Photo-responsive modifications and photo-uncaging concepts are useful for spatiotemporal control of peptides structure and function. While side chain photo-responsive modifications are relatively common, access to photo-responsive modifications of backbone N–H bonds is quite limited. This letter describes a new photocleavage pathway, affording N-formyl amides from vinylogous nitroaryl precursors under physiologically relevant conditions via a formal oxidative C=C cleavage. The N-formyl amide products have unique properties and reactivity, but are difficult or impossible to access by traditional synthetic approaches.

Comamonas testosteroni TA441 degrades steroids aerobically via aromatization of the A-ring accompanied by B-ring cleavage, followed by D- and C-ring cleavage. We previously revealed major enzymes and intermediate compounds in A,B-ring cleavage, β-oxidation cycle of the cleaved B-ring, and partial C,D-ring cleavage process. Here, we elucidated the C-ring cleavage and the β-oxidation cycle that follows. ScdL1L2, a 3-ketoacid Coenzyme A (CoA) transferase which belongs to the SugarP_isomerase superfamily, was thought to cleave the C-ring of 9-oxo-1,2,3,4,5,6,10,19-octanor-13,17-secoandrost-8(14)-ene-7,17-dioic acid-CoA ester, the key intermediate compound in the degradation of 9,17-dioxo-1,2,3,4,10,19-hexanorandrostan-5-oic acid (3aα-H-4α [3′-propionic acid]-7aβ-methylhexahydro-1,5-indanedione; HIP)-CoA ester in the previous study; however, this study suggested that ScdL1L2 is the isomerase of the derivative with a hydroxyl group at C-14 which cleaves C ring. The subsequent ring-cleaved product was indicated to be converted to 4-methyl-5-oxo-octane-1,8-dioic acid-CoA ester mainly by ORF33-encoded CoA-transferase (named ScdJ), followed by dehydrogenation by ORF21 and 22-encoded acyl-CoA dehydrogenase (named ScdM1M2). Then a water molecule is added by ScdN for further degradation by β-oxidation. ScdN is considered to catalyze the last reaction in C,D-ring degradation by the enzymes encoded in the steroid degradation gene cluster tesB to tesR.

Hydrogen embrittlement (HE) has been extensively studied in bulk materials. However, little is known about the role of H on the plastic deformation and fracture mechanisms of nanoscale materials such as nanowires. In this study, molecular dynamics simulations are employed to study the influence of H segregation on the behavior of intergranular cracks in bicrystalline α-Fe nanowires. The results demonstrate that segregated H atoms have weak embrittling effects on the predicted ductile cracks along the GBs, but favor the cleavage process of intergranular cracks in the theoretically brittle directions. Furthermore, it is revealed that cyclic loading can promote the H accumulation into the GB region ahead of the crack tip and overcome crack trapping, thus inducing a ductile-to-brittle transformation. This information will deepen our understanding on the experimentally-observed H-assisted brittle cleavage failure and have implications for designing new nanocrystalline materials with high resistance to HE.

In mammals, sperm-borne regulators can be transferred to oocytes during fertilization and have different effects on the formation of pronuclei, the first cleavage of zygotes, the development of preimplantation embryos and even the metabolism of individuals after birth. The regulatory role of sperm microRNAs (miRNAs) in the development of bovine preimplantation embryos has not been reported in detail. By constructing and screening miRNA expression libraries, we found that miR-202 was highly enriched in bovine sperm. As a target gene of miR-202, co-injection of SEPT7 siRNA can partially reverse the accelerated first cleavage of bovine embryos caused by miR-202 inhibitor. In addition, both a miR-202 mimic and SEPT7 siRNA delayed the first cleavage of somatic cell nuclear transfer (SCNT) embryos, suggesting that miR-202-SEPT7 mediates the delay of first cleavage of bovine embryos. By further exploring the relationship between miR-202 /SEPT7, HDAC6 and acetylated α-tubulin during embryonic development, we investigated how sperm-borne miR-202 regulated the first cleavage process of bovine embryos by SEPT7 and demonstrated the potential of sperm-borne miRNAs to improve the efficiency of SCNT.

Complexation of copper(II) with penta-coordinated N4S ligands in alcohols proceeded through unusual reaction paths involving C-N bond cleavage process. The products are square planar compound with a modified ligand structure that includes an alkoxy group provided by the solvent. The complexes are characterized by single crystal XRD, elemental analyses, IR, UV-visible and EPR spectroscopic techniques. Cyclic voltammetry shows Cu(II)/Cu(I) reduction becomes more difficult with the increase in donor strength of alkoxy group.
