EO also demonstrated moderate antimicrobial activity against Gram-negative and -positive bacteria with MICs ranging from 15.6 to 62.5 μg/mL, except for Candida albicans, which showed the same MIC value of 7.8 μg/mL as the positive control, flucytosine. This is the first report on the chemical composition of P. cypria EO and its insecticidal, toxicant, cytotoxic, and antimicrobial activity.

The essential oil from the fruits of Vismia baccifera Triana & Planch. (Gutttiferae), collected in June 2009, was analyzed by GC/MS. A yield of 0.6% oil was obtained by hydrodistillation. Twenty-seven components were identified by comparison of their mass spectra with those in the Wiley GC-MS Library data base. The major components were trans-cadin-1,4-diene (36.6%), cis-cadin-1,4-diene (18.8%) and β-caryophyllene (11.9%). The essential oil showed a broad spectrum of antibacterial activity against the important human pathogenic Gram-positive and Gram-negative bacteria Staphylococcus aureus (ATCC 25923), Enterococcus faecalis (ATCC 29212), Escherichia coli (ATCC 25992), Pseudomonas aeruginosa (ATCC 27853) and Klebsiella pneumoniae (ATCC 23357) with MIC values ranging from 9 to 37 μg/mL.

Achillea filipendulina is an aromatic plant of family Asteraceae with bright yellow colour flowers. The objective of the study was to extract the essential oil from the flowers of the selected plant using Clevenger apparatus, to analyse the chemical composition of the oil using GC-MS and to screen the essential oil for its antibacterial property against both gram-positive and gram-negative bacteria. GC-MS results revealed that the main chemical constituents present in the oil were trans 2,7 Dimethyl 4,6 octadien-2-ol (27.93%), borneol (21.44%) and santolina triene (7.13%). The results of agar well diffusion assay revealed that the oil showed antibacterial activity against all the tested bacteria except Pseudomonas aeruginosa. Minimum inhibitory concentration (MIC) of oil was determined against the sensitive bacteria. MIC values against Staphylococcus aureus and Escherichia coli were similar (0.312% v/v) and for Bacillus cereus, MIC value was found to be (0.156%). Results indicated the great antibacterial potential of oil of this plant against gram positive and gram negative bacteria.
