Direct in vivo measurements of neurometabolic markers in the brain with high spatio-temporal resolution, sensitivity, and selectivity is highly important to understand neurometabolism. Electrochemical biosensors based on microelectrodes are very attractive analytical tools for continuous monitoring of neurometabolic markers, such as lactate and glucose in the brain extracellular space at resting and following neuronal activation. Here, we assess the merits of a platinized carbon fiber microelectrode (CFM/Pt) as a sensing platform for developing enzyme oxidase-based microbiosensors to measure extracellular lactate in the brain. Lactate oxidase was immobilized on the CFM/Pt surface by crosslinking with glutaraldehyde. The CFM/Pt-based lactate microbiosensor exhibited high sensitivity and selectivity, good operational stability, and low dependence on oxygen, temperature, and pH. An array consisting of a glucose and lactate microbiosensors, including a null sensor, was used for concurrent measurement of both neurometabolic substrates in vivo in the anesthetized rat brain. Rapid changes of lactate and glucose were observed in the cortex and hippocampus in response to local glucose and lactate application and upon insulin-induced fluctuations of systemic glucose. Overall, these results indicate that microbiosensors are a valuable tool to investigate neurometabolism and to better understand the role of major neurometabolic markers, such as lactate and glucose.

Cocaine is one of the most common illicit drugs globally, but the role of serotonin in its mechanism of action is insufficiently characterised. Consequently, we investigated the acute effects of the psychomotor stimulant cocaine on electrical stimulation-evoked serotonin (phasic) release in the nucleus accumbens core (NAcc) of urethane-anesthetized (1.5 g/kg i.p.) male Sprague-Dawley rats using N-shaped fast-scan cyclic voltammetry (N-FSCV). A single carbon fiber microelectrode was first implanted in the NAcc. Stimulation was applied to the medial forebrain bundle using 60 Hz, 2 ms, 0.2 mA, 2 s biphasic pulses before and after cocaine (2 mg/kg i.v.) was administered. Stimulation-evoked serotonin release significantly increased 5 minutes after cocaine injection compared to baseline (153±21 nM vs 257±12 nM; p = 0.0042; n = 5) but was unaffected by saline injection (1 ml/kg i.v.; n = 5). N-FSCV's selective measurement of serotonin release in vivo was confirmed pharmacologically via administration of the selective serotonin reuptake inhibitor escitalopram (10 mg/kg i.p.) which effectively increased the signal in a separate group of rats (n = 5). Selectivity to serotonin was further confirmed in vitro in which dopamine was minimally detected by N-FSCV with a serotonin to dopamine response ratio of 1:0.04 (200 nM of serotonin:1 mM dopamine ratio; p = 0.0048; n = 5 electrodes). This study demonstrates a noteworthy influence of cocaine on serotonin dynamics, and confirms that N-FSCV can effectively and selectively measure phasic serotonin release in the NAcc.

The present study is focused on the electrochemical properties of poly(3,4-propylene­dioxy­thiophene) (Poly(ProDOT)), electrocoated on the single carbon-fiber microelectrode (SCFME) in different electrolytic media, with different solvent dielectric constants (35.9, 41.7, 47.5, 53.3, 59.1 and 64.9). The highest deposition charge density of 24.49 mC cm-2 and the highest specific capacitance of 23.17 mF cm-2 were obtained for Poly(ProDOT) synthesized in a medium with the lowest solvent dielectric constant (e = 35.9). Electrochemical impedance spectroscopy (EIS) results of Poly(ProDOT) coated SCFME measured at open circuit potential showed continuously increased impedance magnitudes as ε was increased from 35.9 to 59.1. For all films, almost capacitive impedance responses at lower frequencies at least were obtained. The highest capacitance was observed for the polymer film synthesized in the medium of e = 35.9. The impedance of this film was also measured in different solvent mixtures with different dielectric constants at open circuit potential.
