AbstractLittle information is available on vegetative and generative performance of apple cultivars during the early growth of trees in orchards. The aim of this five-year study was to evaluate 2 vegetative (trunk cross sectional area (TCSA) and tree height (TH)) and 7 generative parameters (tree yield (TY), fruit number per tree (FNT), crop load (CL), fruit diameter (FD), shape index (SI), fruit surface color (FSC) and fruit color intensity (FCI)) and their inter-correlations (Pearson correlation, regression analyses and PCA) for young apple trees, on 9 apple cultivars (‘Jugala’, ‘Galaval’, ‘Gala Venus Fengal’, ‘Gala Decarli-Fendeca’, ‘Gala Schnitzer (S) Schniga’, ‘Fuji September Wonder’, ‘Crimson Crisp (Co-op 39)’, ‘Jeromine’, and ‘Red Idared’) in a slender spindle training system (2597 trees ha−1) and on 4 apple cultivars (‘Wilton’s Red Jonaprince’, ‘Red Cap Valtod (S)’, ‘Early Red One’, and ‘Red Topaz’) in a super spindle training system (5194 trees ha−1) in Eastern Hungary. The strongest vegetative growth was observed in ‘Red Idared’, while the weakest was in ‘Early Red One’. Most ‘Gala’ mutants showed high yields in all years, except for ‘Galaval’. On the 6 year-old trees, the lowest tree yield was found in ‘Fuji September Wonder’ (8.2 kg tree−1), while the highest was found in ‘Gala Venus Fengal’ (35.8 kg tree−1). The lowest fruit number per tree (15 fruit tree−1) was found in ‘Jeromine’, while the highest (222 fruit tree−1) was in ‘Gala Venus Fengal’. The highest crop load was found in ‘Gala Venus Fengal’ (12.72 fruit per cm2 TCSA), while the lowest was in ‘Jeromine’ (2.13 fruit per cm2 TCSA). The smallest fruit diameter (66.3 mm) was recorded in ‘Gala Schnitzer (S) Schniga’, while highest (93.6 mm) was in ‘Red Idared’. The lowest shape index (0.73) was found in ‘Red Topaz’, while the highest (0.92) was in ‘Red Idared’. The majority of the cultivars reached very good fruit surface color (80–100%). The lowest fruit surface color (40%) was observed in ‘Gala Schnitzer (S) Schniga’, while the highest (100%) was in ‘Jeromine’ and ‘Early Red One’. The highest fruit color intensity was observed in most cultivars with the exception of ‘Jeromine’, ‘Gala Schnitzer (S) Schniga’ and’Fuji September Wonder’. In addition, correlation and regression analyses revealed strong and significant (p = 0.05) relationships between TH vs TCSA, TY vs TCSA, TH vs TY, TY vs FNT, and FCI vs FSC. PCA explained 87% of the total variance and PC1, PC2, PC3, and PC4 accounted for 33, 21, 20, and 13% of the variance, respectively, and correlated with TSCA, TH, TY and FNT; with FNT, CL and FS; with FSC and FCI; and with TH and SI, respectively. In conclusion, our study provides useful tree property data on prospective mutants/cultivars for growers/advisors in order to select the most suitable cultivars for establishing new orchards under climate conditions similar to central Europe.

Apples are seasonal fruits, and it is important to prepare them adequately for storage and ensure proper storage conditions. In this study, we used ten different apple cultivars: ‘Alva’, ‘Auksis’, ‘Connell Red’, ‘Cortland’, ‘Ligol’, ‘Lodel’, ‘Noris’, ‘Rubin’, ‘Sampion’, and ‘Spartan’. We studied the qualitative and quantitative composition of phenolic compounds in the apple and apple extracts antioxidants activity before placing them in the controlled atmosphere chambers and again at the end of the experiment, eight months later. Different concentrations of O2, CO2, and N2, constant temperature, relative humidity, and removal of endogenous ethylene were continually maintained. HPLC analysis showed that the highest amount of 2265.7 ± 152.5 µg/g of chlorogenic acid was found in apple samples of the ‘Auksis’ cultivar stored under variant IV conditions. Different concentrations of gas in the controlled atmosphere chambers caused changes in antioxidant activity in whole apple and apple peel extracts. In our study, we found that the antioxidant activity of apple extracts varied between samples of different apple cultivars and depended on the composition of the controlled atmosphere. Determining the optimal storage conditions is beneficial to providing the consumers with apples that have a known and minimally altered chemical composition of phenolic compounds and the strongest antioxidant activity, which determine the use of apples in the healthy food chain.

The large number of cultivars belonging to the cultivated apple (Malus × domestica Borkh.) reflects an extremely wide range of variability, including for fruit quality traits. To evaluate some characteristics of fruit quality, 22 apple genotypes were selected from a collection of germplasms containing more than 600 accessions, based on different considerations, including the use of fruits (dessert, cooking, processing, juice, cider, multipurpose). The mean water content of the studied apple genotypes was 85.05%, with a coefficient of variation (CV) of 2.74%; the mean ash content was 2.32% with a CV of 22.1%, and the mean total soluble solids was 16.22% with a CV of 17.78%, indicating a relatively small difference between genotypes for these indices. On the contrary, relatively large differences were registered between genotypes for fruit weight, volume, and titratable acidity with means of 119.52 g, 155 mL, and 0.55% malic acid, and CVs of 35.17%, 34.58%, and 54.3%, respectively. The results showed that peel hardness varied between 3.80 and 13.69 N, the toughness between 0.2 and 1.07 mm, the flesh hardness between 0.97 and 4.76 N, and the hardness work between 6.88 and 27.84 mJ. The current study can emphasize the possibility of choosing the appropriate apple cultivars to cross in the breeding process and how future strategies can help apple breeders select breeding parents, which are essential key steps when breeding new apple cultivars. In addition, multivariate analysis has proven to be a useful tool in assessing the relationships between Malus genetic resources.

Fire blight, affecting more than one hundred and thirty species in the Rosaceae, is probably the most destructive disease affecting pear and apple cultivars in many countries. Currently, there are no effective synthetic compounds with systemic properties. Other major problem is the occurrence and spread of strains of Erwinia amylovora with resistance to streptomycin and copper. Taken into consideration the human and environmental health, the use of biocontrol agents either as an alternative or as a supplement within an integrated fire blight management strategy has attracted worldwide attention. In this study, E. amylovora solution of 107 CFU ml-1 was treated with bio-control agents, Bacillus subtilis str. QST 713, B. amyloliquefaciens str. MBI 600 and their mixture (at solution densities of 106, 107 and 108 CFU ml-1 for each one) on Petri dishes, containing King’s B medium and, compared with positive (streptomycin sulphate) and negative (sterile distilled water) controls. In vivo studies were performed on two-year-old apple cv. Gala seedlings grown in 45-cm-diameter pots containing a sterilized mix of soil–sand–peat under controlled greenhouse conditions (85% relative humidity, 25°C temperature and 16h of day light). The plants were irrigated as needed by drip-irrigation and each pot received a mineral solution (NPK: 20–20–20) at 2 g l-1 twice. When plant shoots reached a length of 30-35 cm, bio-control agents, individually and their mixture, were applied to the plants by a hand-sprayer. Obtaining the data, 108 CFU ml-1 of Bacillus spp. suspension mixture showed strongest in vitro antibacterial effect (26mm) among the tested treatments after positive control streptomycin (28.6mm). Parallel to in vitro findings, the mixture was most effective against the pathogen on cv. Gala (66.03%). Findings show that the use of mixture of beneficial microorganisms with individual antagonistic properties against the pathogen can be an effective strategy as a natural alternative to agrochemicals in the scope of good agriculture practices.

Irrigation water productivity is a priority for sustainable orchard management as water resources become more limiting. This study evaluated reduced irrigation (RI) as a management strategy for cider apple (Malus domestica Borkh.) production in 2019 and 2020 in northwestern Washington, which has a Mediterranean climate and averages 14.1 cm of precipitation from June to September. RI was evaluated on three cider apple cultivars, Dabinett, Porter’s Perfection, and Golden Russet, in their third and fourth leaf. Stem water potential (stem ψ) was measured weekly throughout the growing season to monitor water stress and implement the RI treatment: irrigation would be applied when stem ψ values dropped below −1.5 MPa, a threshold indicative of moderate water stress in apples. Soil water potential was monitored throughout the season, vegetative growth was assessed by measuring shoot length and non-destructive imaging of the plant canopy using lateral photography, and yield, fruit quality, and juice quality were measured at harvest. Moderate water stress as indicated by stem ψ did not occur either year, thus irrigation was never applied to the RI treatment plots. There was a negative relationship between average stem ψ and both yield and air temperature (P < 0.0001 for each); as yield increased by 5.9 kg per tree or temperature increased by 3.3 °C, stem ψ decreased by 0.1 MPa. The juice quality attributes of the three cultivars in this study were similar to their historic measures at this site and there were no differences due to irrigation treatment, likely because trees did not reach the threshold to induce physiological stress. Both years, trees in the RI treatment did not differ from the control treatment in vegetative growth, fruit yield, juice yield, or any juice quality attribute, but weight per fruit decreased by 7 g, and fruit firmness (measured only in 2020) increased by 2 N. Results from this study indicate that fruit yield and quality in an establishing orchard can be maintained when irrigation is reduced relative to crop water requirements that are estimated from a calculated water balance or relative to conventional grower practices for this region. This finding highlights the benefits of using plant water status to schedule irrigation.

Drought stress is a significant environmental factor limiting crop growth worldwide. Malus prunifolia is an important apple species endemic to China and is used for apple cultivars and rootstocks with great drought tolerance. N6-methyladenosine (m6A) is a common epigenetic modification on messenger RNAs (mRNAs) in eukaryotes which is critical for various biological processes. However, there are no reports on m6A methylation in apple response to drought stress. Here, we assessed the m6A landscape of M. prunifolia seedlings in response to drought and analyzed the association between m6A modification and transcript expression. In total, we found 19,783 and 19,609 significant m6A peaks in the control and drought treatment groups, respectively, and discovered a UGUAH (H: A/U/C) motif. In M. prunifolia, under both control and drought conditions, peaks were highly enriched in the 3′ untranslated region (UTR) and coding sequence (CDS). Among 4204 significant differential m6A peaks in drought-treated M. prunifolia compared to control-treated M. prunifolia, 4158 genes with m6A modification were identified. Interestingly, a large number of hypermethylated peaks (4069) were stimulated by drought treatment compared to hypomethylation. Among the hypermethylated peak-related genes, 972 and 1238 differentially expressed genes (DEGs) were up- and down-regulated in response to drought, respectively. Gene ontology (GO) analyses of differential m6A-modified genes revealed that GO slims related to RNA processing, epigenetic regulation, and stress tolerance were significantly enriched. The m6A modification landscape depicted in this study sheds light on the epigenetic regulation of M. prunifolia in response to drought stress and indicates new directions for the breeding of drought-tolerant apple trees.

Food authentication is very important to protect consumers, sellers, and producers from fraud. Although several methods have been developed using a wide range of analytical techniques, most of them require sample destruction and do not allow in situ sampling or analysis, nor reliable quantification of hundreds of molecules at the same time. To overcome these limitations, we have developed and validated a new noninvasive analytical workflow for food authentication. The method uses a functionalized strip to adsorb small molecules from the surface of the food product, followed by gas chromatography–mass spectrometry analysis of the desorbed analytes. We validated the method and applied it to the classification of five different apple varieties. Molecular concentrations obtained from the analysis of 44 apples were used to identify markers for apple cultivars or, in combination with machine learning techniques, to perform cultivar classification. The overall reproducibility of the method was very good, showing a good coefficient of variation for both targeted and untargeted analysis. The approach was able to correctly classify all samples. In addition, the method was also used to detect pesticides and the following molecules were found in almost all samples: chlorpyrifos-methyl, deltamethrin, and malathion. The proposed approach not only showed very good analytical performance, but also proved to be suitable for noninvasive food authentication and pesticide residue analysis.

In the framework of the program of creating nomenclatural standards of domestic cultivars initiated by the N.I. Vavilov All-Russian Institute of Plant Genetic Resources (VIR), nomenclatural standards of apple cultivars bred at the Pavlovsk Experiment Station of VIR have been developed. The cultivars in question are ‘Kormay’, ‘Kordonovka’, ‘Kryukovskoye’, ‘Krasnoshchyokoye’, ‘Leningradskaya Zelyonka’, ‘Leningradskiy Sinap’, ‘Leningradskoye Zheltoye’, ‘Lyubimitsa Tarasenko’, ‘Novogodneye’, ‘Novoye Polosatoye’, ‘Pashkevicha Krasnoye’, and ‘Sigovskoye’.
