Lipid peroxidation functional state changes in patients with obstructive jaundice depending on the level of bilirubin in the blood

Obstructive jaundice (OJ) is the most common syndrome among diseases of the hepatopancreatoduodenal region and is found in 12-45% of cases. OJ may be benign and malignant etiology. Despite the evidence of the participation of bilirubin in reducing the bactericidal properties of neutrophils, there are no data currently on changes in the functioning of the antioxidant defense system depending on the level of bilirubin in the blood of patients with OJ of various origins. Research in this direction reveals the possibility for the development of pathogenetic recommendations for influencing these links of the pathogenesis of the disease. The study included men with OJ of non-malignant (OJNMG) (n = 47; mean age - 52.02 ± 5.18 years) and OJ of malignant genesis (OJMG) (I-II stages of the malignant process) (n = 45; mean age - 53.02 ± 4.8 years), divided into three subgroups, depending on the level of bilirubin in the blood. The indicators of practically healthy men as a control (n = 50, average age - 48.7 ± 3.9 years) were used. Spectrophotometric and statistical research methods were used. A statistically significant decrease of superoxide dismutase, glutathione-S-transferase, glutathione-peroxidase, ceruloplasmin, an increase in the values of diene conjugates, malondialdehyde in the group of patients with OJNMG relative to the control was revealed, regardless of the level of bilirubin in the blood. The presence of malignant genesis of the disease with more intense changes in the studied parameters relative to control is accompanied. Comparison of indicators between groups of patients with OJ of different genesis showed a decrease in the values of glutathione-S-transferase and an increase in the level of diene conjugates in patients with OJMG and the level of bilirubin less than 60 μmol / L, as well as an increase in the content of diene conjugates in patients with OJNMG and a level of bilirubin 60- 200 μmol / L in comparison with the corresponding groups of patients with OJNMG. Thus, both in the groups with OJNMG and in the groups with OJMG, there is a significant decrease in the activity of the main antioxidant enzymes and an increase in lipid peroxidation products, regardless of the level of bilirubin in the blood. The presence of malignant genesis is characterized by more intense differences. The revealed changes can serve as additional criteria for optimizing the diagnosis and treatment of this cohort of patients.

Background. The problem of gastric cancer remains unresolved throughout the world, while chronic atrophic gastritis (CAG) increases the likelihood of its development by 15 times. In the Russian Federation, the incidence of gastric cancer (GC) is among the highest, with it prevailing among males. One of the leading mechanisms in molecular pathology of membranes is lipid peroxidation (LPO). The severity of oxidative membrane damage depends on concomitant diseases, contributing to emergence and progression of pathological processes and development of cancer. Currently, the problem of LPO is unsolved in biological systems.The aim of this study was to investigate the state of LPO and antioxidant defense system in CAG and GC. Materials and methods. The parameters were studied in 45 patients with CAG and 50 patients with GC. The control group included 50 practically healthy volunteers without gastrointestinal complaints, who did not have changes in the gastric mucosa according to the fibroesophagogastroduodenoscopy (FEGDS) findings.Results. In patients with CAG, an increase in malondialdehyde, superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase was found in the blood plasma compared with the control group. In patients with CAG, lipid peroxidation was activated, and the malondialdehyde level increased by 3.5 times relative to normal values. At the same time, the body fought against oxidative stress by increasing the activity of antioxidant enzymes, such as superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase. All patients with GC showed pronounced oxidative stress in the blood plasma in the form of a 45-fold increase in malondialdehyde. The activity of the main antioxidant enzyme superoxide dismutase was reduced in GC. Catalase was activated, which indicated pronounced oxidative stress, significant damage to blood vessels, and massive cell death. Glutathione-related enzymes (glutathione S-transferase and glutathione peroxidase) and the antioxidant protein ceruloplasmin were activated, which also indicated significant oxidative stress and severe intoxication in patients with GC.Conclusion. Depending on the stage and type of cancer, an in-depth study of lipid peroxidation and factors of the antioxidant defense system can be used to correct therapy and prevent cancer and can serve as markers of progression and prognosis in gastric cancer.

The effects of selenium (Se) on antioxidant defense system in liver and kidneys of rats with cadmium (Cd)-induced toxicity were examined. Cd exposure (15 mg Cd/kg b.m./day as CdCl2 for 4 weeks) resulted in increased lipid peroxidation (LP) in both organs (p<0.005 and p<0.01). Vitamin C (Vit C) was decreased in the liver (p<0.005), whereas vitamin E (Vit E) was increased in the liver and kidneys (p<0.005 and p<0.05) of Cd-exposed animals. Superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities were decreased in both tissues (p<0.05 and p<0.005), whereas catalase (CAT) activity was decreased only in liver (p<0.005). Glutathione S-transferase (GST) increased in both tissues (p<0.005 and p<0.01). Treatment with Se (0.5 mg Se/kg b.m./day as Na2SeO3 for 4 weeks) significantly increased liver and kidneys SOD and GSH-Px activities (p<0.05 to p<0.005), as well as CAT and GST activities only in the liver (p<0.01). In animals exposed to Se, both the concentrations of Vit C (p<0.01) and Vit E (p<0.005) were increased in both tissues. Co-treatment with Se resulted in reversal of oxidative stress with significant decline in analyzed tissues Cd burden. Our results show that Se may ameliorate Cd-induced oxidative stress by decreasing LP and altering antioxidant defense system in rat liver and kidneys and that Se demonstrates the protective effect from cadmium-induced oxidative damage.

The article presents the results of research on the state of the antioxidant defense system of piglets under nitrate-nitrite toxicosis and the action of corrective factors (methionine, phenarone and methyphene). Nitrate-nitrite load in piglets caused inhibition of the activity of enzymes of the antioxidant defense system, that indicates a decrease in the activity of superoxide dismutase, catalase and glutathione peroxidase in the serum of testing animals. The lowest activity of antioxidant enzymes under chronic nitrate-nitrite toxicosis was on the 60th day of the experiment in the serum of piglets of the control group, where accordingly the activity of superoxide dismutase decreased by 14 %, catalase activity – by 18 %, glutathione peroxidase activity – by 12 %. Lipid peroxidation processes are activated in the blood of animals, namely the level of lipid hydroperoxides and diene conjugates increases under the conditions of experimental nitrate-nitrite load. It should be noted that the highest level of lipid peroxidation products in the serum of piglets with chronic nitrate-nitrite toxicosis was on the 60 th day of the testing, and accordingly the content of lipid hydroperoxides increased by 67 % and diene conjugates – by 33 %. Administration of methionine, phenarone and methyphene under nitrate load contributed to the activation of the antioxidant defense system compared to animals in the control group. The use of antioxidant drugs in sick piglets also contributed to the inhibition of lipid peroxidation, as indicated by a decrease in the content of lipid hydroperoxides and diene conjugates in the serum of experimental group. More pronounced antioxidant effect was shown by administrating methyphene. The mechanism of action of phenarone and methyphene is associated with the direct effect of its components on the inhibition of the absorption of metabolites that have a toxic effect on cell membranes of animals. As a result of the adsorbing action of zeolite, which is part of the drugs, in the digestive tract there is a decrease in the concentration of substances that can be substrates for lipid peroxidation, as well as removal of toxic metabolites in the blood, which are prooxidants. This process occurs by osmosis and diffusion of these substances through the capillaries of the microvilli of the small intestine and their subsequent fixation on the sorbent granules.

Несмотря на достигнутые успехи в диагностике и лечении, на сегодняшний день проблема хронического простатита у пациентов с доброкачественной гиперплазией простаты продолжает оставаться актуальной. Тем временем, выздоровление или же стойкая ремиссия хронического простатита наступает только у 30-35% больных, которые получают традиционное лечение. В патогенезе хронического простатита важное место занимают мембранопатологические процессы, которые обусловлены активацией перекисного окисления липидов и нарушением состояния антиоксидантной системы защиты. У больных с хроническим простатитом и доброкачественной гиперплазией простаты наблюдается существенное усиление процессов липопероксидации на фоне сниженной функциональной способности антиоксидантных систем защиты организма. Поэтому равновесие в оксидантно-антиоксидантной системе является важным звеном в поддержании гомеостаза и, в частности, при патологии предстательной железы, предопределяет включение в комплексное лечение средств антиоксидантного действия. Перспективным можно считать применение препаратов с высоким содержанием биофлавоноидов и антиоксидантных витаминов. Включение в комплексное лечение таких пациентов препарата Кверцетина приводит к сокращению сроков нормализации клинико-лабораторных проявлений заболевания благодаря восстановлению равновесия между перекисным окислением липидов и состоянием антиоксидантной системы защиты Despite the successes achieved in the diagnosis and treatment, to date, the problem of chronic progression in patients with benign hyperplasia of the growth continues to remain relevant. Meanwhile, recovery or persistent remission of chronic prostatitis occurs only in 30-35% of patients who receive traditional treatment. In the pathogenesis of chronic prostatitis, membrane-pathological processes take an important place, which are caused by the activation of lipid peroxidation and impaired state of the antioxidant defense system. In patients with chronic prostate and benign hyperplasia, a significant increase in lipoperoxidation processes is observed against the background of a reduced functional ability of antioxidant defense systems. Therefore, the equilibrium in the oxidant-antioxidant system is an important link in the maintenance of homeostasis and, in particular, with the pathology of the prostate gland, allows the inclusion of antioxidant drugs in the complex treatment. The use of drugs with a high content of bioflavonoids and antioxidant vitamins can be considered promising. The inclusion of such patients in the complex treatment of the drug Quercetin leads to a reduction in the normalization period of the clinical and laboratory manifestations of the disease due to the restoration of the equilibrium between the peroxidation of the lipid peroxidation system and the state of lipid peroxidation

The article contains the research results of the effect of cadmium chloride on the indexes of enzyme and nonenzyme systems of  antioxidant defense system in young cattle, such as the activity of catalase, superoxide dismutase, glutathione peroxidase, glutathione levels of vitamins A and E. It is established that feeding calves at a dose of toxicant 0.04 mg / kg activity of catalase, superoxide dismutase, glutathione peroxidase, glutathione levels of vitamins A and E in the blood of experimental animals decreased throughout the experiment. The lowest indicators of antioxidant in the blood of young cattle is set on the twenty -fourth day of the experiment, which is associated with increased activation of lipid peroxidation and the balance between antioxidant system and lipid peroxidation intensity. Given the cadmium load of young cattle it is used a new integrated drug with antioxidant action «Metisevit», which includes metifen, sodium selenite and vitamin E wich is founded as stimulating effects on the activity of antioxidant protection. In particular,it is established probable increase in activity of catalase, superoxide dismutase, glutathione peroxidase, glutathione levels, vitamin A and vitamin E in the blood of young cattle, which has performed cadmium stress. These changes occur through comprehensive action components of the drug «Metisevit» that leads to the normalization of metabolic processes and free radical in the body of the bull. The results of the research indicate antioxidant drug «Metisevit» in the application of its young cattle and the validity of his administration to improve the body's antioxidant status of chronic cadmium toxicosis.

Background: The prevalence of varicose veins of small pelvic veins in women of reproductive age varies widely – from 5.4 to 80 %, due to the low specificity and low sensitivity of clinical diagnostic techniques, and the absence of biomarkers that can be used to identify the formation and progression of varicose veins of small pelvic veins in women.Aims: To identify changes in the levels of parameters of the system “lipid peroxidation – antioxidant protection” with the development of varicose veins of the pelvis in women, as well as the possibility of their diagnostic use.Materials and methods: We examined 200 women with clinical signs of varicose veins of small pelvis. Control group included 30 women without any pathology of venous system. Treatment group included 137 women with varicose veins of small pelvis: with mild degree of severity – 39 women, with moderate degree of severity – 65 women, with severe degree of severity – 33 women. We performed comparative analysis of lipid peroxidation – antioxidant defense system in women with and without varicose veins of small pelvis taking into account the severity degrees of pathological process.Results: We obtained the most informative indices for lipid peroxidation – antioxidant defense processes in blood of women with varicose veins of small pelvis: concentration of diene conjugate (DC) for the mild degree of severity of pathological process, lipid hydroperoxide (LHP), DC and malonic dialdehyde (MDA) – for the moderate and severe degree of severity, and also changes in the activity of antioxidant enzymes with increase of the severity of the disease. Assessment of the level of catalase (Cat) and glutathione peroxidase (GP) showed their highest informative value at the mild degree of severity. Concentrations of glutathione reductase (GR), glutathione S-transferase (GST) and reduced glutathione (GSH) are more statistically significant markers for severe forms of pathological process.Conclusions: Our research showed the relevance and advisability of the studying the levels of lipid peroxidation – antioxidant defense indices in women with varicose veins of small pelvis. We registered the highest diagnostic value of DC, Cat and GP in the beginning of the disease and of LHP, DC, MDA, GR, GST and GSH – at severe forms of the pathological process. We obtained reference values for DC, MDA, Cat, SOD and GP concentration that can be considered as the predictors of the development of varicose veins of small pelvis.
