ABSTRACT “Medusaviridae” is a proposed family of large double-stranded DNA (dsDNA) viruses so far represented by a sole virus isolated from a hot spring. In the present study, we report the isolation and genome sequencing of a second member of this family, medusavirus stheno, discovered from a freshwater sample with an Acanthamoeba castellanii coculture.

<em>This work investigated the effect of temperature and aeration treatments on the dissolved oxygen (DO) values in freshwater. The study was carried out using a simple water bath reactor in room temperature condition. Based on the results, freshwater sample without the aeration and temperature treatments (control) indicates a fluctuating profile on which the obtained values of DO were 4.21-4.98 mg/L at T<sub>average </sub>= 27.7 °C. However, the aeration treatment was slightly able to enhance the DO value up to 8.12 mg/L from the initial concentration of 3.88 mg/L at T<sub>average </sub>= 27.4 °C. Compared to the aeration treatment, the addition of temperature treatment has extremely increased the DO value up to 21 mg/L from 6.6 mg/L (T<sub>0 minutes</sub> = 27 °C)  for 20 minutes of DO observation (T<sub>20 minutes</sub> = 12.4 °C). Hence, this brief report suggests that the addition of temperature treatment gave a significant effect on the DO value in freshwater than the aeration treatment and control.</em>

Hypsibius vaskelae sp. nov. (Tardigrada, Eutardigrada, Hypsibiidae) is described from a freshwater sample collected in the vicinity of Saint-Petersburg. The new species has wrinkled cuticle, bucco-pharyngeal apparatus with two elongate macroplacoids and septulum, and cuticular bars between the bases of inner and outer claw and near the base of the inner claw on legs I–IV. Hypsibius vaskelae sp. nov. is most similar to Hypsibius marcelli Pilato, 1990 and H. septulatus Pilato, Binda, Napolitano & Moncada, 2004, but differs from both in having wrinkled dorsal cuticle, thinner claws, and presence of lunules on the claws of all legs.

ABSTRACT We describe here the draft genome sequence of the cyanobacterium Tychonema bourrellyi, assembled from a metagenome of a nonaxenic culture. The strain (FEM_GT703) was isolated from a freshwater sample taken from Lake Garda, Italy. The draft genome sequence represents the first assembled T. bourrellyi strain.

In water monitoring, effect of chloride ion in water body on determining COD can not be ignored. Potassium dichromate has a strong oxidizing, and it can oxidize most of the organic in matter. But it generally is used to determine the water body which COD is larger than 30mg/L, and easily influenced by Chloride ion. So we often use alkaline potassium permanganate method which has weak impact by Chloride ion in high chlorine concentration wastewater. For the tidal estuary, we often use acidic potassium permanganate method in upstream and alkaline potassium permanganate method in downstream. The different detection methods cause the disagreement of water quality evaluation. This paper describes the influence of chlorine level on the COD standard solution prepared by laboratory and river freshwater sample from Qiantang River upstream in detail, and analyses the difference between the acid potassium permanganate method and alkaline potassium permanganate by comparing the COD standard solution prepared by laboratory and the freshwater sample. Last, this paper discusses the variation of COD during a tidal period.
