The present investigation entitled “Allelic diversity in apple germplasm for fruit quality and scab resistance using SSR markers” was conducted in division of plant biotechnology, SKUAST-K, Shalimar during the year 2018. In the present study, four SSR markers namely Md-ACS-1, Md-ACO-1, ALO-7 and AM-19 were used for screening of 40 apple cultivars presently under cultivation in SKUAST-K for quality and scab resistance. The results revealed that out of 40 cultivars evaluated, 3 were found homozygous (ACS-1-2/2), 17 were heterozygous (ACS-1-1/2) and 10 were homozygous (ACS-1-1/1), while as no amplification of ACS-1 was found for 10 cultivars. Similarly, for Md-ACO-1, 3 cultivars were found homozygous, 35 as heterozygous and 2 did not show any amplification. As far as presence of Vf gene conferring resistance against scab disease, ALO-7 amplified at 820bp and 570bp alleles, 570 bp fragment was observed in resistant genotypes and was absent in susceptible genotypes, while the 820 amplicon was observed in both resistant and susceptible genotypes. The markers AM19 led to the amplification of 520 bp fragment in few cultivars.

A DNA marker-based study was undertaken to identify the occurrence of major scab resistance genes in some apple cultivars and selections of importance for apple breeding. Unfortunately none of the RAPD-based markers previously reported to detect the Va, Vb, Vr and Vx genes produced unambiguous results. By contrast, the CAPS marker MI8 produced the expected three bands in all cultivars and selections already known or suspected to have the Vf gene, as well as in the Russian cultivar 'Antonovka Polotora Funtovaja' suspected to have Va resistance which however may be allelic to Vf. Vf-carrying selections and newly named cultivars 'Frida' and 'Fredrik' are grown successfully in Sweden without fungicides, suggesting that the Vr resistance breaking scab races 6 and 7 have not yet become a problem. The SCAR marker B12 detected the Vm gene in 'Prairifire', 'Rouville', clones 'OR45T132' and 'OR48T70', and selection '16-36-193'. The SSR locus 0102b10 detected one band at 118 by in 'Reka'. This is presumed to be identical to the Vr gene marker previously reported.

ABSTRACTThe production of high quality fruits is a necessary factor for the adaptation and production of plant species with economic viability. Thus, an experiment was conducted from July 2012 to January 2013 to evaluate the fruit quality of the ‘Eva’ and ‘Princesa’ apple cultivars as a function of nitrogen fertilization in Petrolina, PE, Brazil. The experimental design consisted of randomized blocks, with treatments distributed in a factorial arrangement 2 x 4, corresponding to apple cultivars (Eva and Princesa) and nitrogen doses (40; 80; 120 and 160 kg of N ha-1), with four replications and three plants in each plot. The fruit characteristics, such as fruit mass, skin color (luminosity, chromaticity, and colour angle), size (width and length), pulp firmness, titratable acidity (TA), soluble solids (SS) and the SS/TA ratio, were recorded. Nitrogen doses do not affect fruit quality of studied apple cultivars. The fruit quality attributes are different between apple cultivars: fruit firmness, SS/TA ratio, fruit mass and fruit diameter are superior for Princesa cultivar, while the fruit length for Eva cultivar is superior.

The evolutionary diversity of wild potato species makes them excellent materials for improving the narrow genetic basis of the cultivated potato Solanum tuberosum. Understanding their genetic diversity is important not only to choose the best parents for breeding, but also to design proper crossing schemes and selection strategies. The objectives of this study were to determine the resistance response to Ralstonia solanacearum, Potato virus Y and low temperatures of 21 clones of 12 potato species, and to determine their genetic diversity through simple sequence repeat (SSR) markers. Sources of resistance have been found for all the investigated traits, with high resistance variability not only between but also within species. Combined resistances were also identified, with positive implications for efficient breeding. SSR analysis allowed the detection of 12 loci and 46 alleles across all genotypes, with an average value of 3.8 alleles per locus. Both unique and rare alleles useful for marker-assisted selection were found. SSR-based cluster analysis revealed that resistant genotypes were distributed among all clusters, suggesting that genetically different resistant genotypes were identified. The information obtained in this study is discussed from a breeding perspective.

The effect of row direction (N-S, E-W), plant density (conventional and intensive) and rootstock (M9, MM106 and crab apple) on the sensitivity of 41 apple cultivars to sunburn injury was studied at 6 locations (Derecske, Kálmánáaza, Nagykutas, Nagylapos, Siófok, Tornyospálca). During the observations the cultivars had rootstock-specific properties in respect of sunburn-susceptibility. Accordingly, the injury was decreased in the order: M9, MM106 and seedling rootstocks. The differences in sensitivity depend on the canopy of trees, caused finally by the growing vigour of rootstocks. Accordingly, the highest value of sunburn injury was observed on M9 rootstock, because this rootstock has a dwarfing effect upon the scion cultivars. Thus, vegetative area of these trees grow very slowly and the foliage is not enough compact to protect the fruit from solar radiation. The size and density of the foliage increased in the order: M9, MM 106 and crab apple seedling as rootstock. Moreover, relationships were demonstrated between the diameter of upper part of the crown, the leaf area, the number of fruits per tree and the injury of sunburn. Authors categorized the cultivars in respect of values of sunburn incidence: 1. „Not sensitive", II. „Moderately sensitive" and HI. „Strongly sensitive" categories were constituted. Generally, Topaz and Gala cultivars showed low damage (or were free from symptoms), in contrast to this, Golden mutants suffered relatively much. The most sensitive cultivar was Jonica on all the three rootstocks. We searched for relationship between the fruit quality parameters and the frequency of sunburn. Significant correlation was found in the cases of fruit size and the extent of cover-colour. The latter is interpreted with the fact that the best coloured fruits are found on the periphery of crown as a consequence of more intense irradiation.

The aim of the present study was to determine the effect of nitrogen supply on yield and fruit quality of apple cultivars and to explore the relationships between canopy density and the different fruit quality parameters. The experiments were carried out at Kálmánháza, in Eastern Hungary in a private orchard in 2003-2004. The response of four apple cultivars ('Elstar', 'Gala Prince', 'Granny Smith', 'Idared') to different nitrogen dosages was studied. The assessed and calculated indices were: yield, fruit diameter, fruit height, shape index, fruit mass, firmness, dry matter content and colouration. The results indicated that nitrogen fertilization has a significant effect on the yield and fruit quality of apple cultivars. The calculated specific yield values were reduced by the application of nitrogen via the increase in the volume of the canopy. An opposit trend was observed for fruit diameter, fruit height and fruit weight, which increased with increasing nitrogen supply. The reduced shape index caused flattening of fruits. However, the improvement of fruit quality via increasing nitrogen dosage is only virtual, since these dosages increase the fruit size, but firmness, dry matter content and colouration are diminished, which decreases the value of the fruits on the market. The authors also studied the relationships between canopy density responsible for assimilation and light supply of the fruits and the different fruit quality parameters. The closest linear inversely proportional relationship was found in the case of colouration. There was a negative linear relationship between canopy density and firmness or dry matter content. The relationship between canopy density and fruit mass could be described by a quadratic polinomial function.

Apple cultivars were crossed according to the half-diallel and topcross mating design. Apple scab resistance of seedlings was estimated by using a 0–5 scale: 0 – no infection on leaves, 5 – more than 75% of leaf area infected. Percentage of scab resistant seedlings in crosses involving cultivars Prima varied from 35.8% to 67.6%, involving cultivars Florina and Arbat – 51.9–59.5% and 53.5–72.5%, respectively. Significant differences for resistance to scab were found among the crosses. Both general combining ability (GCA) and specific combining ability (SCA) were highly significant. The significance of GCA for scab resistance shows that genetic variability exists for this trait and selection should result in genetic progress, because the mean square for GCA was larger than that calculated for SCA. Therefore, selection of parents on its phenotype should be effective for development of resistant apple cultivars. The significant SCA estimates indicate that a large number of resistant seedlings from particular crosses should be selected.
