In recent years, a substantial amount of evidence has pointed to the key role of free radicals and other oxidants as the main culprits for aging and degenerative diseases associated with aging, such as cancer, cardiovascular diseases, cataract, decline of the immune system and brain dysfunctions. The objective of this work was therefore to detect variations in total phenol and flavonoid content, and in antioxidant activity of samples of V. montevidensis. Dried samples were submitted to extraction to obtain the hexane, ethyl acetate and ethanol fractions. Phytochemical prospecting and an analysis of the phenolic content and antioxidant activity was carried out. The data was analyzed according to the mean±standard deviation and submitted to analysis of variance followed by Tukey's test with a significance of (p < 0.05). Flavonoids, tannins, coumarins, terpenoids, steroids, alkaloids and anthraquinones were detected in the samples. The content of total flavonoids varied between 3.18±0,58 and 7.22±0,17 g/100g, while the total phenols ranged from 1.13±0.16 to 18.44±0.21 g/100g. The extracts produced EC50 between 339.75±6.56 and 278.33±23.11 μg/mL.

Abstract Objective To investigate the effect of cooking temperature and time on the total phenolic content, total flavonoid content and antioxidant activity of aqueous and ethanolic extracts of garlic. Results The mean total phenolic content of fresh garlic were 303.07 ± 6.58 mg gallic acid equivalent per 100 g (GAE/100 g) and 638.96 ± 15.30 mg GAE/100 g of plant material for the aqueous and ethanolic extracts respectively. The mean total flavonoid content 109.78 ± 6.78 mg quercetin equivalent per 100 g (QE/100 g) and 258.47 ± 12.37 QE/100 g for aqueous and ethanolic extracts respectively. Fourier transform infrared spectral data showed absorptions in the range for carboxylic acids, hydroxyl group, esters, and alcohols, confirming the presence of phenols and flavonoids in the extracts. Cooking temperature had a significant effect on total phenolic content and total flavonoid content while cooking time did not have a significant effect on the phytochemicals and antioxidant activity.

  The focus of the present study was to determine the association of the anti-oxidant activity of Costus speciosus (Koen Ex. Retz.) Sm. with the total phe-nolic content and the total flavonoid content. The antioxidant activity of the extracts was quantified by diphenylpicrylhydrazyl (DPPH) assay. The total phenolic content was determined by the Folin-Ciocalteau method and the total flavonoid content was determined by the aluminium chloride method. Among the four extracts of the plant, the ethanol extract exhibited the high-est antioxidant activity and also the highest total phenolic and flavonoid con-tents. A strong correlation was observed between the antioxidant activity and the total phenolic and flavonoid contents. A statistical model was de-rived to explain this dependence and a non-linear association was observed between the antioxidant activity and the total flavonoid content.

This experiment aims to determine the correlation of total phenolic and flavonoid content of jati putih leaves fraction (Gmelina arborea Roxb.) towards Antioxidant activity .  Sample was extracted by maceration method using ethanol 70% to obtain the ethanol extract (EE), followed by liquid-liquid extraction method to obtain fraction of ethyl acetate (EA) and n-Hexane (EH). The phytochemical screening  and determination of total phenolic and flavonoid content were done by colorimetric method. Antioxidant activity were done by DPPH, FRAP and ABTS methods. Phytochemical screening showed positive results for flavonoids, phenolic and saponins.  The largest total phenolic content was found on EA (11,59 µg/ml ± 0,3 %b/b EAG) and the largest total flavonoid content was on EA (3,88 µg/ml ± 0,02 %b/b EK). The total phenolic and flavonoid content of Jati putih leaves has a correlation with antioxidant activity. The coefficient correlation of activity on reducingDPPH radical was 56,7% (total of phenolic content) and 57,8% (total of flavonoid content) and on iron reduction power in FRAP method  was 99,9% (total of phenolics and flavonoids content). The relationship with the activity in reducing radical ABTS obtained coefficient correlation of 57,0% and 58,1% for total phenolic and flavonoids contents, respectively.

Antioxidant activity, total phenolic and flavonoid content of several indigenous species of ferns in East Kalimantan, Indonesia. Biodiversitas 20: 576-580. This study aimed to determine the total phenolic and flavonoids content and antioxidant activity of ethanol extract of several indigenous species of ferns in East Kalimantan. Total phenolic content was determined by Folin-Ciocalteau method and flavonoid content was measured by colorimetric method. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. The result of photochemical screening indicated that the leaves of Plagiogyria pycnophylla, Plagiogyria glauca, and Stenochlaena palustris contained alkaloid, flavonoid, tannin, saponin, and steroid while Acrostichum aureum contained flavonoid, tannin, saponin and steroid. The result showed extract Acrostichum aureum had the highest total phenolic content (366.4573 ± 2.2117 mg GAE.g-1), flavonoid content (228.6087 ± 2.2548 mg QE.g-1), and very strong antioxidant activity with IC50 value 29.5303 ppm. There is positive correlation between total phenolic content, flavonoid with antioxidant activity.

The influence of extraction techniques with different solvents on the contents of total phenols, flavonoids and antioxidant activity of extracts of grape, sour cherry and black currant is analyzed in the study. Antioxidant activity of the extracts on stable 1,1-diphenyl-2-picryl hidrazyl (DPPH) radical was determined by spectrophotometry. The total phenolic content was determined by using the Folin-Ciocalteu assay while total flavonoid content was measured spectrophotometrically with the application of AlCl3. The changes in the value of these parameters after deep-freezing of these fruits were also observed. A high content of total phenols and flavonoids was detected in extracts. Acetone proved to be the best agent for extraction of grape and sour cherry while the highest content of total phenols with black currant was obtained by methanol. The content of total phenols in fruits in frozen state increased while the content of total flavonoids and of antioxidant activity decreased. The obtained results show that the extracts of grape, cherry and black currant can be used as natural antioxidants.

The present study aimed to compare antioxidant activities, total phenolic and flavonoid content present in leaves and bark of Terminalia arjuna and check whether there is any correlation between phenolic content and flavonoid content with antioxidant activities or not. Phytochemical screening of ethanolic extract of leaves and bark revealed the presence of phenols, flavonoids, tannins, carbohydrates, glycosides, saponin, phytosterols and phytosteroids. Total phenolic content was found to be higher in bark (272.71±3.18 mg/g gallic acid equivalents) as compared to that of leaves (95±3.11 mg/g gallic acid equivalents). Similarly, flavonoid content of ethanolic extract of bark was found to be higher (203.95±5.13 mg/g rutin equivalents) than that of leaves (87.625±4.28 mg/g rutin equivalents). DPPH activity of ethanolic extract of bark (IC50-17.41 µg/ml) was more than that of leaves (IC50-20.22µg/ml). FRAP activity of bark (IC50- 4.781 µM Fe (II) equivalents) is more than that of leaves (IC50-7.572 µM Fe (II) equivalents). Nitric oxide (NO) scavenging activity of bark (IC50-12.87 µg/ml) was higher than that of leaves (IC50-13.91 µg/ml).The present study clearly showed that there is a correlation between total phenolics, flavonoid contents and antioxidant activity of leaves and bark of T. arjuna. The phenolic compounds and flavonoids could be major contributor to antioxidant activity of T. arjuna. Keywords: Terminalia arjuna, DPPH, FRAP, NO, IC50, Antioxidants

Background: Secondary metabolites of plants such as phenol and flavonoids can neutralize free radicals. The antioxidant and cytotoxic activities of plants are related to phenolic or flavonoids compounds. The occurrence of drug resistance to antimicrobial drugs has led to the use of medicinal herbs in the treatment of infections. Antibiotic resistant of Staphylococcus aureus has become a major problem in the treatment of diseases. Objectives: The present study aimed to determine total phenolic content (TPC) and total flavonoids content (TFC) of Polygonatum orientale Desf and Tilia dasystyla and evaluate their antibacterial effects on Staphylococcus aureus bacterium. Total phenolic content of Polygonatum orientale Desf and Tilia dasystyla has not been previously investigated. Methods: Total phenolic and flavonoid content of P. orientale Desf and T. dasystyla extracts were determined using colorimetric methods of Folin-Ciocalteu and aluminum chloride. Antimicrobial activities of the extracts were evaluated by microdilution broth and disc diffusion methods to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. Results: The results showed that total flavonoids content of Polygonatum orientale Desf with the value of 7.9 ± 0.040 mg/g DW extracted with diluted water solvent and boiling method and total phenolic contents of Tilia dasystyla with the value of 62.13 ± 0.073 mg/g DW extracted with methanol solvent and boiling method were the highest amount. Methanol extract of Polygonatum orientale Desf had more antibacterial activities with the MBC and MIC values of 0.140 mg/ mL and 8 ± 0.4 mm zone of inhibition. Conclusions: T. dasystyla and P. orientale Desf contain bioactive compounds such as phenolic and flavonoids that can be used as a promising option in pharmacognostical studies for the treatment of S. aureus infections.

Buas-buas, meniran, secang, and rosella have biological and pharmacological activities as antioxidants. The combination of the four plants is expected to provide a more potent synergistic effect on antioxidant activity. The purpose of this study was to analyze the total phenol content, total flavonoids, and antioxidant effects before and after combination. The combination of extracts, buas-buas, meniran, secang, and rosella which are used in sequence is (1: 1: 1/2: 1/2), (1: 2: 1/2: 1/2), and (2: 1 : 1/2: 1/2). Plants used in the form of simplicia was extracted by maceration method. Radical capture activity uses DPPH and IC50 values are determined. Determination of total phenol is expressed equivalent to gallic acid. Total flavonoids are expressed as quercetin equivalents. The phenol and flavonoid content obtained are then correlated with antiradical activity. The results showed that the best IC50 values were in the combination of ratios (1: 1: 1/2: 1/2) that is (11.0 µg / mL), then (1: 2: 1/2: 1/2) which was 13.3 µg / mL, and (2: 1: 1/2: 1/2) which is 19.4 µg / mL. The highest total phenolic and flavonoid content in the ratio (1: 2: 1/2: 1/2) is 33.57% w/w EAG and 74.00% w/w EQ. Correlation analysis between IC50 values with total phenolic and flavonoid contents showed a positive correlation with R2 values of 0.8236 and 0.0102 with positive slope. Thus, it can be concluded that the total phenol content influences free radical scavenging activity by 82.36%, while the effect of total flavonoid content was only 1.02%.

Dendrobium amoenum has biologically active phytoconstituents valued for traditional Chinese or folk medicine as tonic. Total phenolic and flavonoid contents of its stem extract was estimated by using Folin-Ciocalteu’s reagent and Aluminium chloride methods respectively. Antioxidant activity was determined by DPPH free radical scavenging method. Total phenolic content found in acetone (134.34 μg GAE/mg extract) and chloroform (101.55 μg GAE/mg extract) extract was significantly higher than other solvent extracts. Similarly, these two extracts had significantly high flavonoid content (acetone: 115.73 μg QE/mg extract, and chloroform: 84.16 μg QE/mg extract). Presence of high phenolic and flavonoid contents in these extracts showed the highest antioxidant activity. Highest antioxidant activity of these extracts was determined by their lowest IC50 value (acetone: 53.19 μg/ml and chloroform: 36.48 μg/ml). Significant negative relationship was found between phenolic content and IC50 (R2 = 0.209, p < 0.01) and flavonoid content and IC50 (R2 = 0.389, p < 0.01), which indicates high antioxidant activity due to high phenolic and flavonoid contents. This result revealed that D. amoenum act as an antioxidant agent due to its free radical scavenging activity which plays a crucial role in the development of new chemotherapeutic agents.

Latar Belakang: Antioksidan adalah senyawa yang mampu menangkal atau meredam efek negatif oksidan dalam tubuh, bekerja dengan cara mendonorkan satu elektronnya kepada senyawa yang bersifat oksidan sehingga aktivitas senyawa oksidan tersebut dapat dihambat. Herba poguntano (Picria fel-terrae Lour.) digunakan sebagai obat antidiabetes dan telah dilaporkan mengandung senyawa kimia yang terdiri dari alkaloid, saponin, flavonoid, dan polifenol yang merupakan metabolit sekunder yang bersifat antioksidan. Tujuan Penelitian: Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan serta mengetahui kandungan fenolat dan flavonoid total dari ekstrak etanol herba poguntano. Metode Penelitian: Ekstrak diperoleh secara maserasi dengan pelarut etanol 96%, lalu dipekatkan dengan alat rotary evaporator hingga didapatkan ekstrak kental. Ekstrak diuji dengan metode pemerangkapan radikal bebas DPPH (1,1-diphenyl-2-picrylhydrazyl) serta mengukur kandungan fenolat total dan flavonoid total dengan mengukur absorbansi menggunakan spektrofotometri uv-vis yang diinkubasi berturut-turut selama 60, 90 dan 40 menit pada panjang gelombang 516, 775 dan 432 nm. Hasil: Hasil pengukuran aktivitas antioksidan dengan metode pemerangkapan radikal bebas DPPH dari ekstrak etanol herba poguntano menunjukkan kekuatan antioksidan dalam kategori sangan kuat dengan nilai IC50 21.79 ppm dan kuersetin menunjukkan kekuatan antioksidan dalam kategori sangat kuat dengan nilai IC50 4.9 ppm. Hasil pengukuran kandungan fenolat total dengan metode Folin-Ciocalteau menunjukkan angka 66.29 mg GAE/g ekstrak serta hasil pengukuran kandungan flavonoid total menunjukkan angka 19.26 mg QE/g ekstrak. Kesimpulan: Ekstrak etanol herba poguntano mempunyai aktivitas antioksidan dengan kategori sangat kuat serta memiliki kandungan fenolat total yang tinggi dibandingkan dengan kandungan flavonoid total. Background: Antioxidants are compounds capable of counteracting or reducing the negative effects of oxidants in the body. Antioxidants work by donating an electron to an oxidant compound so that the activity of the oxidant compound can be inhibited. Poguntano (Picriafel-terrae Lour) is used as an antidiabetic drug and has been reported to contain chemical compounds, , as secondary metabolites that have antioxidant properties. Objective: This study aimed to determine antioxidant activity and total phenolic and flavonoid content of the ethanol extract of poguntano. Research Method: . The dried materials were macerated using 96% ethanol and then it was concentrated using rotary evaporator to obtain crude extract. The extract was tested by DPPH free radical scavenging method (1,1-diphenyl-2-picrylhydrazyl) and the total phenolic and flavonoid content was determinedby measuring absorbance using uv-vis spectrophotometry which was incubated successively for 60, 90 and 40 minutes at wavelength 516, 775 and 432 nm. Results: The evaluationof antioxidant activity using DPPH free radical scavenging method ofethanol extract of poguntano showed the antioxidant activitywas in very strong category with IC50 value of 21.79 ppm and quercetin showed the antioxidant activitywas in very strong category with IC50 value of 4.9 ppm. The result of the measurement of total phenolic content by the Folin-Ciocalteu method was66.29 mg GAE/g extract and the result of measurements of total flavonoid content was19.26 mg QE/g extract. Conclusion: Ethanol extract of poguntanohas antioxidant activity in a very strong category and it has higher total phenolic content compared with total flavonoid content.
