TRPC3 and NALCN channels drive pacemaking in substantia nigra dopaminergic neurons

Midbrain dopamine (DA) neurons are slow pacemakers that maintain extracellular DA levels. During the interspike intervals, subthreshold slow depolarization underlies autonomous pacemaking and determines its rate. However, the ion channels that determine slow depolarization are unknown. Here we show that TRPC3 and NALCN channels together form sustained inward currents responsible for the slow depolarization of nigral DA neurons. Specific TRPC3 channel blockade completely blocked DA neuron pacemaking, but the pacemaking activity in TRPC3 knock-out (KO) mice was perfectly normal, suggesting the presence of compensating ion channels. Blocking NALCN channels abolished pacemaking in both TRPC3 KO and wild-type mice. The NALCN current and mRNA and protein expression are increased in TRPC3 KO mice, indicating that NALCN compensates for TRPC3 currents. In normal conditions, TRPC3 and NALCN contribute equally to slow depolarization. Therefore, we conclude that TRPC3 and NALCN are two major leak channels that drive robust pacemaking in nigral DA neurons.

A low-cost computational approach to analyze spiking activity in cockroach sensory neurons

Undergraduates use a spike sorting routine developed in Octave to analyze the spiking activity generated from mechanical stimulation of spines of cockroach legs with the inexpensive SpikerBox amplifier and the free software Audacity. Students learn the procedures involved in handling the cockroaches and recording extracellular action potentials (spikes) with the SpikerBox apparatus as well as the importance of spike sorting for analysis in neuroscience. The spike sorting process requires students to choose the spike threshold and spike selection criteria and interact with the clustering process that forms the groups of similar spikes. Once the spike groups are identified, interspike intervals and neuron firing frequencies can be calculated and analyzed. A classic neurophysiology lab exercise is thus adapted to be interdisciplinary for underrepresented students in a small rural college.

Interspike intervals within retinal spike bursts combinatorially encode multiple stimulus features

Neurons in various regions of the brain generate spike bursts. While the number of spikes within a burst has been shown to carry information, information coding by interspike intervals (ISIs) is less well understood. In particular, a burst with k spikes has k−1 intraburst ISIs, and these k−1 ISIs could theoretically encode k−1 independent values. In this study, we demonstrate that such combinatorial coding occurs for retinal bursts. By recording ganglion cell spikes from isolated salamander retinae, we found that intraburst ISIs encode oscillatory light sequences that are much faster than the light intensity modulation encoded by the number of spikes. When a burst has three spikes, the two intraburst ISIs combinatorially encode the amplitude and phase of the oscillatory sequence. Analysis of trial-to-trial variability suggested that intraburst ISIs are regulated by two independent mechanisms responding to orthogonal oscillatory components, one of which is common to bursts with a different number of spikes. Therefore, the retina encodes multiple stimulus features by exploiting all degrees of freedom of burst spike patterns, i.e., the spike number and multiple intraburst ISIs.

Bursts are typically composed of a few spikes generated within dozens of milliseconds, and individual bursts are separated by much longer periods of silence (∼hundreds of milliseconds). Recent evidence indicates that the number of spikes in a burst, the interspike intervals (ISIs), and the overall duration of a burst, as well as the timing of burst onset, encode information. However, it remains unknown whether multiple ISIs within a single burst encode multiple independent information contents. Here we demonstrate that such combinatorial ISI coding occurs for spike bursts in the retina. We recorded ganglion cell spikes from isolated salamander retinae stimulated with computer-generated movies. Visual response analyses indicated that multiple ISIs within a single burst combinatorially encode the phase and amplitude of oscillatory light sequences, which are different from the stimulus feature encoded by the spike number. Because synaptic transmission in the visual system is highly sensitive to ISIs, the combinatorial ISI coding must have a major impact on visual information processing.

Quantitative and qualitative analysis of asynchronous neural activity

The activity of a sparse network of leaky integrate-and-fire neurons is carefully revisited with reference to a regime of a bona-fide asynchronous dynamics. The study is preceded by a finite-size scaling analysis, carried out to identify a setup where collective synchronization is negligible. The comparison between quenched and annealed networks reveals the emergence of substantial differences when the coupling strength is increased, via a scenario somehow reminiscent of a phase transition. For sufficiently strong synaptic coupling, quenched networks exhibit a highly bursting neural activity, well reproduced by a self-consistent approach, based on the assumption that the input synaptic current is the superposition of independent renewal processes. The distribution of interspike intervals turns out to be relatively long-tailed; a crucial feature required for the self-sustainment of the bursting activity in a regime where neurons operate on average (much) below threshold. A semi-quantitative analogy with Ornstein-Uhlenbeck processes helps validating this interpretation. Finally, an alternative explanation in terms of Poisson processes is offered under the additional assumption of mutual correlations among excitatory and inhibitory spikes.

Stochastic Sensitivity and Method of Principal Directions in Excitability Analysis of the Hodgkin–Huxley Model

We study the probabilistic behavior of the Hodgkin–Huxley neuron model in the presence of random forcing of the external current parameter. The stochastic excitement in the zone of stable equilibria is illustrated by the statistics of interspike intervals and probabilistic distributions of mixed-mode oscillations. For the parametric analysis of this phenomenon, a constructive method for stochastic sensitivity and confidence ellipsoids is suggested. It is shown how to simplify this analysis using the principal direction approach. A constructive application of this technique is demonstrated by analyzing the stochastic excitement in the Hodgkin–Huxley model.
