Effect of Dietary Vitamin C and and #914;-Glucan to Alleviate the Toxic Effect of Copper Sulphate in Tilapia Fish

The majority of researchers have studied the following group of microelements: B, Zn, Mn, Cu, Na, Co, Mo, I, Sn, Cl, Al, V, F, Cr, Hg, Cs, Li, Cd, As, Th, Rb, Cr, W, Ti, Sn, Se, Ba, Br. Sporadically, the following elements have been mentioned too: Au, Ra, Hg and Pb. In this study, the effects of copper treatments and their combination with zinc and lead microelements on the germination of maize and barley were investigated using different concentrations of these microelements. Six treatments were used: 1. Copper-sulphate (CuSO4) applied alone, 2. Zinc-sulphate (ZnSO4) applied alone, 3. Copper applied with zinc, 4. Lead-nitrate (Pb(NO3)2) applied alone, 5. Copper applied with lead and 6. Untreated control. Maize (Kiskun SC 297) and barley caryopsis were treated with copper and zinc solutions in the following concentrations: 0.03%, 0.003% and 0.0003%. Maize and barley caryopsis were treated with these solutions for 12 and 24 hours. Maize and barley caryopsis were also treated with lead solutions Pb(NO3)2 with different concentrations: 0.0005%, 0.005% and 0.05%. Maize and barley were treated with these solutions for 12 and 24 hours. In the combined treatments (3 and 5), the same concentration was used for each microelement as in treatments 1, 2 and 4. Control treatments were treated with water for both plant species. Our results showed that copper microelements significantly inhibit germination compared to the untreated control. The toxicity of copper is higher if concentration increases. Zinc microelements also inhibit germination, however its effect highly depends on the microelement concentration. Treatments of copper + zinc also inhibit germination. The two microelements applied together cause more phytotoxicity than they do alone. Lead is highly toxic to plants even in low concentrations. The toxic effect on germination dramatically increased when lead was applied with copper.

Copper sulphate pentahydrate (CuSO4∙5H2O) is widely used as a pesticide not only in agricultural but in aquaculture farming as well. Copper sulphate is a cheap chemical and able to contaminate the environment, especially water sources, which is crucial for fish harvesting and farming. The copper contamination in some areas is caused over decades because this pesticide has long been used everywhere. Copper ions inhibit invasive aquatic plants and many microorganisms but contaminate soil and natural water resources. The family of copper-containing chemicals is frequently used as algaecides in swimming pools. Despite the high toxicity of copper ions for fish in freshwater ponds, copper sulphate remains one of the prevalent pesticides in fish farming everywhere. High cytotoxicity and accumulation of the copper ions in sediments require study and calculation of the optimal dosage for its use as an antiseptic agent which will not have a detrimental effect on various tissue types of aquatic organisms. The main recognized mechanism which accompanies the toxic effect of copper ions is the generation of oxidative stress. Neural tissue cells are extremely susceptible to oxidative damage and the functions of the CNS are critical to the vitality of organisms. Glial cells maintain the structure and many vital functions of neurons. The cytoskeleton glial fibrillary acidic protein (GFAP), transcriptional nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB) and Poly(ADP-ribose) polymerase (PARP) are critical participants in a cellular response to a toxic agent impact. As this takes place, it could be applied in biomarking of heavy metal toxicity. In the presented study, we investigated the effects of copper ions on PARP, NF-kB, and GFAP expression in the Tigris scraper Capoeta umbla brain tissue. For 96 hours the fish were exposed to copper sulphate at sublethal concentrations, namely 1/2, 1/4 and 1/8 of the LD50 value. Western blot analysis of GFAP and PARP was used to assess further effects in the brain tissue. Every studied dose of copper significantly downregulated the expression of GFAP after 72 hours of treatment. In spite of the common increment in the GFAP content, 48 hours exposure to copper initiated the upregulation of that cytoskeleton marker. Moreover, treatment with copper sulphate induced several changes in the β-actin level, especially in the fish group treated for 72 hours. The observed effect of copper in the fish brain evidences the unspecific toxic effect of the copper ions in the brain tissue cells. The obtained results demonstrated meaningful disturbance in the expression of transcriptional factor NF-kB in the brain of the fish group exposed to copper. The changes found in the fish brain indicate the dose-dependent effect in a concentration range 185–740 µg/L of copper sulphate during 72 hours. However, the exposure to low dose of copper ions showed no effect in the fish group treated for 24 hours. Comparative analyses of the PARP content in the brain of fish exposed to copper for 72 hours was significantly less than in the groups treated with copper for both 24 and 48 hours. Thus, the copper ions in the dose range 185–740 µg/L can suppress PARP expression in a time-dependent manner. The results showed that copper ions could induce astroglial response accompanied by modulations of NF-kB and PARP-1 expression. The data obtained in this study suggest that copper sulphate has a significant effect on astrogliosis and DNA damage in the fish brain.

Introduction: Cigarette smoking induces many chronic illnesses, but in developed countries it is a preventable risk factor. However, by increasing the protective protection mechanism, it may be possible to alleviate the smoke-induced damage. As smoking risk is minimised by vitamin C intake, it is recommended that smokers should take more vitamin C. This inverse correlation between both vitamin C intake and serum levels and smoking was independent of age, sex, body weight, ethnicity, and consumption of alcoholic beverages. The negative association between cigarette smoking and serum vitamin C levels continued, following further adjustment for dietary vitamin C intake. The risk of severe hypovitaminosis C, especially when not accompanied by vitamin supplementation, has been increased in smokers. These data indicate that the inverse relationship between smoking and serum vitamin C levels exists independently of dietary intake, while smoking adversely affects preferences for vitamin C rich foods. Methods: A survey method was performed, with 50 smokers (S) receiving either 500 mg of vitamin C or placebo (P) daily for 4 weeks, and 50 non-smokers receiving vitamin C without supplementation. All finished the hearing. Both groups were equal and C: 14.2 + /- 1.8 pack-years was the amount of cigarettes smoked. Concentrations of plasma vitamin C increased significantly (p < 0.005) only in the vitamin C supplement community.  Results: At SMHRC Hospital Nagpur, we examined the relationship between smoking and vitamin C status, dietary and serum vitamin C levels of 100 participants. Smokers of 20 cigarettes a day had the lowest dietary intake of vitamin C and serum levels, whereas smokers of 1-19 cigarettes a day had lower intake of vitamin C and serum levels (compared to respondents who had never smoked. This inverse correlation between vitamin C and smoking intake and serum levels was independent of age, sex, body weight, race, and consumption of alcoholic beverages. The negative association between cigarette smoking and serum vitamin C levels continued, following further adjustment for dietary vitamin C intake. In smokers, the risk of severe hypo-vitaminosis C, particularly when not accompanied by vitamin supplementation, has increased. AbstractMaternal vitamin intake during pregnancy is crucial for pregnancy outcomes and the child's subsequent health. However, there are few valid instruments for assessing vitamin intake that address the effects of nausea and vomiting during pregnancy (NVP). This study aimed to investigate the validity of a FFQ concerning vitamin intake during early and late pregnancy with and without NVP. The participants comprised 200 Japanese pregnant women who completed the FFQ and from whom blood samples were taken in early and late pregnancy. Energy-adjusted dietary vitamin intakes (vitamin C, folate, vitamin B6, vitamin B12, vitamin A, vitamin E and vitamin D) from FFQ were compared with their blood concentrations. A subgroup of women with NVP was investigated. In early pregnancy, significant correlations between FFQ and biomarkers were observed for vitamin C (r0·27), folate (r0·18) and vitamin D (r0·26) in women with NVP and for vitamin A (r0·18), vitamin B12(r0·24) and vitamin D (r0·23) in women without NVP. No significant correlations were observed in either group for vitamins B6or E. In late pregnancy, similar significant associations were observed for vitamin C (r0·27), folate (r0·22), vitamin B6(r0·18), vitamin B12(r0·27) and vitamin A (r0·15); coefficients were higher among women without NVP. Our study demonstrates that the FFQ is a useful tool for assessing intake of several important vitamins in early and late pregnancy regardless of NVP status.
