Abstract Currently, 73% of the water demand in Balikpapan was only supplied from Manggar Reservoir, and it will continue decreasing parallelly with the increasing population. Regarding the population issues and the government’s planning for the new capital city of Indonesia, Balikpapan will be a buffer city facing serious risk in water supply issues in the future. Therefore, this study seeks to analyze an alternative of water supply. Following the concepts in Water Sensitive Urban Design (WSUD), The rainwater harvesting (RWH) method was chosen for preventive analysis, and Institut Teknologi Kalimantan (ITK) as one of the reputable’s universities in Balikpapan will be taken as a study location. The results obtained that the monthly water demand in ITK was ranged from 3228.34 m3 to 16632.97 m3. Using RWH, water supply analysis was obtained from 3790.62 m3 to 10697.31 m3 in various rainfall durations (0.5 to 2 hours) and 20 years projections (2022 to 2042). Following the obtained water supply. This study also reveals that the savings of total water usage can be reached from 24% to 100%, and around IDR 14,082,002 to IDR 37,035,390 is converted to currency. The highest saving reached due to water supply meets the water demand.

Selenium and iodine are the two central trace elements for the homeostasis of thyroid hormones but additional trace elements such as iron, zinc, and copper are also involved. To compare the primary effects of inadequate intake of selenium and iodine on the thyroid gland, as well as the target organs of thyroid hormones such as liver and kidney, mice were subjected to an eight-week dietary intervention with low versus adequate selenium and iodine supply. Analysis of trace element levels in serum, liver, and kidney demonstrated a successful intervention. Markers of the selenium status were unaffected by the iodine supply. The thyroid gland was able to maintain serum thyroxine levels even under selenium-deficient conditions, despite reduced selenoprotein expression in liver and kidney, including deiodinase type 1. Thyroid hormone target genes responded to the altered selenium and iodine supply, whereas the iron, zinc, and copper homeostasis remained unaffected. There was a notable interaction between thyroid hormones and copper, which requires further clarification. Overall, the effects of an altered selenium and iodine supply were pronounced in thyroid hormone target tissues, but not in the thyroid gland.

Natural disturbances such as pest outbreaks have a significant impact on forest dynamics and services, including the loss of mature stands. From a wood production perspective, these disturbances can lead to long-lasting imbalances in the overall age-class structure of the forest, potentially resulting in a shortage of mature harvestable stands. Researchers from Natural Resources Canada’s (NRCan) Canadian Forest Service (CFS) have made a timber supply analysis of the Dunière forest located in the centre of the Gaspé Peninsula (Québec). This region suffers from an age-class structure imbalance caused by an eastern spruce budworm (ESB) outbreak that ended in 1984, and is consequently facing a reduced annual allowable cut, leading to long- term implications for the regional forest sector. The authors suggest that partial harvesting – the removal of a proportion of timber in a mature stand several years before a final cut is carried out – is a promising opportunity in the ESB-affected area for mitigating mid-term timber supply shortages by smoothing the fibre supply over time.

One of the most crucial elements for the long-term success of shared transportation systems (bikes, cars etc.) is their ubiquitous availability. To achieve this, and avoid having stations with no available vehicle, service operators rely on rebalancing. While different operators have different approaches to this functionality, overall it requires a demand-supply analysis of the various stations. While trip data can be used for this task, the existing methods in the literature only capture the observed demand and supply rates. However, the excess demand rates (e.g., how many customers attempted to rent a bike from an empty station) are not recorded in these data, but they are important for the in-depth understanding of the systems’ demand patterns that ultimately can inform operations like rebalancing. In this work we propose a method to estimate the excess demand and supply rates from trip and station availability data. Key to our approach is identifying what we term as excess demand pulse (EDP) in availability data as a signal for the existence of excess demand. We then proceed to build a Skellam regression model that is able to predict the difference between the total demand and supply at a given station during a specific time period. Our experiments with real data further validate the accuracy of our proposed method.

Abstract Background: Thyroid hormone promotes many developmental and homeostatic functions. Apart from adequate circulating levels, the concentration of the active hormone T3 within tissues may be amplified by type 2 deiodinase (Dio2) by conversion from the precursor T4. Dio2 is critical in auditory development, bone maturation, brain function and control of the hypothalamic-pituitary-thyroid axis. Despite its crucial role, an obstacle to studying Dio2 is that the protein has a short half-life, is at low levels and is often transiently expressed, making it difficult to identify Dio2 in tissues at cellular resolution. Methods: We derived a Dio2-CreERt2 knockin mouse that expresses tamoxifen-dependent Cre recombinase from the endogenous Dio2 gene. When crossed onto Ai6 reporter mice, following tamoxifen treatment, Dio2-CreERt2 expression is detected as fluorescent signal in specific cells in brain regions, pituitary, and other tissues. We showed previously that Dio2 is essential for hearing with rising expression levels in the cochlea prior to onset of hearing. The Dio2-CreERt2 model identified positive cell types in the cochlear spiral ligament, septal divisions and modiolus around the sensory epithelium. Dio2-positive fibrocytes were adjacent to and extended projections around blood capillary networks, the source of T4 supply. Transcriptome analysis of isolated positive cells revealed bone lineage-related origins for many of these cells. Conclusion: The Dio2-CreERt2 model detects Dio2 expression sensitively at cellular resolution. In the cochlea, Dio2-positive cell types reside in vascularized support tissues, suggesting combined endocrine and paracrine-like control of the T3 supply. Analysis of cell origins suggests novel interactions between endocrine and skeletal systems in promoting T3 action required for hearing.
