Objective: This study was aimed to evaluate the antioxidant potential of sinapic acid in both in vitro and in vivo. Recently, we have reported that oral administration of sinapic acid (3,5-dimethoxy 4-hydroxycinnamic acid) an active phyto ingredient widely distributed in rye, mustard, berries, and vegetables has been shown to ameliorate hyperglycemia.Methods: Experimental Type 2 diabetes was induced in male Wistar rats by feeding high-fat diet to induce insulin resistance followed by intraperitoneal administration of a single low dose streptozotocin (35 mg/kg body weight [bw]). Sinapic acid was administered orally at a concentration of 25 mg/kg bw/rat/day for 30 days, and its efficacy was compared with metformin. In vitro, antioxidant scavenging properties of sinapic acid were determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), superoxide, and nitric oxide (NO) assay.Results: Sinapic acid treatment showed a significant decline in the levels of lipid peroxides, hydroperoxides and protein carbonyls in the plasma and vital tissues of diabetic rats. The treatment also improved the antioxidant status in diabetic rats indicating the antioxidant potential of sinapic acid. In addition, the results of DPPH, ABTS, superoxide, and NO radical scavenging assays substantiate the free radical scavenging efficacy of sinapic acid.Conclusion: The results of this study evidenced that sinapic acid possess significant antioxidant properties which in turn may be responsible for its antidiabetic properties.

Diabetes is characterized by hyperglycaemia with alterations of lipid parameters. The effects of Aloe vera (A.V) gel extract on lipid profiles of streptozotocin-induced diabetic rats were investigated. Seventy-two male albino rats were randomly divided into six groups of 12 rats each; control, diabetic control, 4 ml/kg and 8 ml/kg body weight (b.w.) A.V, 10 mg/kg b.w. glibenclimide and non-diabetic 8 ml/kg b.w. A.V groups. They were treated for 14 and 28 days. The plasma and HDL lipid profiles (triacylglycerols, cholesterol and phospholipids) of the diabetic rats were significantly (p<0.05) increased, while those of the VLDL+LDL, RBC, RBC membrane and organs were observed to be significantly (p<0.05) reduced, when compared to the control after 14 and 28 days of treatment. A.V gel extract significantly (p<0.05) ameliorated the disruptions observed in the lipid profiles of the diabetic and thus provide a scientific rationale for the use of Aloe vera as a therapeutic agent.Bangladesh J Med Biochem 2016; 9(1): 11-26

Abstract Background Cardiovascular disease is the most life-threatening diabetic complication. Type II diabetes may lead to damage of the heart muscle. Sodium glucose cotransporter 2 (SGL2) inhibitors are a new class of diabetic medications indicated only for the treatment of type II diabetes. Aim This investigation was assigned to compare the effect of metformin and SGL2 inhibitors (dapaglifilizone) in type II diabetic rats. Design and methods: Eighty rats divided into four groups were used: non diabetic; diabetic; diabetic metformin -treated; diabetic dapagliflizoline- treated. At the end, arterial blood pressure and cardiac performance (cardiac contractility and heart rate) were assessed. Serum glucose, glycosylated hemoglobin (HbA1c), insulin, lipid profile, total antioxidant capacity, malondialdehyde, tumor necrosis factor α were measured. HOMA-IR index was calculated. DNA changes were assessed from hearts and aortea. Aortic endothelial changes recorded using H&E and masson trichome techniques. Results Glycemic index, lipid profile, oxidative stress and inflammatory parameters were significantly improved in both metformin and dapagliflizoline treated groups with also significant improvement in blood pressure, Cardiac performance and reduction in collagen deposition in aortic tissue and DNA fragmentation. Dapagliflizoline treatment results were significantly improved in all parameters compared to metformin treatment. Conclusion SGL2 inhibitors (dapaglifilizone) successfully restored glycemic state, cardiac performance, DNA and endothelial changes in type II diabetic rats compared to metformin.
