Abstract Background The health benefits of n-3 polyunsaturated fatty acids (PUFA), such as cardioprotective, anti-inflammatory and hypo-triglyceridemic properties, have been well documented. However, current dietary modification can easily lead to excessive concentrations of n-6 PUFA and an imbalance in the n-6:n-3 PUFA ratio, resulting in the pathogenesis of obesity and related diseases. This study investigated the effects of various dietary n-6:n-3 PUFA ratios on the growth performance, blood lipid and cytokine profiles, tissue fatty acid composition, and gene expression involved in lipid metabolism in finishing pigs. Seventy-two crossbred [(Duroc × Landrace) × Yorkshire] finishing pigs (68.5 ± 1·8 kg) were fed one of four isoenergetic diets with n-6:n-3 PUFA ratios of 2:1, 3:1, 5:1 and 8:1. Results Pigs fed diets with n-6:n-3 PUFA ratios of 3:1 and 5:1 had a higher average daily gain (ADG) and average daily feed intake (ADFI) (P < 0.05) and a lower feed conversion ratio (FCR) (P < 0.05). The concentrations of triglyceride (TG) and total cholesterol (TC) of pigs fed diets with n-6:n-3 PUFA ratios of 1:1 to 5:1 decreased (P < 0·05), whereas the ratio of concentrations of high-density to low-density lipoprotein cholesterol (HDL-C:LDL-C) in pigs fed a diet with an n-6:n-3 PUFA ratio of 2:1 increased (P < 0·05) compared with those fed a diet with an n-6:n-3 PUFA ratio of 8:1. Pigs fed a diet with n-6:n-3 PUFA ratios of 2:1 and 3:1 had lower concentrations of leptin (LEP) and interleukin 6 (IL-6) (P < 0·05), and higher concentrations of adiponectin (APN) (P < 0·05) than those fed diets with n-6:n-3 PUFA ratios of 5:1 and 8:1. Diets with n-6:n-3 PUFA ratios of 2:1 and 3:1 markedly down-regulated the expression levels of peroxisome proliferator-activated receptor-γ (PPARγ) and lipoprotein lipase (LPL) in both the longissimus dorsi muscle (LM) and subcutaneous adipose tissue (SCAT), but up-regulated the expression level of adipocyte fatty acid binding protein (aP2) in the LM. The expression levels of aP2 and hormone- sensitive lipase were similar to those of PPARγ in the SCAT. Conclusions This study demonstrated that dietary n-6:n-3 PUFA ratios between 3:1 and 5:1 beneficially affected growth performance, optimize blood lipid and adipocytokine levels, enhanced absorption and deposition of n-3 PUFA and regulated lipid metabolism-related genes.

The present study evaluated the influence of dietary protein level on growth performance, fatty acid composition, and the expression of lipid metabolic genes in intramuscular adipose tissues from 18- to 23-month-old Hanwoo steers, representing the switching point of the lean-to-fat ratio. Forty steers with an initial live weight of 486 ± 37 kg were assigned to one of two treatment groups fed either a concentrate diet with 14.5% CP and or with 17% CP for 6 months. Biopsy samples of intramuscular tissue were collected to analyze the fatty acid composition and gene expression at 23 months of age. Throughout the entire experimental period, all steers were restrained twice daily to allow individual feeding. Growth performance, blood metabolites, and carcass traits, according to ultrasonic measurements, were not affected by the experimental diets. The high-protein diet significantly increased the expression of intramuscular PPARα (p < 0.1) and LPL (p < 0.05) but did not affect genes involved in fatty acid uptake (CD36 and FABP4) nor lipogenesis (ACACA, FASN, and SCD). In addition, it downregulated intramuscular VLCAD (p < 0.01) related to lipogenesis but also GPAT1 (p = 0.001), DGAT2 (p = 0.016), and SNAP23 (p = 0.057), which are involved in fatty acid esterification and adipocyte size. Hanwoo steers fed a high-protein diet at 18–23 months of age resulted in a relatively lower lipid turnover rate than steers fed a low-protein diet, which could be responsible for shortening the feeding period.

The present study was conducted to investigate the effects of dietary arachidonic acid (ARA) on growth performance, fatty acid composition and ARA metabolism-related gene expression in larval half-smooth tongue sole (Cynoglossus semilaevis). Larvae (35 d after hatching, 54 (sem 1) mg) were fed diets with graded concentrations of ARA (0·01, 0·39, 0·70, 1·07, 1·42 and 2·86 % dry weight) five times per d to apparent satiation for 30 d. Results showed that increased dietary ARA concentration caused a significant non-linear rise to a plateau in survival rate, final body weight and thermal growth coefficient, and the maximum values occurred with the 1·42 % ARA treatment. As dietary ARA increased to 1·07 or 1·42 %, activities of trypsin, leucine aminopeptidase and alkaline phosphatase levels increased, but they decreased with higher ARA concentrations. The fatty acid composition of tongue sole larvae was almost well correlated with their dietary fatty acid profiles, and the EPA content of the larvae decreased with increasing dietary ARA. Meanwhile, the partial sequences of COX-1a (cyclo-oxygenase-1a), COX-1b (cyclo-oxygenase-1b), COX-2 (cyclo-oxygenase-2), 5-LOX (5-lipoxygenase) and CYP2J6-like (cytochrome P450 2J6-like) were also obtained. Both COX-2 and 5-LOX mRNA expression levels significantly increased to a plateau in an ‘L’-shaped manner as dietary ARA increased to 1·07 or 1·42 %, but no significant differences were found in the gene expression of COX-1a, COX-1b or CYP2J6-like. These results suggest that 1·07–1·42 % dietary ARA was beneficial to the growth performance of larval tongue sole, and the regulation of dietary ARA on the growth performance of larvae was probably involved in altering the mRNA expression of COX-2 and 5-LOX.
