Total Phenolic and Flavonoid Content and Antibacterial Properties of Polygonatum orientale Desf and Tilia dasystyla

Background: Secondary metabolites of plants such as phenol and flavonoids can neutralize free radicals. The antioxidant and cytotoxic activities of plants are related to phenolic or flavonoids compounds. The occurrence of drug resistance to antimicrobial drugs has led to the use of medicinal herbs in the treatment of infections. Antibiotic resistant of Staphylococcus aureus has become a major problem in the treatment of diseases. Objectives: The present study aimed to determine total phenolic content (TPC) and total flavonoids content (TFC) of Polygonatum orientale Desf and Tilia dasystyla and evaluate their antibacterial effects on Staphylococcus aureus bacterium. Total phenolic content of Polygonatum orientale Desf and Tilia dasystyla has not been previously investigated. Methods: Total phenolic and flavonoid content of P. orientale Desf and T. dasystyla extracts were determined using colorimetric methods of Folin-Ciocalteu and aluminum chloride. Antimicrobial activities of the extracts were evaluated by microdilution broth and disc diffusion methods to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. Results: The results showed that total flavonoids content of Polygonatum orientale Desf with the value of 7.9 ± 0.040 mg/g DW extracted with diluted water solvent and boiling method and total phenolic contents of Tilia dasystyla with the value of 62.13 ± 0.073 mg/g DW extracted with methanol solvent and boiling method were the highest amount. Methanol extract of Polygonatum orientale Desf had more antibacterial activities with the MBC and MIC values of 0.140 mg/ mL and 8 ± 0.4 mm zone of inhibition. Conclusions: T. dasystyla and P. orientale Desf contain bioactive compounds such as phenolic and flavonoids that can be used as a promising option in pharmacognostical studies for the treatment of S. aureus infections.

Fruits of two jujube (Ziziphus jujuba Mill.) cultivars were collected at four stages of ripening and analyzed in terms of chemical compounds. The cultivars were grown under the climatic conditions of the Sub-Carpathian area Ramnicu-Valcea in Romania (45°6′17″N, 24°22′32″E). Total phenols content, total flavonoids content, organic acids from jujube fruits and related antioxidant activities were investigated during the ripening stages. The antioxidant activity of alcoholic extracts was investigated by the 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method. Total phenolic content was determined by using the Folin-Ciocalteu assay, while determination of flavonoids was done by aluminium nitrate colorimetric method, and organic acids were separated by reversed phase chromatography. The results obtained have indicated significant differences (p < 0.05) found in bioactive compounds and antioxidant activity, depending on cultivars and ripening stage. Total phenolic content ranged from 1634.44 to 472.38 mg GAE/100 g in methanolic extracts of the two varieties studied, while total flavonoids content ranged from 19.9 to 48.5 mg QE/100 g. As regards the influence of ripening stage, the results show that the highest antioxidant activity was recorded in the stage of white maturity. The impact of maturity stage was significant on organic acid distribution of jujube extracts, and ascorbic acid is the major organic acid found in jujube fruits, followed by malic acid and lactic acid. Jujube fruits are rich in bioactive compounds and can be a complement in human’s healthy eating.

In recent years, a substantial amount of evidence has pointed to the key role of free radicals and other oxidants as the main culprits for aging and degenerative diseases associated with aging, such as cancer, cardiovascular diseases, cataract, decline of the immune system and brain dysfunctions. The objective of this work was therefore to detect variations in total phenol and flavonoid content, and in antioxidant activity of samples of V. montevidensis. Dried samples were submitted to extraction to obtain the hexane, ethyl acetate and ethanol fractions. Phytochemical prospecting and an analysis of the phenolic content and antioxidant activity was carried out. The data was analyzed according to the mean±standard deviation and submitted to analysis of variance followed by Tukey's test with a significance of (p < 0.05). Flavonoids, tannins, coumarins, terpenoids, steroids, alkaloids and anthraquinones were detected in the samples. The content of total flavonoids varied between 3.18±0,58 and 7.22±0,17 g/100g, while the total phenols ranged from 1.13±0.16 to 18.44±0.21 g/100g. The extracts produced EC50 between 339.75±6.56 and 278.33±23.11 μg/mL.

The glucosinolate profiles, phenol and flavonoid contents and the antioxidant activity of Eruca vesicaria longirostris were studied for different organs and origins. Eleven desulpho-glucosinolates (DS-GLSs) were isolated and quantified by lipid chromatography- DAD. Similarity between profiles was obtained. Total DS-GLS content, expressed as sinigrin equivalents (SE) revealed a certain variabilily ranging between (76.07-45.61), (27.01-13.53), (4.52 -18.01), (9.39-3.37) and (1.16-13.99) µmol /g DW for seeds, flowers, leaves, roots and stems, respectively. Results showed that seeds are rich in phenolics as they contain highest amounts of phenolics ranging from 27.6±0.5 to 33.47±0.5 mg GAE/g extract as compared to all other parts. Leaves and flowers had a significantly higher total phenolic content than stems and roots in all samples (p < 0.05). According to statistical analysis, the investigated seed extracts with values between (16.20±0.10-18.50±0.10 mg QE/g) exhibited the highest total flavonoids content, followed by leaves (13.00±0.40-15.80±0.30mg QE/g), flowers (10.40±0.40-12.90±0.90 mg QE/g) and stems (7.80±0.20- 9.80±0.70 mg QE/g). Antioxidant activity tested by DPPH, ABTS and FRAP assays, was higher for seeds, leaves and flowers than the other studied organs. These organs were characterized by a significantly high content in glucoerucin, nasturtin and epiprogroitrin, respectively.

  The focus of the present study was to determine the association of the anti-oxidant activity of Costus speciosus (Koen Ex. Retz.) Sm. with the total phe-nolic content and the total flavonoid content. The antioxidant activity of the extracts was quantified by diphenylpicrylhydrazyl (DPPH) assay. The total phenolic content was determined by the Folin-Ciocalteau method and the total flavonoid content was determined by the aluminium chloride method. Among the four extracts of the plant, the ethanol extract exhibited the high-est antioxidant activity and also the highest total phenolic and flavonoid con-tents. A strong correlation was observed between the antioxidant activity and the total phenolic and flavonoid contents. A statistical model was de-rived to explain this dependence and a non-linear association was observed between the antioxidant activity and the total flavonoid content.

Infectious diseases are the second leading cause for worldwide death. Treatment of infections continues to be difficult in modern time because of the severe side effects of some drugs and the growing resistance to antibacterial agents. Over the past few decades the use of antibiotics is under threat as many commonly used antibiotics have become less effective against certain illnesses due to emergence of multi drug-resistant bacteria. In the present study the effects of 3 types of solvents, chloroform, ethyl acetate and methanol were investigated to determine the presence of various phytochemical constituent, total phenolic content, total flavonoids content and in vitro antimicrobial activity from rhizomes of Iris kashmiriana (Kashmir Iris) and Iris ensata (Japanese Iris), belong to family Iridaceae. The reason for selecting in vitro method was to minimize the usage of experimental animals. The antimicrobial activity of chloroform, ethyl acetate and methanol extract of rhizomes of Iris kashmiriana and Iris ensata were evaluated on bacterial strains of Bacillus cereus, Pseudomonas auregenosa, Proteus vulgaris and Eschirichia coli and fungal strains of Candida albicans and Aspergillus niger by agar well diffusion method. The preliminary phytochemical studies and quantitative analysis of alkaloids, phenol and flavonoids were performed by well reported method. These extracts were further subjected to TLC (Thin layer chromatography analysis).  The chemical contents of the Iris kashmiriana and Iris ensata were presented as total phenolic content and total flavonoids content. Phytochemical screening of the extract showed the presence of some common compounds like phenols, terpenoids, flavonoids, carbohydrate etc. The antimicrobial potential of the plant extract was evaluated against different bacterial species which shows significant inhibitory action against all the tested bacterial and fungal strain. Methanolic extract was found to be more active than chloroform and ethyl acetate extracts. It reveals that the methanol soluble components of the plant are highly active against the above mentioned microorganism. Mangifera indica L. belongs to the family Anacardiaceae and is considered as “King of all Fruits”. Mango kernels are discarded as waste after the industrial processing and it has several proven medicinal benefits. Attempts were made to study its antitumour and antimicrobial activities. In the current research work, 15 local cultivars of Mangifera indica L. were collected with a motto to screen the best cultivar having high total phenolic content, flavonoid content, antitumour property and antimicrobial activity. Banganapalli cultivar of mango showed high total phenolic content and total flavonoid content i.e. 63.5±1.1 mg GAE/g and 16.7±0.5 mg quercetin/g followed by Royal special cultivar (TPC-58.7±0.6 mg GAE/g TFC-16.2±0.6 mg quercetin/g). Mangifera indica L. cultivar Banganapalli which showed highest total phenolic content and total flavonoid content was screened for its antitumour and antimicrobial properties. Antitumour property was tested by using potato disc assay which recorded 40.12% tumour inhibition percentage. Antimicrobial activity was assessed by agar diffusion method by taking 3 test microorganisms viz. Bacillus subtilis subsp. subtilis DSM 10, Staphylococcus aureus MTCC 737 and Escherichia coli MTCC 46. The measured area of inhibition is around Staphylococcus aureus MTCC 737 in 8.5±0.3 mm followed by E.coli MTCC 46 (8.2±0.3 mm) and Bacillus subtilis sub subtilis (6.6±0.5 mm). The present study showed that the mango kernels which were generally discarded as waste has antitumour and antibacterial properties and further studies need to be carried out.

Antioxidant activity, total phenolic and flavonoid content of several indigenous species of ferns in East Kalimantan, Indonesia. Biodiversitas 20: 576-580. This study aimed to determine the total phenolic and flavonoids content and antioxidant activity of ethanol extract of several indigenous species of ferns in East Kalimantan. Total phenolic content was determined by Folin-Ciocalteau method and flavonoid content was measured by colorimetric method. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. The result of photochemical screening indicated that the leaves of Plagiogyria pycnophylla, Plagiogyria glauca, and Stenochlaena palustris contained alkaloid, flavonoid, tannin, saponin, and steroid while Acrostichum aureum contained flavonoid, tannin, saponin and steroid. The result showed extract Acrostichum aureum had the highest total phenolic content (366.4573 ± 2.2117 mg GAE.g-1), flavonoid content (228.6087 ± 2.2548 mg QE.g-1), and very strong antioxidant activity with IC50 value 29.5303 ppm. There is positive correlation between total phenolic content, flavonoid with antioxidant activity.

The current investigation is intended to evaluate the content of phytochemical constituents and antioxidant potential of hydroalcoholic extracts of stem and root of Grewia serrulata DC (HAESGS & HAERGS) and leaf and bark of Grewia Nervosa (Lour.) panigrahi (HAELGN & HAEBGN). Initially, all the extracts at different concentrations were estimated for their total phenolic content and total flavonoid content. The study was further extended for their antioxidant potential evaluation using various in vitro methods such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical scavenging assays. The total phenolic content (mg gallic acid equivalent per gram of extract) was high in HAELGN (170.82±0.19) and HAERGS (123.00±0.48) than HAESGS (111.2±0.26) and HAEBGN (119.60±0.23). The total flavonoid content (mg quercetin equivalent per gram) is greater in HAERGS (71.24±0.50) and HAESGS (65.68±0.27) than HAELGN (55.82±0.35) and HAEBGN (62.38±0.45). The IC50 values (µg/ml) of different plant extracts inferred that DPPH radical scavenging activity is greater in HAELGN (42.91±0.88) and HAEBGN (53.87±0.35) than HAESGS (126.73±1.20) and HAERGS (88.87±1.25). However, hydroxyl and superoxide radical scavenging activity is more in HAERGS (135.41±1.19 & 88.00±1.42) and HAELGN (172.28±1.91 & 108.163±1.09) than HAESGS (237.3±1.65 & 110.074±0.87) and HAEBGN (204.7±1.04 & 125.54±1.07). The results of present comprehensive analysis demonstrated that both the plants Grewia serrulata DC and Grewia Nervosa (Lour.) panigrahi possess high phenolic, flavonoid contents and potential antioxidant activity, and could be used as a valid source of natural antioxidants and might be utilized for pharmacological screening of various therapeutic activities.
