A new species of Ptyctolaemus Peters, 1864 is described from Htamanthi Wildlife Sanctuary, Sagaing Division, Myanmar. The new species differs from P. gularis and Ptyctolaemus aff. gularis from Tibet, China, by having relatively longer limbs and different colorations of the gular region, and it differs from P. collicristatus by having much longer limbs and a less developed nuchal crest in males. Moreover, the new species differs genetically from Ptyctolaemus aff. gularis from Tibet, China, and P. collicristatus by an uncorrected percentage distance of 23.5% and 24.8%, respectively, inferred from mitochondrial NADH dehydrogenase subunit 2 gene sequences. This discovery increases the number of known Ptyctolaemus species to three.

The Coregonus lavaretus (Linnaeus, 1758) complex is a morphologically and genetically diverse group of whitefish. Its taxonomic structure has been controversial for almost a century. At least 25 forms of C. lavaretus have been described in Siberia, but there is still no consensus on their intraspecific structure and taxonomy. Coregonus lavaretus pidschian (Gmelin, 1789) was described as a subspecies of C. lavaretus. Recently, it was assumed that this subspecies is also a complex. The purpose of this study was to compare the distributions of pidschian-like whitefish haplotypes in two basins of large Siberian rivers, Yenisei and Lena, and to assess the gene flow between basins of these rivers, which were connected after the last glaciation. The sequence of the following mitochondrial DNA genes, 16S rRNA (partial), tRNA-Leu (full), NADH dehydrogenase subunit 1 (full), tRNA-Ile (full), and tRNA-Gln (partial), were used for the inference of intraspecific genetic structure of C. l. pidschian. Whitefish haplotypes were clustered into two groups according to their distribution between two large Siberian river basins; however, there were shared haplotypes indicating events of migration and hybridization, which could occur when Bolshoi Yenisei and Lena river systems were connected after the last glaciation (the Late Pleistocene).

Abstract In this research, a pair primer targeting NADH dehydrogenase subunit 4 (ND4) gene of porcine (Sus scrofa) mt-DNA was designed and tested for its specificity. The ND4 primer was compared for its robustness against an established primer pair designed to amplify the NADH dehydrogenase subunit 5 (ND5) gene. The samples include non-halal meat animals (dog, porcine, and rat) and halal meat animals (chicken, cow, and horse). DNA from raw meat was isolated by modified Chloroform Isoamyl-Alcohol method and then was analyzed quantitatively with NanoDrop™ spectrophotometer followed by amplification using PCR technique. The amplification results proved that the two pairs of primers were specific, resulting in amplification of 120 bp DNA target for ND4 and 227 bp for ND5. It can be concluded that the two primers can differentiate between halal and non-halal animals. However, to determine the sensitivity of each primer, further research is needed.

This study explored the physiological and molecular mechanisms of yield increase in maize under stable soil water content (SW) conditions. Results of the study showed that under SW conditions, corn yield increased by 38.72 and 44.09% in 2019 and 2020, respectively. Further, it was found that dry matter accumulation, economic coefficient and photosynthetic rate also increased by 31.24 and 25.67%, 5.45 and 15.38% as well as 29.60 and 31.83% in 2019 and 2020 respectively. However, the results showed that both the activity of antioxidant enzymes and content of osmotic adjustment substances decreased in maize under SW conditions. When compared with soil moisture content of dry and wet alternation (DW) conditions, SW could not only significantly promote growth and yield of maize but also increase the economic coefficient. Transcriptome profiles of maize leaves under the two conditions (SW and DW) were also analyzed and compared. It was found that 11 genes were highly up-regulated in the photosynthesis pathway. These genes included photosystem II protein V (PsbE), photosystem II protein VI (PsbF), photosystem II protein D1 (PsbA), photosystem II protein D2 (PsbD) and ATP synthase CF1 beta subunit (atpB). Further, it was found that four genes were up-regulated in the oxidative phosphorylation pathway., These were ATP synthase CF1 epsilon subunit (atpE), ATP synthase CF1 beta subunit (atpB), NADH dehydrogenase subunit 4L (ndhE) and NADH dehydrogenase subunit 6 (ndhG). In conclusion, the physiological mechanism of stable soil water content (SW) to increase corn yield may be the enhancement of photosynthetic capacity and energy metabolism.

The aim of this study was to provide molecular characterization, together with phylogenetic analysis, of Taenia pisiformis cysts isolated from rabbit. On the basis of morphological features and molecular analysis, the cysticerci were identified as T.pisiformis metacestodes. PCR was performed with three different protocols to obtain partial sequences of 12S ribosomal RNA (12S rRNA), NADH dehydrogenase subunit 1 (nad1), and cytochrome oxidase subunit 1 (cox1) of Taenia spp. The products from the PCRs were sequenced. Interpretation of the sequencing results of the obtained amplicons, by comparing them with the GenBank database, proved that the causative agent, in this case, was T. pisiformis. The phylogenetic analysis of the received sequences identified a new haplotype. The received data can be used to supplement the species description. To our knowledge, this is the first molecular confirmation of T. pisiformis metacestodes infection in the rabbit, in Poland.

Abstract More than thirty species of the genus Onchocerca (Nematoda; Filarioidea) were responsible for onchocercosis (a vector-borne parasitic disease) of wild and domestic ungulates. “Between 2016 and 2017”, the presence of Onchocerca flexuosa (Wedl, 1856) was detected and investigated in 17 Japanese Sika deer (Cervus nippon) captured in Gifu and Shiga Prefectures, Japan. The worms were found within characteristics subcutaneous nodules dispersed in different regions of the body of all 17 deer. The all collected nodules were examined stereo-microscopically. The parasites were extracted from the nodules and identified through morphological and histopathological examinations. Molecular identification through sequencing of the following genes; internal transcribed spacer subunit 2 (ITS2)–28S ribosomal RNA (28S rRNA), cytochrome c oxidase subunit 1 (cox1) and mitochondrially encoded NADH dehydrogenase subunit 2 (NAD2) were performed. The histopathological, molecular and phylogenetic analysis demonstrated that, the filarial nematode isolated from Gifu and Shiga Prefectures in Japan is O. flexuosa. This is the first report about presence of O. flexuosa in Japanese Sika deer (Cervus nippon) in Gifu and Shiga Prefectures.

A new species of the gekkonid genus Hemiphyllodactylus is described from forested karst hills near Zhutangxiang town, Lancang Lahu Autonomous County, Yunnan, China. Hemiphyllodactylus zhutangxiangensis sp. nov. is distinguished from all other congeners in morphology and an uncorrected pairwise sequence divergence of greater than 14% based on 1039 base pairs of the mitochondrial NADH dehydrogenase subunit 2 gene (ND2). The new species is defined by the following characters: a maximum SVL of 44.42 mm; 7–9 chin scales; enlarged postmentals; five circumnasal scales; 2–4 internasal scales; 8–11 supralabial scales; 8–11 infralabial scales; subdigital lamellae on fingers II–V (3–5)-(4–6)-(4 or 5)-(4 or 5); subdigital lamellae on toes II–V (4 or 5)-(4 or 5)-(4–6)-(4 or 5); dorsal scales 11–15; ventral scales 5–7; 20–23 continuous femoroprecloacal pores; having pale-grey base color on the body with various darker transverse dorsal blotches; a dark postorbital stripe extending to at least to the base of the neck; no dark dorsolateral or ventrolateral stripe on trunk; and postsacral marking bearing a dark fork-like pattern with anteriorly projecting arms. The new species brings the species total of Hemiphyllodactylus in China to 11.

Abstract Mitochondrial complex I is the main site for electron transfer to the respiratory chain and generates much of the proton gradient across the inner mitochondrial membrane. Complex I is composed of two arms, which form a conserved L-shape. We report the structures of the intact, 47-subunit mitochondrial complex I from Arabidopsis thaliana and the 51-subunit complex I from the green alga Polytomella sp., both at around 2.9 Å resolution. In both complexes, a heterotrimeric γ-carbonic anhydrase domain is attached to the membrane arm on the matrix side. Two states are resolved in A. thaliana complex I, with different angles between the two arms and different conformations of the ND1 (NADH dehydrogenase subunit 1) loop near the quinol binding site. The angle appears to depend on a bridge domain, which links the peripheral arm to the membrane arm and includes an unusual ferredoxin. We propose that the bridge domain participates in regulating the activity of plant complex I.

The present study was conducted to get more information about the genome and locate the taxonomic position of Schizothorax niger in Schizothoracinae through mitochondrial 13 protein-coding genes (PCGs). These PCGs for S. niger were found to be 11409 bps in length ranging from 165 (ATPase 8) to 1824 bps (NADH dehydrogenase subunit 5) and encode 3801 amino acids. In these PCGs, 4 genes overlap on the similar strands, while one shown on the opposite one: ATPase 6+8 and NADH dehydrogenase subunit 4+4L overlap by 7 nucleotides. Similarly, ND5-ND6 overlap by 4 nucleotides, while ATP6 and COIII overlap by 1 nucleotide. Similarly, four commonly used amino acids in S. niger were Leu (15.6 %), Ile (10.12 %), Thr (8.12 %), and Ala (8.7 %). The results presented that COII, COIII, NDI, ND4L, and Cytb had substantial amino acid conservation as compared to the COI gene. Through phylogenetic analysis, it was observed that S. niger is closely linked with S. progastus, S. labiatus, S. plagiostomus, and S. nepalensis with high bootstrap values. The present study provided more genomic data to know the diversity of the mitochondrial genome and its molecular evolution in Schizothoracinae.
